A novel process for enhancing activity of an oligopeptide or polypeptide comprising the steps of: providing an oligopeptide or polypeptide, dissolving the oligopeptide or polypeptide in an organic solvent, heating, removing the solvent, and recovering an oligopeptide or polypeptide with enhanced activity is disclosed. Also disclosed are novel oligopeptides and polypeptides enhanced by the process according the invention.

Viral, endoparasitoid and/or host genes that specifically inhibit the immune response of insect pests, useful for broadening the host range of insect viruses. Symbiont viruses of insect pests are genetically modified to express immune-suppressing proteins or biologically active fragments thereof and, optionally toxins, to increase the virus host range and/or improve the efficacy of insect pathogens.

Disclosed is a chimeric isoprenoid synthase polypeptide including a first domain from a first isoprenoid synthase joined to a second domain from a second, heterologous isoprenoid synthase, whereby the chimeric isoprenoid synthase is capable of catalyzing the production of isoprenoid reaction products that are not produced in the absence of the second domain of the second, heterologous isoprenoid synthase. In vertebrates, many cytokines and growth factors have been identified as activators of the JAK/STAT signaling pathway. In Drosophila, JAK and STAT molecules have been isolated, but no ligands or receptors capable of activating the pathway have been described. We have characterized the unpaired (upd) gene, which displays the same distinctive embryonic mutant defects as mutations in the Drosophila JAK (hopscotch) and STAT (stat92E) genes. Upd is a secreted protein, associated with the extracellular matrix, that activates the JAK pathway. We propose that Upd is a ligand that relies on JAK signaling to stimulate transcription of pair-rule genes in a …

Thiazolidinediones Inhibit Lipoprotein Lipase Activity In Adipocytes, Subramanian Ranganathan, Philip A. Kern

The thiazolidinediones troglitazone and BRL 49653 improve insulin sensitivity in humans and animals with insulin resistance. Adipose tissue lipoprotein lipase is an insulin-sensitive enzyme. We examined the effects of thiazolidinediones on lipoprotein lipase expression in adipocytes. When added to 3T3-F442A, 3T3-L1, and rat adipocytes in culture, troglitazone and BRL 49653 inhibited lipoprotein lipase activity. This inhibition was observed at concentrations as low as 0.1 μM and within 2 h after addition of the drug. Lipoprotein lipase activity was inhibited in differentiated adipocytes as well as the differentiating cells. Despite this decrease in enzyme activity, these drugs increased mRNA levels in …

Pesticides used in urban environments can be transported in runoff to downstream waters and cause adverse environmental consequences. This experiment assessed the effects of post-application irrigation depth (0, 6.4, and 12.7 mm) and formulation (liquid and granular) on concentration and transport of diazinon (a pesticide commonly used for lawn insect control) in runoff from “tall” fescue (Festuca arundinacea Schreb.) plots. The post-application irrigation was applied using rainfall simulators immediately following diazinon application. The rainfall simulators were again used approximately 2 h after diazinon application to apply the equivalent of a heavy rainfall (64 mm/h for approximately 1.5 h) to generate …

The Legionnaires' disease bacterium, Legionella pneumophila, is a facultative intracellular pathogen that invades and replicates within two evolutionarily distant hosts, free living protozoa and mammalian cells. Invasion and intracellular replication within protozoa are thought to be major factors in the transmission of Legionnaires' disease. We have recently reported the identification of a galactose/N-acetyl-d-galactosamine (Gal/GalNAc) lectin in the protozoan host Hartmannella vermiformis as a receptor for attachment and invasion by L. pneumophila (Venkataraman, C., B.J. As a part of a National Pork Producers Council educational program, our research and extension team at the University of Kentucky was linked with an independent commercial swine producer to test the NCPIG model against observed commercial on-farm data. This experience provided improved information for model development as well as increased producer insight into the data input needs and potential benefits of modeling. Detailed production information comparisons between the NCPIG model and producer data are presented for summer time conditions to assess the validity of the NCPIG model for simulation of grow-finish swine performance. A simple and implantable sustained release drug delivery device with an inner core containing an effective amount of a low solubility agent covered by a non-bioerodible polymer coating layer that is permeable to the low solubility agent is disclosed. A method for treating a mammal to obtain a desired local or systemic physiological or pharmacological effect by surgically implanting such a sustained release drug delivery device into a mammal in need of treatment is also disclosed.

