Pharmaceutically Engineered Nanoparticles For Enhancing Immune Responses To Hiv-1 Tat And Gag P24 Proteins, Jigna D. Patel

These studies were aimed at investigating the potential application of nanoparticles engineered from oil-in-water microemulsion precursors for enhancing immune responses to HIV-1 Tat and Gag p24 proteins. Both of the HIV-1 proteins have been reported to be critical in the virus life cycle and are being evaluated in clinical trials as vaccine candidates. Anionic nanoparticles were prepared using emulsifying wax as the oil phase and Brij 78 and sodium dodecyl sulfate as the surfactants. The resulting nanoparticles were coated with Tat and were demonstrated to produce superior immune responses after administration to BALB/c mice compared to Tat adjuvanted with Alum. …

In this digital age, it is more important than ever to have good methods for processing images. We focus on the removal of blur from a captured image, which is called the image deblurring problem. In particular, we make no assumptions about the blur itself, which is called a blind deconvolution. We approach the problem by miniming an energy functional that utilizes total variation norm and a fidelity constraint. Thimet oligopeptidase (TOP, EC 3.4.24.15) and neurolysin (EC 3.4.24.16) are zincdependent metallopeptidases that metabolize small bioactive peptides. The two enzymes share60 % sequence identity and their crystal structures demonstrate that they adopt nearly identicalfolds. They generally cleave at the same sites, but they recognize different positions on somepeptides, including neurotensin, a 13-residue peptide involved in modulation of dopaminergiccircuits, pain perception, and thermoregulation.On the basis of crystal structures and previous mapping studies, four residues(E469/R470, M490/R491, H495/N496, and R498/T499, TOP residues listed first) in thesubstrate-binding channel appear positioned to account for differences in specificity. Although attractive interactions between π systems (π-π interaction) have been known for many years, understanding of its origin is still incomplete. Quantitative measuring of π-stacking is challenging due to the weak nature of the π-π interaction. This dissertation aims at elucidating a quantitative conformational analysis by NMR ring current anisotropy of an organic compound capable of intramolecular π-stacking in solution and studying charge effects on the stacking of π-systems. The decision to relocate or to age in place can be a difficult one, mitigated by a variety of influencing factors such as finances, physical abilities, as well as social and instrumental support from family and others. This study focuses on the stresses of residential relocation to independent and assisted living facilities among older women living in Lexington, Kentucky. Participation entailed three semi-structured interviews as well as saliva and blood sampling over a period of 6 months, beginning within one month of the move. This project utilized synthesis and in vitro assays to generate antagonist SARs at various nAChR subtypes. Alkylation of the pyridino nitrogen of the nicotine molecule afforded subtype specific antagonists at a42* nAChR subtypes and nAChR subtypes that mediate nicotine-evoked dopamine release. Using this data, a series of mono-azaaromatic quaternary salts were produced and evaluated in binding and functional assays for a42* and a7* nAChR subtypes and nAChR subtypes that mediate nicotine-evoked dopamine release. Additionally, bis-azaaromatic quaternary salts were synthesized and evaluated in the same assays. Two potent lead compounds were identified. This theses presents the synthesis and characterization of Cd(II)-, Pb(II)- and Hg(II)-aminoethanethiolates in aqueous media. 2-Aminoethanethiolate, a versatile sulfur andnitrogen (S/N) based ligand was used due to its resemblance to the naturally occurringamino acid, cysteine. Cellular proliferation is a key characteristic of hematopoietic stem and progenitor cells (HSC/HPCs) that allows for the production of all blood cell lineages during an individuals lifetime. While this feature of stem cells is strictly regulated during steadystate and stress hematopoiesis, it also contributes to the development of myeloproliferative disorders, such as chronic myelogenous leukemia, essential thrombocythemia, and polycythemia vera. It should come as no surprise then, that common treatments for these diseases often target the proliferative nature of the dysfunctional HSC/HPCs. Colleotrichum graminicola is the causal agent of anthracnose stalk rot, and is one of the most common and aggressive pathogens of maize. The goal of my Ph.D. project was to contribute to a better understanding of the biology of the interaction between C. graminicola and its host. C. graminicola produces two type of asexual spores: one is produced on the surface of infected tissues and is thought to be involved in the spread of the disease in the field. USA soybean germplasm has a narrow genetic base that could be augmented by alleles from the wild species Glycine soja which positively influence agronomic traits. The objective of this study was to identify such alleles for yield and yield component QTL (quantitative trait loci). Two populations of 150 BC2F4 lines were generated from a mating between recurrent parent Glycine max 7499 and donor parent Glycine soja PI 245331 with one line in each population tracing back to the same BC2 plant. The site-specific management of alfalfa has not been well-evaluated, despite the economic importance of this crop. The objectives of this work were to i) characterize the effects of soil moisture deficits on alfalfa and alfalfa yield components and ii) evaluate the use of canopy reflectance patterns in measuring treatment-induced differences in alfalfa yield. Ethnic identity is a significant psychological variable for the study of African Americans in the United States and often associated with psychological health. However, the nature of this relationship