The movement ecology of raccoons varies widely across habitats with important implications for the management of zoonotic diseases such as rabies. However, the spatial ecology of raccoons remains poorly understood in many regions of the United States, particularly in the southeast. To better understand the spatial ecology of raccoons in the southeastern United States, we investigated the role of sex, season, and habitat on monthly raccoon home range and core area sizes in three common rural habitats (bottomland hardwood, upland pine, and riparian forest) in South Carolina, USA. As long as there is food, there will also be bacteria that are capable of compromising the quality and safety of food. This dissertation aims to study bacteria capable of spoiling meat, causing illness, or both. The literature review summarizes the understanding of several bacterial pathogens, current issues in combating those pathogens in the food supply, and how spoilage Pseudomonas behave in the meat supply. Chapter 2 explored the impact of organic acid treatments on the spoilage microbiome of raw beef during extended storage. The tomato processing industry generates large amounts of waste, which creates disposal problems and negatively impacts the environment. Tomato waste contains lycopene, high-value health- and wellness-promoting bioactive, and it is a potential source of natural lycopene for food and nutraceutical applications. However, isolation of lycopene from its source requires toxic solvents, and lycopene is unstable during storage. The objective of this study was to extract lycopene from tomato waste and to convert it to a shelf-stable and easy-to-use free-flowing powder form using green extraction and particle formation processes based on supercritical carbon dioxide (SC-CO2) technology. This dissertation is a compilation of 5 chapters culminating in a bioeconomic model of the interactions between an integrated crop-livestock system and native rangeland grazing system. The model is developed using data from the eastern Sandhills of Nebraska, a location where cropping systems of Eastern Nebraska border one of the last intact grasslands remaining in North America.

MicroRNAs ( miRNAs ) are important regulators of genes expression. Their levels are precisely controlled through modulating the activity of the microprocesser complex ( MC ) . Here, we report that JANUS, a homology of the conserved U2 snRNP assembly factor in yeast and human, is required for miRNA accumulation. JANUS associates with MC components Dicer-like 1 ( DCL1 ) and SERRATE ( SE ) and directly binds the stem- loop of pri-miRNAs. In a hypomorphic janus mutant, the activity of DCL1, the numbers of MC, and the interaction of primary miRNA transcript ( pri-miRNAs ) with MC are reduced. …

