Soils disturbed by strip mining practices may have increased colloid loads moving to groundwater resources, also enhancing the transport of contaminants into our water resources. We hypothesize that contaminant transport within soils following mining is enhanced by colloid mobility. Two sites were chosen for this study, a 30-year old reclaimed strip mine in southwest Virginia and a recently mined area from eastern Kentucky. Intact reclaimed soil monoliths were retrieved from sandstone derived soils in southwestern Virginia. Reclaimed monoliths from eastern Kentucky were recreated in the lab. Microglia-mediated neuroinflammation plays an important role in the pathogenesis of Parkinson's disease (PD). Uncontrolled microglia activation produces major proinflammatory factors including cyclooxygenase 2 (COX-2) and inducible nitric oxide synthase (iNOS) that may cause dopaminergic neurodegeneration. Peroxisome proliferator-activated receptor γ (PPARγ) agonist pioglitazone has potent antiinflammatory property. We hypothesize pioglitazone protects dopaminergic neuron from lipopolysaccharide (LPS)-induced neurotoxicity by interacting with relevant signal pathways, inhibiting microglial activation and decreasing inflammatory mediators.

This study evaluated the efficacy of various attention-related, neuropsychological, and symptom validity measures in the detection of feigned ADHD in an undergraduate sample. Performance was compared between a group of presumed normal students (HON), a group of diagnostically "clean" ADHD students asked to respond to the best of their ability (ADHD), and a group of motivated, coached feigners (FGN). Feigners were educated about symptoms and characteristics of ADHD, provided with a scenario to help them relate to the plight of a student who might seek diagnosis, admonition to feign believably, and a significant monetary incentive for "successful feigning" ($45). Consumers evaluate products in the market place using their senses and often form mental representations of product properties. These mental representations have been studied extensively. Imagery has been shown to interact with perception within many perceptual modalities including vision, auditory, olfactory, and motor. This dissertation draws on the vast visual imagery literature to examine imagery in the haptic, or touch, modality. The current project examined the ability of a novel series of guandine and amidine-containing nicotine analogs to interact with several native and recombinantlyexpressed mammalian neuronal nicotinic-acetylcholine receptor (nAChR) subtypes. Rational drug design methods and parallel organic synthesis was used to generate a library of guanidine-containing nicotine (NIC) analogs (AH compounds). A smaller series of amidine-containing nicotine analogs (JC compounds) were also synthesized. In total, >150 compounds were examined. Compounds were first assayed for affinity in a high-throughput [3H]epibatidine radioligand-binding screen. Previous research has suggested that lobeline, a plant alkaloid derived from Lobelia inflate, has potential to be an efficacious pharmacotherapy for the treatment of methamphetamine dependence. In addition to attenuating methamphetamineinduced dopaminergic alterations in vitro, lobeline has been shown to decrease the primary rewarding effects and discriminative stimulus properties of methamphetamine in rats. It is of clinical interest to assess the utility of lobeline to decrease methamphetamine conditioned cues as these cues have been shown to significantly contribute to relapse.

