Serum amyloid A (SAA) is a family of proteins, the plasma levels of which may increase >1000-fold in acute inflammatory states. We investigated the role of SAA in sepsis using mice deficient in all three acute-phase SAA isoforms (SAA-TKO). SAA deficiency significantly increased mortality rates in the three experimental sepsis mouse models: cecal ligation and puncture (CLP), cecal slurry (CS) injection, and lipopolysaccharide (LPS) treatments. SAA-TKO mice had exacerbated lung pathology compared to wild-type (WT) mice after CLP. A bulk RNA sequencing performed on lung tissues excised 24 h after CLP indicated significant enrichment in the expression of genes associated …

Platelets express several surface receptors that could interact with different viruses. To understand the mechanisms of HIV-1′s interaction with platelets, we chose the EcoHIV model. While EcoHIV is an established model for neuroAIDS, its effects on platelets are ill-defined. Our results indicate that EcoHIV behaves differently from HIV-1 and is cleared from circulation after 48 h post- infection. The EcoHIV course of infection resembles an HIV-1 infection under the effects of combined antiretroviral therapy (cART) since infected mice stayed immunocompetent and the virus was readily detected in the spleen. EcoHIV-infected mice failed to become thrombocytopenic and showed no signs of …

Background and aims: Genome and epigenome wide association studies identified variants in carnitine palmitoyltransferase 1a (CPT1a) that associate with lipid traits. The goal of this study was to determine the role of liver-specific CPT1a on hepatic lipid metabolism.
Approach and results: Male and female liver-specific knockout (LKO) and littermate controls were placed on a low-fat or high-fat diet (60% kcal fat) for 15 weeks. Mice were necropsied after a 16 h fast, and tissues were collected for lipidomics, matrix-assisted laser desorption ionization mass spectrometry imaging, kinome analysis, RNA-sequencing, and protein expression by immunoblotting. Female LKO mice had increased serum alanine …

The mosquito family Culicidae is divided into 2 subfamilies named the Culicinae and Anophelinae. Nix, the dominant male-determining factor, has only been found in the culicines Aedes aegypti and Aedes albopictus, 2 important arboviral vectors that belong to the subgenus Stegomyia. Here we performed sex-specific whole-genome sequencing and RNAseq of divergent mosquito species and explored additional male-inclusive datasets to investigate the distribution of Nix. Except for the Culex genus, Nix homologs were found in all species surveyed from the Culicinae subfamily, including 12 additional species from 3 highly divergent tribes comprising 4 genera, suggesting Nix originated at least 133 to …

Preparing and manipulating pure magnetic states in molecular systems are the key initial requirements for harnessing the power of synthetic chemistry to drive practical quantum sensing and computing technologies. One route for achieving the requisite higher spin states in organic systems exploits the phenomenon of singlet fission, which produces pairs of triplet excited states from initially photoexcited singlets in molecular assemblies with multiple chromophores. The resulting spin states are characterized by total spin (quintet, triplet, or singlet) and its projection onto a specified molecular or magnetic field axis. These excited states are typically highly polarized but exhibit an impure spin …

Cell proliferation requires metabolic reprogramming to accommodate biosynthesis of new cell components, and similar alterations occur in cancer cells. However, the mechanisms linking the cell cycle machinery to metabolism are not well defined. Cyclin D1, along with its main partner cyclindependent kinase 4 (Cdk4), is a pivotal cell cycle regulator and driver oncogene that is overexpressed in many cancers. Here, we examine hepatocyte proliferation to define novel effects of cyclin D1 on biosynthetic metabolism. Metabolomic studies reveal that cyclin D1 broadly promotes biosynthetic pathways including glycolysis, the pentose phosphate pathway, and the purine and pyrimidine nucleotide synthesis in hepatocytes. Proteomic …

CD8þ T cells from HLA-A2.1-transgenic mice, but not wild-type mice, immunized with the amino- terminus region (aa 41e152) of dense granule protein 6 (GRA6Nt) of Toxoplasma gondii secreted large amounts of perforin and granzyme B in response to GRA6Nt through antigen presentation by HLA-A2.1 in vitro. When those CD8þ T cells were transferred into chronically infected HLA-A2.1-expressing NSG mice deficient in T cells, cerebral cyst burden of the recipients of HLA-A2.1-transgenic T cells, but not of WT T cells, became significantly less than that of control mice with no cell transfer. Furthermore, the significant reduction of the cyst burden by …

Background: Magnetic resonance imaging (MRI) is a non-invasive imaging modality which, in conjunction with biopsies, provide a qualitative assessment of tumor response to treatment. Intravenous injection of contrast agents such as fluorine (19F) nanoemulsions labels systemic macrophages, which can, then, be tracked in real time with MRI. This method can provide quantifiable insights into the behavior of tumor-associated macrophages (TAMs) in the tumor microenvironment and macrophage recruitment during therapy.
Methods: Female mice received mammary fat pad injections of murine breast or colon cancer cell lines. The mice then received an intravenous 19F nanoemulsion injection, followed by a baseline 19F MRI. …

