School principals face critical events as part of their professional roles. These critical events can profoundly impact their ability to lead, which is why this phenomenological study explored the central question, how do principals experience critical events that include perceived trauma? Using Moustakas’ (1994) process of transcendental phenomenological analysis, this research investigated the multifaceted emotional, professional, and personal responses of these educational leaders, revealing the complexities and consequences of their experiences. Semistructured interviews with fourteen current and former principals provided rich, in-depth qualitative data that captured the essence of their experiences. The extent to which Puerto Rico’s indigenous communities resisted, survived, and were transformed by colonization is unclear despite extensive ethnohistorical research. Historical claims of extinction are attributed to colonial census information but are strongly opposed by islanders who claim Indigenous Taíno ancestry (Nieves-Colón et al. 2019). Ethnic composition of Puerto Ricans is primarily European, African, and Indigenous and these compositions reflect migrations and admixture. Aphasia, a common communication disorder following a stroke, significantly impacts psychosocial wellbeing and quality of life. Despite advancements in rehabilitation frameworks, traditional impairment-based interventions prevail, leaving gaps in addressing holistic needs. This study aims to compare patient-reported outcome measures (PROMs) between intensive comprehensive aphasia programs (ICAP), modified ICAPs (mICAPs) and individual usual care (UC) service delivery models for aphasia intervention. Eighteen participants with aphasia were broadly recruited for this study. Each completed a series of PROMs pre-and post-treatment examining psychosocial wellbeing, communicative participation, and health related quality of life. Arctic and boreal regions, in particular, have warmed more than double the global average and are experiencing increasing precipitation, thawing permafrost, shrinking snow seasons, and increasing shrub- and tree-line encroachment. Pattee Creek was once a free-flowing stream that meandered across the Missoula Valley, but has been channelized, piped, and degraded by logging, mining, agriculture, and urbanization. This thesis used a mixed-method approach to examine the impact of human activities on the Pattee Creek watershed. Beginning with a historical overview, the story of how the creek has been utilized and why it was buried was told. An analysis of historical photos from 1937 to 2019 revealed drastic changes in the land cover of the watershed and the channel of lower Pattee Creek. Changing climate, vegetation, and fire exclusion are altering and homogenizing fire-regime attributes compared to historic conditions. Fire-regime changes are commonly quantified using departure metrics, which are often based on measures of central tendency (i.e. changes in the mean). These metrics can mischaracterize complex changes to the distributional characteristics of fire-regime attributes. Here we develop a fire-regime departure metric that quantifies nonparametric distributional changes to fire-regime attributes over time. We use this metric to compare fire frequency and severity between historical (~1600-1880) and contemporary (1880- 2021) time periods in western US forests. An increasing number of spontaneous mutations in the GABA transporter gene SLC6A1 (GAT-1) have been associated with pediatric epilepsy and neurodevelopmental disorders. The molecular mechanisms and impact of mutations on GABA homeostasis and neuronal circuit function are poorly understood. Furthermore, there are currently no validated therapeutic approaches to treat patients. Pre-mRNA intron splicing is essential to the production of many eukaryotic proteins, but it is relatively uncommon for sequence analysis tools to account for this process in their methods. In this dissertation I present three tools for biological sequence analysis, demonstrating the benefits that they derive from their explicit awareness of intron splicing. First, I present Mirage2, an updated implementation of the Mirage multiple sequence alignment program, which produces alignments of protein isoforms according to spliced genome mapping coordinates. The information included are a complete biological profile, any taphonomic features or pathological conditions present, and a literature review on forensic odontology due to the severe nature of the teeth. This individual is consistent with a 5'2"-5'6" adult male ranging between the ages of 35-50 of ambiguous population affinity. Evidence of osteoarthritis and bone spurs are present along the vertebral spine, clavicle, and articulation points throughout the post-cranial skeleton. In the arid and semi-arid western U.S., migratory waterbirds (e.g., shorebirds, wading birds, and waterfowl) rely on wetland landscapes that function as nodes in broader continental habitat networks (e.g., flyways). Here, I integrate citizen science and GPS animal tracking technologies with satellite imagery and cloud computing to quantify the effects of climate and land use change on wetland ecosystems that sustain migratory waterbirds in the western U.S. Four decades of wetland surface water hydrology were first reconstructed from over 150,000 individual satellite images covering 3.1 million ha to track monthly changes in the extent and duration of wetland inundation. The Serban lab has previously developed and reported on a single application drug delivery system for the treatment of otitis externa. Otitis externa also known as outer ear infections is a very common ailment that impacts large portions of both human and domestic animal populations. Typical treatment for otitis externa typically requires an extended dosing regimen over several days that is inconvenient and can lead to poor clinical outcomes due to poor patient compliance. River recreation in the U.S. increased dramatically in the 1970s as boating technology and recreational skill level shifted, thus a number of federally- and state-managed river areas instituted use limits over the years to prevent the environmental and social impacts perceived to have stemmed from increased use. By introducing sister methodologies for both historical and modern contexts, this research bridges the methodological gap in care analysis, enhancing the original IOC with contextual environmental variables and, for modern individuals, DNA analysis.

The COP9 signalosome is a highly conserved eukaryotic complex regulating protein degradation via deneddylation of Cullin-RING E3 ligases. CSN5, the COP9’s fifth component, contains the catalytically active domain for CSN deneddylation. The complex is inactive without CSN5; however, CSN5 engages in COP9-independent binding with several other proteins, typically promoting either destruction or stabilization of its partners. Many of its confirmed interaction partners are also implicated in tumorigenesis (prominent examples being p27 and p53) and a complex cancer interactome has been established for CSN5. The overall aim of this dissertation was to investigate the influence of female sex hormones on the response to exercise in premenopausal and postmenopausal women. Study one examined the effect of menstrual cycle phase on thermoregulation during prolonged exercise in the heat. Notably, elevated core temperature and basal aldosterone was apparent in the mid-luteal phase corresponding with elevations in estrogen and progesterone. Aldosterone significantly increased following exercise in the heat but remained elevated in the mid-luteal phase, suggesting a previously unexplored menstrual phase-aldosterone hormonal additivity under the stress of uncompensable heat.
