Non-steroidal anti-inflammatory drugs (NSAIDs) are among the most widely used medications globally. There are generally two types: selective (COX-2) and traditional NSAIDs (COX-1). They are primarily used for the treatment of pain. They gained attention after a study about their basic mechanisms highlighted their toxicity.
Several studies have reported an association between NSAIDs and risk of myocardial infarction (MI). However, the direction of the relationship is not conclusive. Further studies are needed to ascertain the direction of this relationship and evaluate the present situation with available drugs. A challenge for fire researchers is obtaining data from those fires that are most dangerous and costly. While it is feasible to instrument test beds, test plots, and small prescribed burns for research, it is uncommon to successfully instrument an active wildland fire. With a focus on very specific facets of wildland fire, researchers have created many unique models utilizing matchsticks, cardboard, liquid fuel, excelsior, plywood, live fuels, dead fuels, and wood cribs of different packing densities. Humans learn to interact with many complex dynamic systems such as helicopters, bicycles, and automobiles. This dissertation develops a subsystem identification method to model the control strategies that human subjects use in experiments where they interact with dynamic systems. This work provides new results on the control strategies that humans learn.
We present a novel subsystem identification algorithm, which can identify unknown linear time-invariant feedback and feedforward subsystems interconnected with a known linear time-invariant subsystem. These subsystem identification algorithms are analyzed in the cases of noiseless and noisy data.

Integrins are cellular adhesion molecules that bind cells to the extracellular matrix. The integrin α6β4, a receptor for laminins, is predominantly expressed on epithelial cells where it is present at the basal surface adjacent to the basement membrane. This integrin plays a critical role in maintaining normal cellular functions, yet has also been implicated in promoting invasion and metastasis in human malignancies. While overexpression of the integrin α6β4 has been detected in select human cancers, the clinical significance of integrin α6β4 expression in a number of malignancies has not been determined. Metformin is an oral biguanide that is prescribed to over 120 million people worldwide for the treatment of conditions including type II diabetes mellitus, polycystic ovarian syndrome, and gestational diabetes. This hypoglycemic agent is rapidly emerging as a potential cost-effective anti-oncogenic agent. Over the past decade multiple epidemiologic studies have consistently associated metformin with decreased cancer incidence and cancer-related mortality. More recently numerous preclinical and clinical studies have demonstrated anti-cancer effects of metformin, leading to the proposal of numerous clinical trials to better understand this drug and its mechanism of action.

Abdominal aortic aneurysm (AAA) is a devastating disease that exhibits permanent lumen expansion typically in the infrarenal aorta. AAA is prevalent among aged population, especially in males. Despite the incidence in women is lower, studies indicate the tortuosity is more severe and aortic rupture risk is higher in women. In most patients, AAA remains asymptomatic until it ruptures leading to sudden and fatal hemorrhage.

Lung tumors secrete multiple factors that contribute to cachexia, a severe wasting syndrome that includes loss of muscle mass, weakness, and fatigue. 80% of advanced lung cancer patients experience cachexia, which cannot be reversed by nutritional interventions, diminishes response to and tolerance of cancer treatments, and increases morbidity and mortality. Despite a multitude of clinical trials, there are currently no approved treatments. This deficiency suggests that not all of the factors that contribute to cachexia have been identified.

Skeletal muscle is a major contributor to whole-body metabolism as it serves as a depot for both glucose and amino acids, and is a highly metabolically active tissue. An intrinsic molecular clock mechanism exists within skeletal muscle that regulates the timing of physiological processes. A key function of the clock is to regulate the timing of metabolic processes to anticipate time of day changes in environmental conditions. Skeletal muscle possesses remarkable plasticity in responses to altered mechanical load. An established murine model used to increase mechanical load on a muscle is the surgical removal of the gastrocnemius and soleus muscles, thereby placing a functional overload on the plantaris muscle. As a consequence, there is hypertrophic growth of the plantaris muscle. We used this model to study the molecular mechanisms regulating skeletal muscle hypertrophy.