A major challenge to integrating public metabolic resources is the use of different nomenclatures by individual databases. This paper presents md_harmonize, an open-source Python package for harmonizing compounds and metabolic reactions across various metabolic databases. The md_harmonize package utilizes a neighborhood-specific graph coloring method for generating a unique identifier for each compound via atom identifiers based on a compound’s chemical structure. The resulting harmonized compounds and reactions can be used for various downstream analyses, including the construction of atom-resolved metabolic networks and models for metabolic flux analysis. Parts of the md_harmonize package have been optimized using a variety of computational …

Dysregulated eEF2K expression is implicated in the pathogenesis of many human cancers, including triple-negative breast cancer (TNBC), making it a plausible therapeutic target. However, specific eEF2K inhibitors with potent anti-cancer activity have not been available so far. Targeted protein degradation has emerged as a new strategy for drug discovery. In this study, a novel small molecule chemical is designed and synthesized, named as compound C1, which shows potent activity in degrading eEF2K. C1 selectively binds to F8, L10, R144, C146, E229, and Y236 of the eEF2K protein and promotes its proteasomal degradation by increasing the interaction between eEF2K and the …

IGC Proceedings (1977-2023)
In the transition period between the rainy and dry seasons there is decreased water, temperature and luminosity, reducing forage production and quality. Because of this variability in forage mass production, it is essential to quantify forage mass for use in planning and decision making about pastures management. The objective was to evaluate Brachiaria spp. (Syn. Urochloa spp.) cv. Mulato II (Convert HD364) growth, biomass production managed under continuous grazing with steers fed with protein supplement feed in-trough and on-ground during the seasonal transition period. The experiment was carried out in the Beef Cattle Sector of Jatai Federal University …

Production And Persistence Of Self-Regenerating Annual Clovers In The Pacific Northwest, Ian Silvernail, Serkan Ates
IGC Proceedings (1977-2023)
Self-regenerating annual legumes are commonly included in dryland pastures to increase the production of high-quality forages to meet the nutritional demands of growing and lactating animals. Balansa clover (Trifolium michelianum Savi), not as widely evaluated as subterranean clover (Trifolium subterraneum L.), has shown promise in Western Oregon. However, there is a paucity of information on the productivity and persistence of annual clover cultivars that have various flowering times. This study investigates the biomass production and persistence of early, mid and late flowering subterranean clover and balansa clover cultivars in Corvallis, Oregon. Averaged across the cultivars, the total annual …

IGC Proceedings (1977-2023)
Red clover remains a globally important forage legume grown mostly in humid temperate climates on approximately 4 million ha. In the past 25 years global red clover seed production has stabilized around 10 million kg yr-1, although there is evidence that seed production is increasing globally at around 0.34 million kg yr-1 since the early 2000s. Globally many red clover programs exist targeting specific global regions with a focus on improving persistence, biomass yield, seed yield, and a few other minor traits. Breeding programs are increasing persistence by selecting for specific disease resistances, specific agricultural conditions such …

Managing Vegetation In Grassland Habitats To Enhance Livestock Or Wildlife Objectives, B. B. Sleugh, S. Flynn, W. Hatler, C. Hart, S. Ingram, Joao B. Cason, D. Ovalle
IGC Proceedings (1977-2023)
Sustainably stewarding grassland systems involves applying various practices to manipulate forage interactions with other plants, the environment, and grazing animals to meet resource manager objectives. These interactions can result in invasion or encroachment and increased abundance of weeds which hinder attainment of management objectives. Weeds influence the structure and function of pasture ecosystems whether forages are grown in improved pastures, rangeland, or grassland communities. They degrade pasture quality and reduce livestock performance by interfering with forage establishment, yield, and quality by competing for resources. Weeds reduce the feed value of forage, decrease pasture carrying capacity, and can be toxic or …

IGC Proceedings (1977-2023)
‘Maton’ cereal rye (Secale cereale L.) and ‘TAM-90’ annual ryegrass (Lolium multiflorum Lam.) are used for winter annual pastures to background stocker cattle in the southeastern US. Rye and ryegrass were sod-seeded into bermudagrass [Cynodon dactylon (L.) Pers.] pastures and fixed-stocked at different rates under continuous and 8-paddock rotation methods during each of 7 years to: 1) compare stocking methods for daily gain per animal (ADG) and gain/ha; and 2) quantify relationships of ADG with forage mass and forage allowance. Each of 3 levels of stocking rate, 4.9, 6.4, and 8.6 hd/ha, significantly affected ADG at 1.24, …