African swine fever virus (ASFV) is circulating in many swine-producing countries, causing significant economic losses. It is observed that pigs experimentally vaccinated with a live-attenuated virus (LAV) but not a killed virus (KV) vaccine develop solid homologous protective immunity. The objective of this study was to comparatively analyze antibody profiles between pigs vaccinated with an LAV vaccine and those vaccinated with a KV vaccine to identify potential markers of vaccineinduced protection. This study investigates two critical areas in bioinformatics: enhancing transparency in medical image analysis and advancing the discovery of Anti-CRISPR (Acr) proteins, which have potential in developing more precise and controlled CRISPR-Cas gene editing tools. Kaposi sarcoma-associated herpesvirus is the etiological agent of several pathologies including Kaposi sarcoma, primary effusion lymphoma, and multicentric Castleman’s disease. Here, we investigated both the tumor microenvironment (TME) in KS lesions as well as plasma from KSHV-infected individuals and how metabolic dysfunction and dysregulated cytokines might have a systemic effect on KS tumorigenesis. Based on previously published KS tumor transcriptomics, one aspect of our investigation involved characterizing cell surface glycoproteins with potential to serve as therapeutic targets against KSHV-infected cells or KS tumors or as biomarkers for disease. The Nebraska Water Quality Index, under development by the Nebraska Department of Environment and Energy, summarizes in a single value eight environmental parameters that have been monitored in Nebraska for nearly 20 years. Water quality parameters including those used in the Nebraska Water Quality Index have been shown in previous studies to impact bacterial growth. As such, this index has the potential to correlate with the freshwater microbial community. Short train rail lines across the United States are seeing increased national funding to reduce toxic chemical spills caused by train derailments, the most notable of which happened in February 2023 in East Palestine, Ohio. A year prior, the film White Noise (2022) featured a similar toxic train derailment incident, taking place, too, in Eastern Ohio, and featuring actors from the town of East Palestine. This research focused on health management through the identification of suitable interventions in stressed livestock. For this investigation, we conducted two experiments. First, we determined whether administration of dexamethasone or fish oil to heat-stressed livestock to reduce systemic inflammation would mitigate metabolic deficits, enhance nutrient utilization, and improve health. We found that these interventions helped to reduce circulating inflammatory cytokines and improve respiration, muscle glucose metabolism, lipid homeostasis, cardiovascular function, and organ pathology in lambs exposed to 30 days of heat stress. Investigating the influences of single nucleotide polymorphisms (SNP) associated with conceptus death (CD) on day 16 of pregnancy could distinguish cows who have inferior value for fertility traits. We hypothesized that CD is associated with SNP genotypes that disrupt maternal recognition and lead to early pregnancy loss. RNA sequences from Holstein cow pregnancies (n = 15) with normal conceptuses (Norm) or CD were subjected to SNP discovery via Qiagen CLC Genomics Workbench. Human-animal interactions (HAI) are commonplace in society and play a consequential role in a variety of situations such as companion animal ownership, agriculture, or youth programs such as 4-H or FFA. Interacting with animals has been shown to provide developmental benefits to children. This dissertation investigates work-based learning as a method to prepare students for agricultural careers through three interconnected studies. The first two studies investigate secondary education programs in the United States, while the third study explores postsecondary education in Rwanda. In Study 1, a philosophical research approach explores how the new Supervised Agricultural Experience (SAE) approach, SAE for All, aligns five Foundational SAE components with learning theories and supports Career and Technical Education priorities. Herbivory is often believed to always negatively affect the growth of plants, but there are plant species that can actually benefit from being consumed by herbivores. These plants have a phenotypic trait known as “compensatory growth.” A plant that exhibits compensatory growth is able to increase its intrinsic growth rate as a response to taking damage caused by herbivory, and as a result, produce more biomass. Very few mathematical models used to describe plant-herbivore interactions have accounted for plant compensatory growth. Locating potential mates in non-native habitats is one of the most important challenges faced by invasive vertebrate species. The Argentine black and white tegu lizard (Salvator merianae) is a major invasive reptile species in the contiguous United States and is rapidly expanding its range across Florida and the Southeast, in part due to inadequate management strategies and tools. Introduction—Approximately 1% of the world’s population is impacted by epilepsy, a chronic neurological disorder characterized by seizures. One-third of epileptic patients are resistant to AEDs, or have medically refractory epilepsy (MRE). One non-invasive treatment that exists for MRE includes the ketogenic diet, a high-fat, low-carbohydrate diet. Despite the KD’s success in seizure attenuation, it has a few risks and its mechanisms remain poorly understood. The KD has been shown to improve metabolism and mitochondrial function in epileptic phenotypes. Potassium channels have implications in epileptic conditions as they have dual roles as metabolic sensors and control neuronal excitation.

We document rare finds of fossil bats from two localities representing the Pleistocene epoch in southern and central Nebraska, Albert Ahrens locality (No-104, late Irvingtonian age, Middle Pleistocene), Nuckolls County, and Litchfield (Sm-102, late Rancholabrean age, latest Pleistocene), Sherman County. The Albert Ahrens local fauna with strong boreal influence has produced two bats, Lasiurus cf. borealis and Cf. Myotis sp. The Litchfield local fauna, also showing a strong boreal influence, has yielded two bats, Eptesicus fuscus and Cf. Myotis, among a diverse Pleistocene fauna of small vertebrates and a pollen record indicating a boreal mixed conifer and deciduous woodland, …

Despite effective antiretroviral therapy, HIV co-morbidities remain where central nervous system (CNS) neurocognitive disorders and cardiovascular disease (CVD)-pathology that are linked with myeloid activation are most prevalent. Comorbidities such as neurocogntive dysfunction and cardiovascular disease (CVD) remain prevalent among people living with HIV. We sought to investigate if cardiac pathology (inflammation, fibrosis, cardiomyocyte damage) and CNS pathology (encephalitis) develop together during simian immunodeficiency virus (SIV) infection and if their co-development is linked with monocyte/ macrophage activation. We used a cohort of SIV-infected rhesus macaques with rapid AIDS and demonstrated that SIV encephalitis (SIVE) and CVD pathology occur together more frequently …