is sometimes unclear. One reason for the confusion may be that ethnic identity is often confounded with acculturation as they are sometimes used interchangeably in research. Zero-valent metals such as bulk iron and zinc are known to dechlorinate toxicorganic compounds. Enhancement in reaction rates has been achieved through bimetallicnanosized particles such as nickel/iron (Ni/Fe) and palladium/iron (Pd/Fe). Batchdegradation of model compounds, trichlroethylene (TCE) and 2,2'-dichlorobiphenyls(DCB), were conducted using bimetallic Ni/Fe and Pd/Fe nanoparticles. Completedegradation of TCE and DCB is achieved at room temperature. Zero-valent iron, as themajor element, undergoes corrosion to provide hydrogen and electrons for the reductivecatalytic hydrodechlorination reaction. The second dopant metals of nickel and palladium(in nanoscale) act as catalyst for hydrogenation through metal hydride formation thatproduces completely dechlorinated final product. Protein farnesylation is an essential post-translational modification required for the function of numerous cellular proteins including the oncoprotein Ras. The farnesyl transferase (FTase) catalyzed reaction is unique because farnesyl diphosphate (FPP), the farnesyl group donor for the reaction, forms a significant portion of a target protein binding site. The major goal of this research was to exploit this unique property of the FTase reaction and determine if changing the structure of the farnesyl donor group would affect FTase protein targeting. The Learning Classifier System (LCS) and its descendant, XCS, are promising paradigms for machine learning design and implementation. Whereas LCS allows classifier payoff predictions to guide system performance, XCS focuses on payoff-prediction accuracy instead, allowing it to evolve "optimal" classifier sets in particular applications requiring rational thought. This research examines LCS and XCS performance in artificial situations with broad social/commercial parallels, created using the non-Markov Iterated Prisoner's Dilemma (IPD) game-playing scenario, where the setting is sometimes asymmetric and where irrationality sometimes pays. Yellow perch (Perca flavescens) exhibit an estrogen stimulated sexual sizedimorphism (SSD) wherein females grow faster and larger than males. In an effort togain better understanding of this phenomenon, several genes associated with sexualdevelopment, reproduction and growth were cloned, including prolactin (PRL),somatolactin (SL), insulin-like growth factor-I (IGF-Ib), the estrogen receptors (ER?? andER??a) and ovarian aromatase (CYP19A1). The studies presented in this dissertation were conducted with the objective ofgaining greater understanding into the mechanisms of successful aging, the role ofmitochondria dysfunction in traumatic brain injury, and also on the mechanisms ofimproved learning and cognitive function in the aging.Aging is usually characterized by impairments in physiological functionsincreasing its susceptibility to dementia and neurodegenerative disorders. Alzheimer's disease (AD) is a progressive, irreversible, neurodegenerative disease. The key to understanding AD is to elucidate the pathogenesis of neuron degeneration in specific brain regions.We hypothesize that there is increased DNA oxidation in AD brain compared to age-matched control subjects, especially in mitochondrial DNA (mtDNA), and that the changes in DNA bases will affect protein expression in mitochondria and contribute to neurodegeneration in AD. A promising approach investigated here is to utilize thin film multilayer structures where the thickness of a catalyst layer at an exposed edge of photolithographically defined pattern determines the diameter of the nanotubes/nanowires grown from it. This can in turn be incorporated into photolithographically defined post structures resulting in an array of suspended nanowires for line-of-site shadow lithography. NADPH-cytochrome P450 reductase (CPR) is a flavoprotein containing both FAD and FMN and functions as the electron donor protein for several oxygenase enzymes found on the endoplasmic reticulum of eukaryotic cells, including cytochrome P450s involved in drug metabolism and cholesterol biosynthesis. As many as three enzymes in the cholesterol biosynthetic pathway have been demonstrated, or proposed, to use CPR as a redox partner: squalene monooxygenase, which converts squalene to 2,3-oxidosqualene; lanosterol demethylase, a cytochrome P450 (CYP51); and 7-dehydrocholesterol reductase, the final step in cholesterol synthesis. Sarcocystis neurona is an apicomplexan parasite that is a major cause of equine protozoal myeloencephalitis (EPM). During intracellular development of S. neurona, many genes are temporally regulated. To better understand gene regulation, it is important to identify and characterize regulatory elements controlling gene expression in S. neurona. To perform this study, it was essential to establish transfection system for this parasite. Hence, the 5 flanking region of the SnSAG1 gene was isolated from a genomic library and used to construct expression plasmids. Salamanders of the genus Ambystoma are a unique model organism system because they enable natural history and biomedical research in the laboratory or field. We developed Sal-Site to integrate new and existing ambystomatid salamander research resources in support of this model system. Variable-rate technology (VRT) adds complexity to application equipment, thereby confounding the assessment of applicator performance. The intent of this investigation was to assess the rate response of various VRT granular applicators: two spinner spreaders (A and B), and two pneumatic applicators (C and D). Variable-rate (VR) tests were conducted to quantify the rate response characteristics (delay and transition times) for the applicators. A sigmoidal function was used to model the rate response for five of the six tests. Applicator A exhibited a linear response during decreasing rate changes.
