Amphibians have experienced major declines and are highly vulnerable to the impacts of climate change. Yet we still lack sufficient information to understand where amphibians are most vulnerable to changes in climate and how effects might manifest. In this dissertation, I worked to understand risks due to climate change and investigate effective indicators of survival in amphibians. To predict which species might be most affected by increasingly dry conditions, in Chapter 1, I extended the climate change vulnerability assessment framework to rank amphibian vulnerability based on projected climate change and species life history traits. Agrobiodiversity, the diversity of crops that been co-produced through humanenvironment interactions over millennia, constitutes an important part of humanity’s cultural heritage. Agrobiodiversity is as much about the local ecosystem and biotic resources, such as soil, water, pollinators, and microbes, as it is about human processes like sociocultural and linguistic practices, including farming methods, knowledge, skills, and natural resource management. At the center of these dynamics are the smallholder farmers who engage in a range of stewardship practices that contribute to the diversification and evolution of the native crops that in turn support their food security. To me, I see humans as part of nature – as we are mammals, animals, part of the food chain, biological beings no higher than others on our planet. We have simply constructed this false narrative around us within our societies, minds and media that embeds this division between us and nature, between us and wildlife. Issues related to diversity, equity, and inclusion are becoming increasingly important to park and protected area managers. Recently, several Executive Orders have established policies and priorities for steering public lands to better serve the diversity of the US public. Certain groups, compared to the US population at large, are underrepresented as visitors to parks and protected areas in the US, including BIPOC communities (Black, Indigenous, and other People of Color), women, people with disabilities, veterans, people with lower socioeconomic status, and the elderly. The neuronal calcium sensor 1 (NCS-1), an EF-hand Ca binding protein, and Ric-8A coregulate synapse number and probability of neurotransmitter release. Recently, the structures of Ric-8A bound to Gα have revealed how Ric-8A phosphorylation promotes Gα recognition and activity as a chaperone and guanine nucleotide exchange factor. However, the molecular mechanism by which NCS-1 regulates Ric-8A activity and its interaction with Gα subunits is not well understood. Given the interest in the NCS-1/Ric-8A complex as a therapeutic target in nervous system disorders, it is necessary to shed light on this molecular mechanism of action at atomic level.