A dithiol compound, benzene-1,3-diamidoethanethiol (BDETH2), also known as N,N'-bis(2-mercaptoethyl) isophthalamide or N,N'-bis(2-mercaptoethyl)-1,3-benzenedicarboxamide, capable of binding divalent metal ions, has been synthesized and characterized. Clostridium thermocellum is a thermophilic bacterium that converts biomass to ethanol directly; however, high sensitivity of this bacterium toward ethanol limits its commercial utility. To elucidate the effect of ethanol on the growth of this bacterium a metabolite analysis of C. thermocellum was performed. The hypothesis of the project was that exogenous ethanol alters the metabolite profile of C. thermocellum. For metabolite analysis, capillary electrophoresis-electrospray ionization-mass spectrometry method (CE-ESI-MS) was developed due to highly polar and charged nature of metabolites. It is well documented that both quantitative and qualitative changes in the murine hematopoietic stem cell (HSC) population occur with age. In mice, the effect of aging on stem cells is highly strain-specific, thus suggesting genetic regulation plays a role in HSC aging. In C57BL/6 (B6) mice, the HSC population steadily increases with age, whereas in DBA/2 (D2) mice, this population declines. Our lab has previously mapped a quantitative trait locus (QTL) to murine chromosome 2 that is associated with the variation in frequency of HSCs between aged B6 and D2 mice. Plant microRNAs play important roles in plant growth and development. Here we investigated the roles of miRNAs in the plant abiotic stress, development and viral infection. MicroRNA membrane array analysis using five different abiotic stress treatments resulted in the identification of 8 novel stress inducible miRNA-families. Functional studies on novel stress inducible miR168 revealed its functional relation with abiotic stress. Over expression of miR168 in Arabidopsis showed upregulation of four stress related miRNAs (miR163, miR167, miR398 and miR408). The immunoproteasome, known to play an important role in MHC class I antigen processing and presentation, have been linked to neurodegenerative diseases and hematological cancers. However, the pathophysiological functions of the immunoproteasome in these diseases are still not very well established. This can be attributed mainly to the lack of appropriate molecular probes that selectively target the immunoproteasome catalytic subunits. One of the principle factors that limits the areas to which seeded bermudagrass can be adapted is low temperature. Therefore, increasing the winter tolerance of seeded bermudagrass cultivars has been a goal of turfgrass breeders for many years. Design of an efficient breeding method for developing cultivars with increased tolerance to cool and freezing temperatures could be enhanced by having heritability estimates for cold tolerance traits. Additionally, the identification of correlated traits can be useful in improvement of cultivar development. Southern Magnolia (Magnolia grandiflora) is a primitive tree species that has attracted attention because of its horticultural distinctiveness, the wealth of natural products associated with it, and its evolutionary position as a basal angiosperm. Terpenoid constituents were determined from Magnolia leaves and flowers. Magnolia leaves constitutively produced two major terpenoids, andamp;acirc;-cubebene and germacrene A. However, upon wounding Magnolia leaves biosynthesized a significant array of monoand sesquiterpenoids, including andamp;acirc;-pinene, trans-andamp;acirc;-ocimene, andamp;aacute;-gurjunene, andamp;acirc;-caryophyllene and andamp;acirc;-cubebene, along with fatty acid derivatives such as cis-jasmone, for up to 19 hours after treatment. Anthracnose, caused by the fungal pathogen Colletotrichum trifolii, is one of the most destructive diseases of alfalfa worldwide. Cloning and characterization of the host resistance (R) genes against the pathogen will improve our knowledge of molecular mechanisms underlying host resistance and facilitate the development of resistant alfalfa cultivars. However, the intractable genetic system of cultivated alfalfa, owing to its tetrasomic inheritance and outcrossing nature, limits the ability to carry out genetic analysis in alfalfa. Group I intron-derived ribozymes are catalytic RNAs that have been engineered to catalyze a variety of different reactions, in addition to the native self-splicing reaction. One such ribozyme, derived from a group I intron of Pneumocystis carinii, can modify RNA transcripts through either the excision or insertion of RNA sequences. These reactions are mediated through the trans excision-splicing (TES) or trans insertionsplicing (TIS) reaction pathways. Atherosclerosis is a chronic inflammatory disease thought to be initiated by endothelial cell dysfunction. Research described in this dissertation is focused on the role of zinc deficiency in endothelial cell activation with an emphasis on the function of the transcription factors nuclear factor-κB (NF-κB), peroxisome proliferator activated receptor (PPAR), and the aryl hydrocarbon receptor (AhR), which all play critical roles in the early pathology of atherosclerosis. Microhistological analysis is a highly effective microscopic technique of determining botanical composition of animal diets by visual recognition of fecal plant fragments, and has been widely used in range studies. The objective of this study was to use microhistological techniques to predict the botanical composition of domesticated horse diets when grazing mixed cool-season pasture. Samples of tall fescue [Lolium arundinaceum (Schreb.) S.J. Darbyshire], Kentucky bluegrass (Poa pratensis L.) and orchardgrass (Dactylis glomerata L.) were evaluated for microscopically unique characteristics. The treatment of Mn and SO42- contaminated mine drainage via a sulfate reducing bioreactor is expected to result in near-permanent immobilization of significant amounts of Mn and a portion of the sulfates within the matrix. This study tested several different combinations of organic amendments and inorganic substrates in an attempt to optimize sulfate reducing conditions and Mn removal capacity. The speed and accuracy of conservation planning could be improved if maps indicating areas where grassed waterways should be placed to reduce erosion could be easily created. For five central Kentucky fields, elevation data were obtained with real time kinematic (RTK) global positioning system (GPS) and from US Geological Survey (USGS) digital elevation models (DEMs). Terrain attributes were calculated from these datasets which were used as predictor variables for neural network and logistic regression analyses. Standardized Ileal Digestible Tryptophan To Lysine Ratios In Growing Pigs Fed U.S. Type And Non U.S. Type Feedstuffs, Anthony David Quant