Characteristics Of Lactic Acid Bacteria Strains Isolated From Tibetan Plateau And Their Effects On Silage Quality Of Italian Ryegrass (Lolium Multiflorum Lam.) At Low Temperature, Siran Wang, Mudasir Nazar, Niaz Ali Kaka
IGC Proceedings (1977-2023)
Temperature is an important factor affecting ensilage. In cold regions, low temperature could be an adverse environmental factor during ensiling. However, little research has focused on improving silage quality at low temperatures. This study aimed to examine two lactic acid bacteria (LAB) strains (LCG9 and TG1) isolated from the Tibetan Plateau, and evaluate their effects on the silage quality of Italian ryegrass (Lolium multiflorum Lam.) at three temperatures (10° C, 15° C and 25° C). The isolated strains and one commercial inoculant (G, Lactobacillus plantarum MTD-1) were evaluated by morphological, physiological and biochemical tests. Strains G, LCG9, TG1 and …

It Takes More Than Calcium To Neutralize Soil Acidity, Edwin L. Ritchey, Christopher D. Teutsch, John H. Grove
IGC Proceedings (1977-2023)
Proper pH management is the foundation to a good soil fertility program. Soil pH influences nutrient availability and root growth and function. Just because an agricultural product contains calcium does not mean that it will change soil pH. The effectiveness of three calcium products in raising soil pH was compared to an untreated check in acid soils. A field trial was conducted at 16 locations across Kentucky and a laboratory incubation study was conducted at the University of Kentucky Research and Education Center. The same application rates were used for the three products, whether in field trials or laboratory incubations. …

A Novel Tractor Operated Grass Seed Harvester Developed In India, Chandra Shekhar Sahay, D. S. Thorat, P. S. Pathak
IGC Proceedings (1977-2023)
The demand of green and dry fodder in India is estimated to increase to 1170 and 650 m tonne whereas availability is expected to be at 411.3 and 488 m tonne in 2025, respectively, depicting deficit of about 64.9% green fodder and 24.9% dry fodder (Vision 2030, ICAR - IGFRI, Jhansi, 2011). In forages, availability of quality seed is only 25-30% in cultivated fodder and less than 10 % in range grasses and legumes (Vision 2050, IGFRI). Prices paid for grass seeds of native species vary from Rs.5,000 to 6,500 per kg for clean, un-haired seeds due to excessive use …

Making Round Bale Silage – Lessons Learned From An On-Farm Survey Of Baleage In Kentucky, Jimmy C. Henning, Jeff Lehmkuhler, Brandon Sears, Corinne Belton, L. Berg, Don Sorrell, April Wilhoit
IGC Proceedings (1977-2023)
The ability to make silage in round bale packages allows producers to avoid rain damage and produce high quality stored forage. The proliferation of bale wrappers (both individual and inline types) has made this technology more available to producers. Although the process for making baleage is well documented, producers have experienced poor fermentation and in some cases botulism toxicity from baleage. The parameters of good silage are well known and include pH below 5.0 and lactic acid concentrations above 3% on a DM basis. However, tests of farmer- produced baleage reveals that often these target values are not achieved. To …

Yellotas: A Unique Yellow Serradella Cultivar With Potential For Permanent Pasture Environments, G. Martin, Rowan W. Smith, E. J. Hall, Matthew Newell, R. Haling, R. C. Hayes
IGC Proceedings (1977-2023)
Yellow serradella (Ornithopus compressus L.) has been identified as a priority self-regenerating annual legume species for permanent pasture environments in south-eastern Australia. However, most yellow serradella genotypes exhibit high levels of hard seed and slow rates of hard seed breakdown, which reduces regeneration density in the years following the year of sowing . One cultivar, Yellotas, exhibits a much faster rate of hard seed breakdown and has been identified as one of only a handful of cultivars of that species with promising persistence in permanent pasture environments. In addition, this cultivar is substantially easier to de-hull than other cultivars …

A Solution To Weed Control In Grassland Containing White Clover, E. Bardsley, T. Schulz, R. Bentley, V. Van Damme, D. Gurney, N. Blanchier, B. B. Sleugh
IGC Proceedings (1977-2023)
Productive grass with white clover can lead to advantages both in forage quantity and quality, economics and in meeting wider expectations detailed in recent EU and UK policy. The ability to achieve this agronomic success is currently difficult due to a lack of options for broad spectrum weed control that also allow establishment or preservation of a white clover population. The aim of this paper is to demonstrate that 3730XL, developed by Corteva Agriscience, is a solution to this critical success factor. Data is presented from 16 efficacy trials (10 from established grassland and 5 from newly sown) where white …

