The McMurdo Dry Valleys (DVs) of South Victoria Land, Antarctica, constitute the largest ice-free region on the continent and one of Earth’s most Mars-analog environments. Their hyper-arid polar desert conditions offer a unique setting for investigating surface weathering and mineralogical processes under extreme climates. This study presents the first regional-scale mapping of alteration and crystalline weathering minerals across the McMurdo DVs. Synovitis is a key contributor to the development of OA, and early modulation of the synovial environment may help limit downstream cartilage damage. This study compared the clinical and biochemical effects of intra-articular triamcinolone acetonide (TA) and autologous conditioned serum (ACS) in an equine model of IL-1β–induced synovitis. Six healthy adult horses were used in a crossover design involving five groups: PBS (negative control), IL-1β (positive control), IL-1β + ACS, IL-1β + TA, and an exploratory ACS-alone group administered post hoc to isolate its effects without IL-1β interference. Expanding the adoption of anaerobic digestion (AD) is limited by system instabilities caused by feedstock composition and variability. Understanding how changes in feedstock and operational parameters influence microbiome dynamics is essential for developing biology-driven strategies to improve digester stability.
Two semi-continuous reactors, an experimental and a control, operated over 39 weeks across five phases with varying feedstocks and organic loading rates. Substrate-specific enrichment cultures were developed for bioaugmentation and implemented in select phases to assess its impact. Dextrorphan is a Phase I equine metabolite of dextromethorphan, a widely used anti-tussive / cough suppressant medication marketed and available in numerous over-the-counter formulations in the United States. Dextromethorphan is an Association of Racing Commissioners International (ARCI) drug class 4 penalty class B substance and a Horseracing Integrity and Welfare Authority (HIWU) CONTROLLED S7 B substance and is regulated as such in horse racing. In horses, dextromethorphan is metabolized to its Phase I metabolite, dextrorphan, which is next glucuronidated and the resultant dextrorphan-glucuronide conjugate then excreted at remarkably high concentrations in equine urine. Amidst the opioid crisis, understanding the genetic basis of opioid use disorder (OUD) is crucial for identifying biological mechanisms and intervention points. However, genome-wide association studies (GWASs) have been hampered by inadequate sample sizes and often the use of control populations not assessed for prior opioid exposure. Because opioid exposure is a prerequisite for the development of OUD, consideration of exposure history in controls is important. Electronic health record data (EHR) paired with genomic information allow a broader sampling of patients with OUD and exposed controls. Metabolic homeostasis is maintained through tightly coordinated signaling networks that integrate diverse physiological inputs. Central to this regulation are transcription factors, which sense nutrient, hormonal, and vitamin-derived signals and translate them into coordinated gene expression programs. Transcription factor activity is governed by DNA binding, recruitment of coregulatory proteins, and interactions with specific response elements in gene promoters, which allows them to selectively modulate diverse metabolic pathways. Owing to their multifactorial and tunable nature, transcription factors have emerged as promising targets for the treatment and diagnosis of metabolic diseases, including metabolic dysfunction-associated steatotic liver disease (MASLD). Novacetimonas hansenii is a model organism for bacterial cellulose (BC) production, a biopolymer with broad industrial applications. To better understand its carbon source-dependent metabolism, we conducted the first time-resolved transcriptomic analysis of N. hansenii ATCC 23769 grown on five carbon sources (glucose, fructose, mannitol, galactose, and sucrose) across five time points (24–120 h). Glucose, fructose, and mannitol supported growth and efficient BC production, whereas galactose and sucrose resulted in poor yields, likely due to limited transport and enzymatic capacity. Solid-state NMR revealed that cellulose crystallinity and polymorphism were carbon source-dependent, with reduced bcsD expression correlating with lower crystallinity. Transcriptomic profiling …

Eggplant exhibits a diverse range of fruit colors, making it an excellent model for studying fruit pigmentation and its genetic regulation. While genes responsible for green and photosensitive purple fruit have been identified, the genetic basis of the nonphotosensitive (NPS) fruit trait in eggplant has remained elusive. In this study, we characterized a major quantitative trait locus (QTL), SmNPS10.1, on chromosome 10 using QTL-seq. By combining linkage-based gene mapping with progeny testing, we fine-mapped SmNPS10.1 to a 33.58-kb interval, within which we identified SmMYB113, an R2R3-MYB transcription factor that regulates anthocyanin biosynthesis, as the candidate gene. The selective forces shaping parental care have been studied for over 50 years. While theoretical and experimental work has yielded qualitative progress, the large body of empirical work testing predictions about parental investment based on life-history trade-offs has yet to be synthesized. We first provide an overview of the core life-history theory exploring how selection might shape parental care. We then conduct a systematic review and meta-analysis on studies that experimentally manipulated brood size in birds, a widely used experimental approach to manipulate parental investment. The aim of this study was to test relationships between adolescent insecure attachment (anxious and avoidant) with mothers and fathers and three outcomes: food addiction, eating disorders, and weight status, in a large multi-regional sample of Czech adolescents. Anonymous, self-reported data were collected from 3,677 adolescents (1,688 boys, 45.9% and 1,989 girls, 54.1%). Both anxious and avoidant attachment with mothers were positively associated with food addiction (p <.001) and eating-disorder risk (p <.001). Importantly, avoidant attachment with fathers uniquely and additively explained variance in both food addiction (p <.001) and eating-disorder risk (p =.003), after controlling for maternal attachment. Weight status was unrelated to attachment and was predicted by sex and parental education only. Study findings highlight the salience of insecure attachment with both mothers and fathers for food addiction and eating disorders among adolescents and identify the parental relationship as a key potential preventative strategy.