Kentucky’s disappearing native grassland communities provide habitat for native flora and fauna. A study was conducted to compare the efficacy of herbicides in control of the invasive Japanese honeysuckle (Lonicera japonica) applied at times when most native species are dormant. Six herbicide mixtures (glyphosate, glyphosate + imazapyr, glyphosate + imazapic, imazapyr, triclopyr + difluphenzopyr, and metsulfuron + difluphenzopyr) were applied in three seasons to assess the effect of application timing of each mixture on honeysuckle control. Environmental and anthropogenic stressors negatively affect amphibians in a variety of ways, often increasing their vulnerability to pathogen infection and mortality. Sampling for the pathogenic fungus Batrachochytrium dendrobatidis (Bd) was conducted in order to: 1) determine the presence of chytrid infection in stream-associated plethodontid salamanders of southeastern Kentucky, and 2) evaluate differences in infection intensity between salamanders residing in intact forest streams, timber-harvested streams, and surface-mined streams. Intra-articular glucocorticoid injections are commonly used to treat synovitis and osteoarthritis in horses. These agents are highly effective at relieving pain, swelling, and other symptoms of joint inflammation. The drugs also have therapeutic benefits by down regulating the expression of cytokines and protease enzymes that participate in the degradation of articular cartilage. However, detrimental effects on chondrocyte function and cell viability that is independent of osteoarthritis pathogenesis have been described and linked to glucocorticoid use. American chestnut was once a crucial component of North American forests, but it was functionally eliminated by the introduction of the chestnut blight fungus. Chestnut is recently experiencing resurgence, but the introduced Asian chestnut gall wasp, a specialist herbivore, threatens chestnut recovery. I characterized this invasion and the interactions developing with host associates as the gall wasp spread in North America. Gall wasp dispersal is attributable to host plant distribution, effects of prevailing winds during adult insect emergence, and to topography. The brown marmorated stink bug (Halyomorpha halys) is an invasive, agriculturally destructive pest present in most of the United States. Because organic farmers have few effective control options to combat this pest, three potential control options were investigated. Screens of different mesh sizes (0.42cm, 0.32cm, and 0.10cm) that covered vegetable crops were evaluated for their ability to exclude H. halys, allow the entry of beneficial insects, and protect the crop from various types of damage. Taxonomic changes are made based on checkered beetle (Coleoptera: Cleridae) types of the Natural History Museum, London (BMNH).Lectotypes are designated (and holotypes and paralectotypes recognized) for 44 species of Hydnocerinae, including the type species for Isolemidia, Parmius, Paupris, Allelidea, Blaesiopthalmus and Lemidia, four species of Enoclerus (Clerinae), and 14 species of Cymatodera (Tillinae). Annotations include comments on additional type material, new type locality, previous (type series) locality, and questionable or missing types. The current mixed methods study utilized an ecological framework to examine public perspectives toward sexual behaviors among long-term care residents with dementia. Analyzing attitudes of the public is an integral component of understanding the entire ecological system that affects the development and overall well-being of a long-term care resident. Attitudes were examined using a multiple segment factorial vignette with a probability sample of 329 respondents from a southern state. Results indicate that attitudes were not statistically affected by sex or the elapsed time since diagnosis, and age, spousal disposition, and degree of intimacy predicted attitudes. Emerald ash borer, Agrilus planipennis Fairmaire, is an invasive wood boring beetle native to eastern Asia which was first detected in North America in 2002. All North American Fraxinus (ash) species are suitable hosts and susceptible to attack. Emerald ash borer larvae feed on phloem beneath the bark of infested trees resulting in girdling and mortality in as little as five years following initial infestation. Since its introduction near Detroit, Michigan, emerald ash borer has rapidly spread throughout much of the United States and portions of Canada, threatening the persistence of ash in invaded regions.