Climate Change And Legume Performance In Grassland Agroecosystems, Lynn E. Sollenberger, M. M. Kohmann
IGC Proceedings (1977-2023)
We reviewed the literature to assess the effect of climate change factors on forage legumes. Whether growing in monoculture or mixtures with grasses, exposing legumes to elevated CO2 (eCO2) generally leads to sustained increases in forage accumulation (FA) and N fixation, but elevated temperature (eT) in conjunction with eCO2 usually reduces magnitude of these responses. In legumes, nodules represent large C sinks, precluding photosynthetic acclimation to eCO2 observed in non-N fixing plants. Greater N fixation in legume-grass mixtures exposed to eCO2 is due to greater percentage of legume N derived from symbiotic fixation and …

Cattle Diet Composition Under A Short- Duration Grazing With And Without Deferment In Puna Rangelands, R. E. Tácuna, E. R. Flores
IGC Proceedings (1977-2023)
The treatments of rest and deferment periods have shown to be effective tools to improve range condition, but little is known about their effect on the diet of animals. The present research aimed to evaluate the effect of the deferment period when applied to a short-term grazing system. The experimental design was a completely random factorial where the treatments resulted from combination of two grazing management systems: short-duration with deferment period (WD) and no deferral period (ND) both evaluated for three years. Forage yield (4.9 vs. 4.5 Tm DM ha-1 year-1) like range condition (55.5 vs. 50.7%) …

Pasture supplementation is an alternative to take advantage of the forage mass deferred in the rainy season and maintain or provide weight gain in periods of rain absence. The objective was to compare the structural characteristics, mass production, density and population demography of tillers of Urochloa brizantha cv. Paiaguás and Urochloa spp. cv. Convert under fixed and continuous stocking with steers supplemented with pelleted concentrate supplied in the trough or on the ground in the water/dry transition period. The experiment was carried out in the Jatai Federal University, from March to June 2020, in a completely randomized design and …

Harvesting hay as baleage can allow for more timely harvest, especially in spring months when curing conditions are not ideal. Forage conserved as baleage undergoes anaerobic fermentation in which the sugars are converted to lactic acid, lowering final pH. To successfully conserve forage as dry hay, moisture concentrations must be less than 18%. In contrast, the ideal moisture concentration for baleage is between 55 and 65%. There are a number of producers harvesting and wrapping hay that is not quite “dry enough” to bale but is too dry to ensile. This produces a product commonly referred to by producers as …
Cool-Season Perennials And Stability In Year-Round Forage Production Systems, C. D. Teutsch
Cool-Season Perennials And Stability In Year-Round Forage Production Systems, C. D. Teutsch
IGC Proceedings (1977-2023)
Changes in long-term climate normals have resulted in warmer and wetter summers and milder winters in the humid eastern United States. This will likely impact regional forage species adaptation in the long-term and varietal adaptation in the short term. Variety evaluation has been occurring at the University of Kentucky for almost 100 years. There are several considerations for selecting forage species and varieties including regional and local adaptation, productivity, distribution of growth, palatability, nutritive value, anti-quality factors, tolerance to stress, and persistence. Two of the most important criteria are long-term productivity and persistence under grazing, both of which are currently …
Impact Of Fertilizer Type, Seeding Coating, And Duration Of Exposure On The Germination Of Red Clover Seed, C. D. Teutsch, Edwin L. Ritchey
Impact Of Fertilizer Type, Seeding Coating, And Duration Of Exposure On The Germination Of Red Clover Seed, C. D. Teutsch, Edwin L. Ritchey
IGC Proceedings (1977-2023)
Legumes are important components in grassland ecosystems. Red clover is one the most used legumes in the transition zones states like Kentucky. To maintain legumes in grass pastures, improved red clover varieties are often overseeded in the late-winter or early-spring. In many cases seed is mixed with fertilizer and top-dressed onto pastures. Little data are available on the impact of fertilizer type or duration of exposure on the germination of raw and coated red clover seed. The objective of this study was to evaluate the impact of two fertilizer types, muriate of potash and a blended fertilizer (urea, diammonium phosphate, …
Effect Of The Hydrological Keyline Design On The Growth And Quality Of The Naturalized Grassland In The Dry Central Zone Of Chile, P. Toro-Mujica
Effect Of The Hydrological Keyline Design On The Growth And Quality Of The Naturalized Grassland In The Dry Central Zone Of Chile, P. Toro-Mujica
IGC Proceedings (1977-2023)
The effect of climate change in the central, semi-arid zone of Chile has altered precipitation patterns, shortening the growing season of the naturalized annual grasslands and reducing forage yields. Water harvesting techniques such as the hydrological keyline design, may improve the distribution of precipitation, reduce erosion, and enhance soil water harvesting. It may therefore reduce the impact of climate change in rainfed areas. A trial was implemented to evaluate the effect of the keyline water harvesting practice on soil variables, and pasture production and quality in three farms of the semi-arid O'Higgins region. Rains began in April and amounted to …
