Introduction: Lifetime exposure to interpersonal violence or abuse has been associated with several chronic diseases, including adult-onset diabetes, yet this pattern has not been confirmed by sex and race within a large cohort.\Methods: Data from the Southern Community Cohort Study collected between 2002−2009 and 2012−2015 were used to explore the relationship between lifetime interpersonal violence or abuse and diabetes (N=25,251). Prospective analyses of lower-income people living in the southeastern U.S. were conducted in 2022 to examine the risk of adult-onset diabetes associated with lifetime interpersonal violence or abuse by sex and race. Previous study from our lab has revealed a new role of CD47 in regulating adipose tissue function, energy homeostasis and the development of obesity and metabolic disease in CD47 deficient mice. In this study, the therapeutic potential of an antisense oligonucleotide (ASO) targeting to CD47 in obesity and its‑associated complications was determined in two obese mouse models (diet induced and genetic models). Although the presence of glycogen in platelets was established in the 1960s, its importance to specific functions (i.e., activation, secretion, aggregation, and clot contraction) remains unclear. Patients with glycogen storage disease often present with increased bleeding and glycogen phosphorylase (GP) inhibitors, when used as treatments for diabetes, induce bleeding in preclinical studies suggesting some role for this form of glucose in hemostasis. In the present work, we examined how glycogen mobilization affects platelet function using GP inhibitors (CP316819 and CP91149) and a battery of ex vivo assays. Platelet secretion requires Soluble N-ethylmaleimide Sensitive Attachment Protein Receptors (SNAREs). Vesicle SNAREs/Vesicle-Associated Membrane Proteins (v-SNAREs/ VAMPs) on granules and t-SNAREs in plasma membranes mediate granule release. Platelet VAMP heterogeneity has complicated the assessment of how/if each is used and affects hemostasis. To address the importance of VAMP-7 (V7), we analyzed mice with global deletions of V3 and V7 together or platelet-specific deletions of V2, V3, and global deletion of V7. We measured the kinetics of cargo release, and its effects on three injury models to define the context-specific roles of these VAMPs. Loss of V7 minimally affected dense and α …

Influenza-A virus (IAV) infects yearly an estimated one billion people worldwide, resulting in 300,000–650,000 deaths. Preventive vaccination programs and antiviral medications represent the mainstay of therapy, but with unacceptably high morbidity and mortality rates, new targeted therapeutic approaches are urgently needed. Since inflammatory processes are commonly associated with measurable changes in the cell membrane potential (Em), we investigated whether Em hyperpolarization via TREK-1 (K2P 2.1) K þ channel activation can protect against influenza-A virus (IAV)-induced pneumonia. We infected mice with IAV, which after 5 days caused 10–15% weight loss and a decrease in spontaneous activity, representing a clinically relevant infection. …

Platelets contribute to COVID-19 clinical manifestations, of which microclotting in the pulmonary vasculature has been a prominent symptom. To investigate the potential diagnostic contributions of overall platelet morphology and their α-granules and mitochondria to the understanding of platelet hyperactivation and micro-clotting, we undertook a 3D ultrastructural approach. Because differences might be small, we used the high-contrast, high-resolution technique of focused ion beam scanning EM (FIB-SEM) and employed deep learning computational methods to evaluate nearly 600 individual platelets and 30 000 included organelles within three healthy controls and three severely ill COVID-19 patients. Vascular smooth muscle cells (vSMCs) exert a critical role in sensing and maintaining vascular integrity. These cells abundantly express the low-density lipoprotein receptor–related protein 1 (LRP1), a large endocytic signaling receptor that recognizes numerous ligands, including apolipoprotein E–rich lipoproteins, proteases, and protease-inhibitor complexes. We observed the spontaneous formation of aneurysms in the superior mesenteric artery (SMA) of both male and female mice in which LRP1 was genetically deleted in vSMCs (smLRP1–/– mice). Quantitative proteomics revealed elevated abundance of several proteins in smLRP1–/– mice that are known to be induced by angiotensin II–mediated (AngII-mediated) signaling, suggesting that this pathway was dysregulated. …

Aims The metabolic failure of macrophages to adequately process lipid is central to the aetiology of atherosclerosis. Here, we examine the role of macrophage angiotensin-converting enzyme (ACE) in a mouse model of PCSK9-induced atherosclerosis.
Methods and results Atherosclerosis in mice was induced with AAV-PCSK9 and a high-fat diet. Animals with increased macrophage ACE (ACE 10/10 mice) have a marked reduction in atherosclerosis vs. WT mice. Macrophages from both the aorta and peritoneum of ACE 10/10 express increased PPARα and have a profoundly altered phenotype to process lipids characterized by higher levels of the surface scavenger receptor CD36, increased uptake of …

Traumatic brain injury (TBI) continues to be a significant cause of morbidity and mortality worldwide. Despite significant progress in understanding the complex pathophysiology of TBI, the underlying mechanisms remain poorly understood. The primary brain damage is acute and irreversible. However, secondary brain injuries often develop gradually over months to years, creating an opportunity for critical therapeutic interventions. Conventional understandings of perception have long undergirded traditional readings of Modernist texts in adopting a predominantly subject-centered perspective that separates subject from object in a vertical and hierarchical relationship. I argue that a consideration of Virginia Woolf’s short stories in dialogue with four generations of twentieth-century Catalan women writers who followed her work closely suggests an entirely different epistemological framework of perception in which subject and object are fluidly and horizontally organized. Mercè Rodoreda, Concha Alós, Montserrat Roig and Carme Riera establish a horizontal fictional dialogue that constitutes a return to matter that decenters the subject, resulting in an alternative …