The current study investigated maternal determinants of mental health service use, namely, individual child therapy, among preadolescent children diagnosed with ADHD. The Behavioral Model of Health Care Utilization (Andersen, 2008) was used as a theoretical framework for the study. Data from the last three rounds of ECLS-K dataset were employed to test a longitudinal model using Bayesian analysis. Socio-demographic variables and maternal mental health were tested as exogenous variables and mother-child relationship variables, discipline variables, and perceived maternal concern about child’s overall behavior and child’s emotional symptoms were tested as intervening variables. Hippodamia convergens is a widely distributed insect predator in the United States and parts of Canada and Mexico. Several insectaries collect overwintering adults from aggregation sites in the Rocky Mountains during their winter dormancy. Collected beetles are then sold throughout the United States for augmentative biological control. This practice could have negative impacts on local populations of Hippodamia convergens in the Eastern United States. The function and developmental regulation of the altruistic worker and soldier caste is the central element contributing to major advantages of eusociality over solitary animals. The soldier caste is the first evolved sterile caste in termites. Their primary function is believed to be colony defense. However, the function and development of termite soldiers remains largely unknown. Concern that youth do not have enough fruit and vegetable intake lead to two strategies implemented to influence intake in the school environment: the Healthy Hunger-Free Kids Act (HHFKA) and Fighting with Food: Battling Chemical Toxicity with Good Nutrition program (FF), which could influence phytochemical content and knowledge regarding phytochemicals, respectively. Currently, the U.S. Preventive Services recommends biennial screening mammography for women ages 50-74. This study examined the associations between socioeconomic status and rurality on breast cancer screening behavior. Methods. Data for this study come from the Centers for Disease Control and Prevention (CDC) Behavioral Risk Factor Surveillance System Survey (BRFSS). The advent of RNA-seq technologies provides an unprecedented opportunity to precisely profile the mRNA transcriptome of a specific cell population. It helps reveal the characteristics of the cell under the particular condition such as a disease. It is now possible to discover mRNA transcripts not cataloged in existing database, in addition to assessing the identities and quantities of the known transcripts in a given sample or cell. However, the sequence reads obtained from an RNA-seq experiment is only a short fragment of the original transcript. A key determinant of geriatric frailty is sarcopenia, the age-associated loss of skeletal muscle mass and strength. Although the etiology of sarcopenia is unknown, the correlation during aging between the loss of activity of satellite cells, which are endogenous muscle stem cells, and impaired muscle regenerative capacity has led to the hypothesis that the loss of satellite cell activity is also a cause of sarcopenia. We tested this hypothesis in male sedentary mice by experimentally depleting satellite cells in young adult animals to a degree sufficient to impair regeneration throughout the rest of their lives. BACKGROUND: Dolichyl phosphate-linked mono- and oligosaccharides (DLO) are essential intermediates in protein N-glycosylation, C- and O-mannosylation and GPI anchor biosynthesis. While many membrane proteins in the endoplasmic reticulum (ER) involved in the assembly of DLOs are known, essential proteins believed to be required for the transbilayer movement (flip-flopping) and proteins potentially involved in the regulation of DLO synthesis remain to be identified.
METHODS: The synthesis of a series of Dol-P derivatives composed of citronellyl-based photoprobes with benzophenone groups equipped with alkyne moieties for Huisgen "click" chemistry is now described to utilize as tools for identifying ER proteins involved in regulating …

Biomass production is spatially distributed resulting in high transportation costs when moving dedicated biomass crops and crop residues. A multifaceted approach was taken to address this issue as the low bulk and energy density of biomass limits transportation efficiency. Two systems were analyzed for the conversion of biomass into a denser feedstock applicable to on-farm use. Pelletization was able to densify the material into a solid fuel. Using a pilot scale flat ring pellet mill, the density of the material was able to be increased to at least 4.4 times that of uncompressed material. Pellet durability was found to be …

Biochars have been created and characterized from a variety livestock manure biomass including poultry, dairy, and swine. However, no research has been conducted on the physical and chemical makeup of biochar pyrolyzed from horse muck. Two horse muck derived biochars containing either straw (HS) or woodchip (HW) bedding were pyrolyzed at 700°C and characterized for their physical and chemical properties. Tests revealed both biochars had high alkalinity, moderate specific conductivity, and low surface area as compared to other biochars in the literature. HS contained more mineral structures than HW.