NIMA promotes entry into mitosis in late G2 by some mechanism that is after activation of the Aspergillus nidulans G2 cyclin-dependent kinase, NIMXCDC2/NIMECyclin B. Here we present two independent lines of evidence which indicate that this mechanism involves control of NIMXCDC2/NIMECyclin B localization. First, we found that NIMECyclin B localized to the nucleus and the nucleus-associated organelle, the spindle pole body, in a NIMA-dependent manner. Analysis of cells from asynchronous cultures, synchronous cultures, and cultures arrested in S or G2 showed that NIMECyclin B was predominantly nuclear during interphase, with maximal …

We have investigated the controversial involvement of components of the SNARE (soluble N-ethyl maleimide–sensitive factor [NSF] attachment protein [SNAP] receptor) machinery in membrane traffic to the apical plasma membrane of polarized epithelial (MDCK) cells. Overexpression of syntaxin 3, but not of syntaxins 2 or 4, caused an inhibition of TGN to apical transport and apical recycling, and leads to an accumulation of small vesicles underneath the apical plasma membrane. All other tested transport steps were unaffected by syntaxin 3 overexpression. Botulinum neurotoxin E, which cleaves SNAP-23, and antibodies against α-SNAP inhibit both TGN to apical and basolateral transport in …

Multidirectional Ecg Coherent Optimal Timing Of Defibrillation Shocks, Abhijit R. Patwardhan, Fabio M. Leonelli

A method for of delivering a defibrillation shock to a heart at an optimal time to stop ventricular fibrillation which involves obtaining an electrocardiogram of a heart in at least two directions, determining a time-coherency of the electrocardiogram based upon each of the at least two directions, and computing a tracking function from the time-coherency. An optimal time to apply a defibrillation shock to the heart is determined by locating a local maximum on the tracking function. The method utilizes spacia characteristics of the ventricular fibrillation. The method can be incorporated into implantable cardioverter defibrillators utilizing existing hardware technology.

The gene sequence for ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) large subunit (LS) εN-methyltransferase (protein methylase III or Rubisco LSMT) is disclosed. This enzyme catalyzes methylation of the ε-amine of lysine-14 in the large subunit of Rubisco. In addition, a full-length cDNA clone for Rubisco LSMT is disclosed. Transgenic plants and methods of producing same which (1) have the Rubisco LSMT gene inserted into the DNA, and (2) have the Rubisco LSMT gene product or the action of the gene product deleted from the DNA are also provided. Spring dead spot is the most destructive disease of bermudagrass in Kentucky. The most serious outbreaks occur under high maintenance conditions; e.g., high nitrogen fertility, low mowing height, and frequent traffic. Moderate to severe outbreaks can occur under low-maintenance conditions as well.
High levels of resistance to spring dead spot in bermudagrass varieties have not yet been demonstrated experimentally. Limited data suggest that those varieties with best winter hardiness are somewhat less susceptible to the disease. To date, no confirmed cases of spring dead spot have been documented in “Quickstand” bermudagrass, which may indicate that this variety will suffer less …

Three spatial data sets consisting of high spatial resolution (1 m) remote sensing images acquired in 12 spectral bands, an on-the-go yield map, and a Digital Elevation Model were co-registered and evaluated for spatial variability studies in a Geographic Information Systems environment. Separate on-the-go yield maps were developed for 3, 5, and 12 statistically significant mean yield classes. For each yield class, the corresponding mean spectral and elevation data were extracted. The relationship between mean spectral and yield data was strongly linear (r = 0.99). Also, a strong linear relationship between mean yield and elevation data (r = 0.92) was …

A method is provided for reducing the amount of wear particulates generated by a total joint orthopaedic implant. The implant consists of two matched articulating components wherein at least one of the components is made of polymer or other material with similar physical properties. The method includes the steps of placing the total joint orthopaedic implant in a fluid bath and articulating the implant in the fluid bath for at least 1,000 cycles. The articulation may be performed in three stages. In the first, the implant is articulated under a load of substantially 0.1-500 Newtons with sliding speeds of substantially …