Adverse childhood experiences (ACEs), which refer to exposure to traumatic life events before age 18, and instability in caregiving experiences contribute to poor physical, mental, behavioral, and developmental health outcomes for both children and adults. Previous research shows that these experiences can have negative effects both in the short term and throughout a person’s life. However, children’s strengths, which are the internal and external assets that support their development, can help protect against the impact of ACEs. Maintaining a membrane electrical potential of biological cells is a dynamic process, as some cells have a continually changing potential, like pacemaker cells, while other cells may function with large or small changes in the membrane potential. Additionally, some cells may change their electrical potential when stimulated or inhibited by electrical signals, chemical compounds, or both—either simultaneously or episodically. The persistent leak of K+ through two-pore-domain potassium channels (K2P) and of Na+ through Na+ leak channels (NALCNs) and the action of pumps and exchangers are primarily responsible for maintaining a resting potential. Systematists have long been fascinated by the astounding variation in species diversity across the various branches of the tree of life, a net result of the uneven rates at which lineages undergo speciation and extinction over time. The past 30 years have seen the development and widespread application of tools that allow diversification to be quantified and characterized in empirical data sets. Male mice fed high-fat diet become obese, but female mice are resistant to diet-induced weight gain. We previously found that circulating estradiol in females protects their daily rhythms from disruption by high-fat feeding to prevent diet-induced obesity. The goal of this study was to determine the effects of estradiol on daily metabolic rhythms in male mice. Male C57BL/6J mice were treated with estradiol and fed high-fat diet for 2 weeks. Measures of rodent drinking (lick) patterns are used to test how various manipulations affect fluid intake. Decades of research indicate that manipulating the satiating potency of a stimulus affects the number of licking bursts whereas manipulating the hedonic value of the solution affects the size of the licking bursts or rate of licking. This type of analysis has been largely applied to manipulations that modulate drinking that is already occurring (i.e., indirect controls or modulators of intake), but there is a dearth of studies with detailed measures of licking due to stimuli that are primary drivers of intake which happen …

Three research poems are created as an autoethnographic response to feedback on a research article on the author’s experiences as a gender non-conforming person in women’s public restrooms. The feedback loop includes the sharing of lived experiences by others and theoretical (why?) discussions in the context of the gendered policing and politicalization of women’s restrooms. The first found poem includes words, phrases, and descriptions of images used to label spaces designated as women’s restrooms. Chronic alcohol consumption increases the risk of osteoporosis and fracture by disrupting bone remodeling, in part by enhancing osteoclastogenesis. However, the cellular mechanisms underlying this process remain incompletely defined. We analyzed scRNA-seq data from osteoclasts differentiated in vitro from bone marrow mononuclear cells obtained from macaques following 12 months of chronic ethanol or isocaloric control solution consumption. Module scoring, trajectory inference with generalized additive modeling (tradeSeq), and CellChat-based analyses of intercellular communication were applied to uncover ethanol-induced changes in metabolic reprogramming, lineage progression, and signaling network dynamics. Studies on the earliest life stages are essential to our ecological understanding of avian demography; however, monitoring technologies that allow tracking of small birds are still limited in a variety of ways. One critical limitation, until recently, has been the development of methods for attaching transmitters to young birds with precocial development (e.g., Galliformes, Charadriiformes, etc.). Indeed, chronic alcohol consumption (CAC) rewires macrophages/monocytes toward a hyper-inflammatory phenotype. Prior studies showed increased gut permeability and dysbiosis. Translocation of host- and microbial-derived metabolites could trigger the hyper-inflammatory responses generated by macrophages/monocytes. However, the exact changes in these metabolites remain poorly defined due to confounders that complicate clinical studies and the differences between human and rodent gut microbiomes.