Elucidating the relationship of the gut microbiome in Alzheimer's Disease (AD) risk and pathogenesis is an area of intense interest. Since 60 to 80% of AD risk is related to genetics and APOE alleles represent the most impactful genetic risk factors for AD, their mechanism(s) of action are under intense scrutiny.
First, I conducted a study on APOE targeted replacement mice to investigate the impact of APOE alleles on the murine gut microbiome. The state of Kentucky is home to many rural counties which experience high levels of outward migration due to their relatively unfavorable economic conditions. While migration trends nationally have begun to plateau, migration flows from county to county show a much more volatile story. This study will examine the relationship between economic opportunity and migration flow estimates in Kentucky’s counties through a multiple regression approach with the response variable being annual migration flow estimates, with multiple predictor variables showing the economic composition of the county. Collecting, analyzing, and reporting accurate, reliable data is necessary for achieving health equity. Although data alone will not lead to equitable health outcomes for all people, it helps health professionals, researchers, and policymakers understand health disparities and develop effective interventions, programs, and policies to reduce them. Necessary data includes disaggregated health information on diverse demographics, including people with disabilities. However, despite this need, disability data gaps exist. Fortunately, numerous solutions exist to address this issue, including implementing minimum data standards on disability and engaging all levels of health organizations in data practices.

The heavy element content (“metallicity”) of the Universe is a record of the total star formation history. Gas-phase metallicity in galaxies, as well as its evolution with time, is of particular interest as a tracer of accretion and outflow processes. However, metallicities from the widely-used electron temperature (Te) method are typically ∼ 2× lower than the values based on the recombination line method. This “abundance discrepancy factor” (ADF) is well known and is commonly ascribed to bias due to temperature fluctuations. We utilize medium-resolution JWST/NIRSpec observations of 164 galaxies at z = 2.0-9.3 from the Cosmic Evolution Early Release Science (CEERS) survey to investigate the evolution of the excitation and ionization properties of galaxies at high redshifts. Our results represent the first statistical constraints on the evolution of the [O III]/Hβ versus [N II]/Hα, [S II]/Hα, and [O I]/Hα “BPT” diagrams at z > 2.7, and the first analysis of the O32 versus R23 diagram at z > 4 with a large sample. In recent research, there has been a growing interest in understanding the impact of informative cluster size (ICS) on statistical inference for clustered data. In the non-parametric context, the problem for testing equality of distribution functions has been the main consideration. We are aiming to develop inferential procedures for the Wilcoxon-Mann-Whitney effect, also known as the non-parametric relative effect, involving two or more groups. Computationally, results from both simulated and real-world data have shown promising results that our proposed tests effectively account for ICS and they particularly outperform other methods in the literature designed for ignorable cluster sizes. Background: Performance-based tests for patients with anterior shoulder dislocation are lacking. This study determined the reliability and validity of the supine moving apprehension test designed to assess the ability to control anterior instability loads. Methods: Thirty-six participants were recruited (18 healthy individuals, and 18 patients following anterior shoulder dislocation). Healthy participants performed the supine moving apprehension test on 2 separate occasions to determine test-retest reliability. Patients completed the supine moving apprehension test and the Western Ontario Shoulder Instability index before and 6 months after surgical stabilization of their shoulder. The genetic etiologies of common diseases are highly complex and heterogeneous. Classic methods, such as linear regression, have successfully identified numerous variants associated with complex diseases. Nonetheless, for most diseases, the identified variants only account for a small proportion of heritability. Challenges remain to discover additional variants contributing to complex diseases. Expectile regression is a generalization of linear regression and provides complete information on the conditional distribution of a phenotype of interest. Hans Osieck (1910-2000) was a Dutch composer, pianist, and teacher. His musical portraits, Acht Korte Karakterschetsen voor piano vierhandig (Eight Short Character Sketches for Four Hands), are a valuable addition to the piano duo repertoires for intermediate and advanced students. In Eight Sketches, Osieck musically portrays eight of his students using simple compositional techniques, much like the brushstrokes and color palette of an artistic portrait. As a result of their programmatic nature and relatable subjects, Osieck’s Eight Sketches can be particularly useful for students developing their imaginations and learning to perform expressively. The purpose of this project is introducing South Korean composer Seung Ki Hong’s Arirang Variations (2015). Hong’s post-2013 work has focused on incorporating traditional Korean melodies into Western instrumental music; Arirang Variations exemplifies this process. The theme is “Arirang,” a folksong that Koreans have sung to express their joys and sorrows for centuries. Hong transforms this iconic melody through a series of variations in various dance styles such as a tango and a Viennese waltz.

This project considers the Cambodian tocay gecko’s cry as the motif which embodies the harmonic structure in Chaconne for Tokkae for Violin and Piano, Op. 96 by Dr. Myung Whan Johann Kim.