The objective of these studies was to determine the optimum standardized ileal digestible tryptophan to lysine (SID Trp:Lys) ratio for growing pigs fed both U.S.–type (corn–soybean meal) and non–U.S.–type (wheat–barley) diets.

When feeding U.S.–type diets, average daily gain (ADG) and plasma urea nitrogen (PUN) concentrations improved as SID Trp:Lys ratios increased, yielding optimum ratios of 15.32% (P<0.001) and 15.25% (P<0.001), respectively. Averaging these resulted in an optimum SID Trp:Lys ratio of 15.29%. In non–U.S.–type diets, feeding increasing SID Trp:Lys ratios improved ADG and PUN, displaying optima of 15.99% (P=0.048) and 15.29% (P=0.054), respectively. This produced an average optimum SID Trp:Lys ratio of 15.64%. The final study included the other dietary essential amino acids (EAA) at a higher level, and ADG and PUN improved as SID Trp:Lys …

Potato yellow dwarf virus (PYDV) is the type member of the genus Nucleorhabdovirus. The virus replicates in the nuclei of infected cells and mature virions accumulate in the perinuclear space after viral cores bud through the inner nuclear membrane. The virus was first described as an extremely destructive pathogen of potato (Solanum tuberosum) and other members of family Solanaceae. To date, much of the research examining the properties of this muscle group has been conducted on 2 primary muscles: the thyroarytenoid and posterior cricoarytenoid. Consequently, it is unknown whether the remaining intrinsic laryngeal muscles evidence this highly refined phenotype or if they retain a phenotype more similar to prototypical skeletal muscle. NM23-H1 represents the first identified metastasis suppressor, exhibiting reduced expression in breast carcinoma and melanoma, and an ability to inhibit metastatic growth without significant impact on the transformed phenotype. Although its molecular mechanism of action is not fully understood, NM23-H1 possesses at least three enzymatic activities that may mediate metastasis suppressor function. It catalyzes nucleoside diphosphate kinase (NDPK) activity, as well as protein histidine kinase and 3’-5’ exonuclease activities. As 3’-5’ exonucleases are generally required for maintenance of genomic integrity, this activity represents a plausible mediator to underlie the metastasis suppressor function of NM23-H1 protein. The JAK/STAT pathway is a non-receptor tyrosine kinase signaling pathway that is well conserved and highly re-utilized in many mammalian and Drosophila developmental processes. Compared to dozens of ligands and receptors in mammalian JAK/STAT, Drosophila JAK/STAT pathway is simpler with one receptor and three ligands, Upd, Upd2 and Upd3, which have similar amino acid sequences. Previous literature shows that upd and upd2 exhibit the same dynamic striped expression pattern in embryos and have semi-redundant functions during embryogenesis. Computational methods such as linear free energy relationships (LFERs) offer a useful high-throughput solution to quickly evaluate drug developability, e.g. membrane permeability, organic solvent/water partition coefficients, and solubility. LFERs typically assume the contribution of structural components/functional groups to the overall properties of a given molecule to be constant and independent. This dissertation describes a series of studies in which linear free energy relationships were developed to predict solvation of small organic molecules in lipid formulations, specifically, triglyceride containing solvents and phospholipid-based liposomes.
