Diabetic individuals have an increased risk for developing cardiovascular disease due to stiffening of the left ventricle (LV), which is thought to occur, in part, by increased AGE/RAGE signaling inducing fibroblast differentiation. Advanced glycated end-products (AGEs) accumulate within the body over time, and under hyperglycemic conditions, the formation and accumulation of AGEs is accelerated. AGEs exert their effect by binding to their receptor (RAGE) and can induce myofibroblast differentiation, leading to increased cell migration. Previous studies have focused on fibroblast migration during wound healing, in which diabetics have impaired fibroblast migration compared …

The neurosteroid, 5α-pregnan-3α-ol-20-one (known as “allopregnanolone” or 3α,5α-THP), is produced in the midbrain ventral tegmental area (VTA), independent of peripheral sources of progestogens, where it has potential actions at N-methyl-D-aspartate (NMDA) and GABAA receptors to facilitate rodent sexual behavior. Progestogens can also have anti-anxiety effects, but whether these involve actions of centrally-derived 3α,5α-THP or these receptors to support reproductively-relevant behavior is not well understood. We investigated the extent to which 3α,5α-THP’s actions via NMDA and/or GABAA receptors in the midbrain VTA influence reproductive behaviors. Estradiol-primed, ovariectomized/adrenalectomized (OVX/ADX) rats received midbrain VTA …

Topical drug delivery is an attractive alternative to conventional methods because of advantages such as non-invasive delivery, by-pass of first pass metabolism, and improved patient compliance. However, several factors such as skin, physicochemical properties of the drug, and vehicle characteristics influence the permeation. Within a formulation, critical factors such as concentration of drug, physical state of drug in the formulation, and organoleptic properties affect the flux across the skin. The aim of the study was to develop and investigate topical semisolid preparations (creams and gels) with ibuprofen as the model drug …

© 2020 by the authors. Licensee MDPI, Basel, Switzerland. Insects that undergo metamorphosis from juveniles to adults provide an intriguing opportunity to examine the effects of life stage, species, and the environment on their gut microbiome. In this study, we surveyed the gut microbiomes of 13 species of dragonfly collected from five different locations subject to different levels of human impact. Juveniles were collected as nymphs from aquatic habitats while airborne adults were caught at the same locations. The gut microbiome was characterized by next generation sequencing of the bacterial 16S rRNA gene. Life stage was an important factor, with …

Avocado oil is prized for its high nutritional value due to the substantial amounts of triglycerides (TGs) and unsaturated fatty acids (FAs) present. While avocado oil is traditionally extracted from mature fruit flesh, alternative sources such as avocado seed oil have recently increased in popularity. Unfortunately, sufficient evidence is not available to support the claimed health benefit and safe use of such oils. To address potential quality issues and …

See http://ivyspring.com/terms for full terms and conditions. The sigma 1 receptor (S1R) is widely expressed in the CNS and is mainly located on the endoplasmic reticulum. The S1R is involved in the regulation of many neurotransmission systems and, indirectly, in neurodegenerative diseases. The S1R may therefore represent an interesting neuronal biomarker in neurodegenerative diseases such as Parkinson's (PD) or Alzheimer's diseases (AD). Here we present the characterisation of the S1R-specific 18F-labelled tracer 18F-IAM6067 in two animal models and in human …

Formulation Development Of Curcumin-Piperine Solid Dispersion Via Hot-Melt Extrusion To Enhance Solubility And Bioavailability Of Curcumin, Abdulmajeed Althobaiti
Electronic Theses and Dissertations
Curcumin has been known to have a variety of biological activities in different diseases, including inflammation, cancer, and diabetes. The limited bioavailability of curcumin is due to its low solubility and extensive metabolism, rendering curcumin unusable as an oral therapeutic agent. The aim of the study is to enhance the solubility and bioavailability of curcumin by preparing curcumin-piperine solid dispersion (SD) utilizing the hot-melt extrusion technique. Three physical mixtures with different ratios were prepared as following: F1 (5% w/w piperine, 15% w/w curcumin, 80% w/w Soluplus®), F2 (10% w/w piperine, 30% w/w curcumin, 60% w/w Soluplus®), and F3 (15% w/w …

Approaches To Alleviate The Vigabatrin Induced Retinal Toxicity, Anitha Police
Electronic Theses and Dissertations
Safety of therapeutics is one of the primary concerns during the treatment of any ailments. Most of the toxic effects of drugs occurs due to accumulation of drug in organs, which is dependent on the dosage of the drug. The ideal therapy should minimize the risk of causing adverse events at the same time elicit intended therapeutic effects. Vigabatrin (VGB) is an antiepileptic drug used primarily in the treatment of infantile spasms and it acts by inhibiting gamma amino butyric acid (GABA) transaminase thereby increasing inhibitory neurotransmitter GABA levels. At therapeutic doses, VGB accumulates in retina leading to peripheral visual …

Development Of A Liposomal Formulation For Brain Targeting, Ziru Zhang
Electronic Theses and Dissertations
The blood-brain barrier (BBB), formed by the endothelial cells of the brain capillaries, inhibits the penetration of many therapeutic compounds into the brain. Liposomes, which have unique physicochemical characteristics, have been widely investigated for the drug delivery system across BBB. In the current study, FITC-dextran was used as a model drug that encapsulated in a liposomal formulation to investigated its ability to overcome the blood-brain barrier for targeted brain delivery of therapeutic agents. Here, non-targeted liposome (NT LPs) and RVG-modified liposome (RVG LPs) were prepared. The NT LPs and RVG LPs were about 97 and 101 nm in diameter with …

Labor In The Field Is Much Changed: The Chickasaws And The Civilization Plan, 1790-1837, Jeffrey D. Washburn
Electronic Theses and Dissertations
Between 1790 and 1840, the U.S. federal government instituted its so-called “civilization plan,” which encouraged Southern Indians to embrace republican values, patriarchy, yeoman farming, and individual property rights. The policies included in the civilization plan were highly influential, as the Native South transitioned from an economy built on the deerskin trade into one of cottage industries, plow-based agriculture, and ranching. Through a detailed examination of one Southern Indian community, the Chickasaws, my dissertation defines the civilization plan in terms of the ideology that the federal government envisioned and how the plan was interpreted on the ground level. My work centers …
Role Of Insulin-Like Growth Factor 1 Receptor In The Regulation Of Astrocyte Structure, Function And Cognition, Disha Prabhu
Role Of Insulin-Like Growth Factor 1 Receptor In The Regulation Of Astrocyte Structure, Function And Cognition, Disha Prabhu
Electronic Theses and Dissertations
Levels of Insulin-like growth factor I (IGF-1) are reduced in several neurological disorders, including aging and neurodegeneration. Aged animals have reduced IGF1 signaling and exhibit impairment in cognition. Supplementing rodents with IGF-1 reverses some of the cognitive deficit, suggesting that IGF-1 signaling could potentially be targeted to ameliorate cognitive decline in older adults. For this, a thorough understanding of the mechanisms by which IGF-1 regulates the function of various cell types in the brain is needed. A majority of studies have focused on the effects of IGF-1 regulating neurons. However, astrocytes that form roughly about 40% of brain mass also …