Purpose: Pharmacoepidemiology is a key discipline for evidence-based decision-making, yet its educational programs have not been systematically assessed for alignment with international core competencies. Our objectives were to evaluate curricula alignment of pharmacoepidemiology training programs with ISPE-recommended core competency themes and identify key curriculum content development areas.
Methods: The ISPE Core Competencies Workgroup and ISPE leadership developed a curriculum and needs assessment survey, incorporating feedback from global members. The survey was electronically distributed to leaders of pharmacoepidemiology training programs (e.g., department chairs and program directors) using the ISPE contact database, followed by snowball sampling to enhance representation. Objective: Brief behavioral interventions targeting pain (Physical Self-Regulation, PSR) or sleep (Brief Behavioral Intervention for Insomnia, BBTI) show promise for interdisciplinary management of chronic pain, though their feasibility with chronic musculoskeletal orofacial pain (MSK-OFP) populations remains underexplored. This pilot study assessed the feasibility of a randomized clinical trial (RCT) of these interventions in a university-affiliated tertiary OFP clinic and explored clinical changes in pain, sleep, and other secondary outcomes.

This study aimed to elucidate the mechanism by which colonic fecal microbiota transplantation (FMT) improves lipid metabolism in low-birth-weight neonatal calves via modulation of the gut microbial ecosystem. In a completely randomized design, seventy newborn Holstein calves (1 day old, 32.63 ± 3.13 kg BW) were assigned to two groups (35 calves/treatment; 30 females and 5 males per group). Aim:
Molar-Incisor (PerioC-MIP) and Generalized (PerioC-G) Grade C Periodontitis could have distinctive etiopathogenesis behind their unique clinical patterns. Thus, this study aimed to distinguish these two phenotypes by analyzing the subgingival metagenomic profile and the inflammatory markers levels.
Methods:
In this cross-sectional comparative study, Gingival Crevicular Fluid (GCF) and Subgingival Biofilm (SB) were collected from 18 PerioC-MIP North Americans and 14 periodontally healthy controls (HC) from the same location (HC-MIP) and 20 PerioC-G Brazilians and 20 controls (HC-G). From GCF, immunoenzymatic analysis was performed. SB functional and taxonomic bacterial content was determined using shotgun metagenomics sequencing.

Immunotherapies have greatly improved outcomes for patients with renal cell carcinoma (RCC), yet response rates remain suboptimal and the factors promoting therapy resistance versus sensitivity are incompletely understood. Currently, no preclinical model of orthotopic renal cancer exists that permits evaluation of tumor antigen–specific (TAS) CD8+ tumor-infiltrating lymphocytes (TILs). To address this deficiency, we developed a mouse renal cancer model that permits tracking of adoptively transferred TAS CD8+ T cells. Prostate cancer progression and therapeutic resistance are closely linked to alterations in redox homeostasis and cell cycle regulation. Nuclear factor erythroid 2–related factor 2 (NRF2) is a master regulator of antioxidant responses, yet the mechanisms integrating cell cycle signaling with NRF2 activity in prostate cancer remain poorly defined. In this study, we identify polo‑like kinase 1 (PLK1) as a novel regulator of NRF2 through direct phosphorylation at serine 253 (S253). Using bioinformatic prediction, mass spectrometry, in vitro kinase assays, and cell‑based analyses, we demonstrate that PLK1 phosphorylates NRF2 at S253 in a cell cycle–dependent manner, with maximal phosphorylation during mitosis. …

Maternally inherited bacterial endosymbionts are common in arthropods. Many of these bacteria manipulate host reproduction to favor the production of infected females, thereby increasing bacterial prevalence in the host population, even if cost is incurred to the host. However, there are also many factors that may inhibit symbionts’ ability to spread in host populations, including environmental constraints, symbiont-imposed costs on the host, interactions with co-infecting bacteria, or host biology. Dung beetles provide ecosystem services in pasture systems but are vulnerable to chemical residues in livestock dung. Parasiticides containing macrocyclic lactones (MLs) are used to control parasites in livestock and are excreted unmetabolized in dung, exposing non-target organisms to these compounds. We used a multi-scale approach to examine whether ML residues alter dung beetle foraging ecology. At the field scale, we assessed dung beetle abundance and community composition using traps baited with dung from untreated cattle and cattle treated with ivermectin, moxidectin, or eprinomectin across eight farms and natural areas. Hollow stems have independently evolved multiple times across the plant kingdom and play crucial roles in plant development and various environmental adaptations. However, the mechanisms underlying stem hollowness remain poorly understood. Water spinach (Ipomoea aquatica) is one of the few hollow-stemmed plants in the Convolvulaceae family (eudicot: asterid), and its hollow stems are essential for thriving in aquatic environments. Using histochemical staining and transcriptome analysis, we found that programmed cell death (PCD) is involved in cavity formation at water spinach shoot tips. Single-cell and spatial transcriptome analyses further revealed that ethylene and reactive oxygen species (ROS) likely drive …
