Exposure to environmental pollutants poses numerous risk factors for human health, including increasing incidence of cardiovascular disease and diabetes. Persistent organic pollutants, such as polychlorinated biphenyls (PCBs) have been strongly linked to the development of these chronic inflammatory diseases and the primary route of exposure is through consumption of contaminated food products. Thus, the gastrointestinal tract is susceptible to the greatest levels of these pollutants and is an important facet to study.

Abdominal aortic aneurysms (AAAs), a permanent dilation in the abdominal region of the aorta, is a highly sexually dimorphic disease. AAAs prevalence is ranging from 4-10 fold higher in males than females. Defining the mechanistic basis for reduced (in females) or increased (in males) AAA formation and progression may uncover potential therapeutic targets. The majority of studies examining sexual dimorphism focus on the role of sex hormones. However, genes residing on sex chromosomes, in addition to sex hormones, may contribute to sexual dimorphism of AAAs. Adult skeletal muscle is highly plastic and responds readily to environmental stimuli. One of the most commonly utilized methods to study skeletal muscle adaptations is immunofluorescence microscopy. By analyzing images of adult muscle cells, also known as myofibers, one can quantify changes in skeletal muscle structure and function (e.g. hypertrophy and fiber type). Skeletal muscle samples are typically cut in transverse or cross sections, and antibodies against sarcolemmal or basal lamina proteins are used to label the myofiber boundaries.

The purpose of this research was to evaluate and compare the performance, aesthetic and material properties of women’s t-shirts composed of three different sustainable fibers (lyocell, organic cotton, and viscose from bamboo) before and after repeated home laundering. A convenience sample was comprised of thirty-nine t-shirts. Thirteen black t-shirts from each fiber content were used for testing according to ASTM and AATCC standards. The evaluations and measurements were performed initially and after wash intervals one, five, ten, and twenty. The t-shirts were tested for fabric weight, fabric count, color fastness, smoothness appearance, pilling, dimensional stability and skewness.

Fungi in the genus Trichoderma have been characterized as biocontrol agents of plant pathogens since the 1930s. The use of biologicals for disease management has increased in recent years, typically marketed as a safer alternative to chemical applications. However, biologicals often lack consistent control across varying environmental conditions. To overcome the loss in efficacy due to environmental conditions, biologicals can be combined with common fungicide seed-treatments to provide improved control. The objectives of this study are to characterize two carrier proteins in the horse that significantly decrease in humans following anabolic androgenic steroid administration: sex hormone-binding globulin (SHBG) and thyroxine-binding globulin (TBG). For SHBG characterization, qPCR, RNA sequencing, and immunohistochemistry were performed on testes and equine livers. Free and total testosterone immunoassays were utilized to confirm the presence of a carrier protein in equine circulation. SHBG was detected in the testes using qPCR, RNA sequencing, and IHC, indicating the presence of the isoform androgen-binding protein (ABP). SHBG was not detected in any liver samples. Nearly extirpated from the Central Appalachians, USA by the mid-1900s as a result of human persecution, loss of forests, and absence of large mammal carrion, remnant populations of common ravens (Corvus corax) have recolonized portions of their historical range. One such area of recolonization is southeastern Kentucky where the species is listed as state threatened. Southeastern Kentucky appears to have extensive suitable breeding habitat, but raven records remain relatively rare with sightings and a few nests being confirmed during the past three decades. Changes in land use such as mountaintop removal mining with valley fills (MTR/VF) affect chemical, physical, and hydrological properties of headwater streams. Although numerous stream taxa have experienced significant declines from MTR/VF, stream salamanders appear to be particularly sensitive. Yet, the specific mechanism(s) responsible for the population declines has eluded researchers. We sampled salamander assemblages across a continuous specific conductivity (SC) gradient in southeastern Kentucky and estimated occupancy rates and abundance estimates along this gradient. Conventional Appalachian surface-mine reclamation techniques repress natural forest regeneration, and tree plantings are often necessary for reforestation. Reclaimed Appalachian surface mines harbor a suite of mammal herbivores that forage on recently planted seedlings. Anecdotal reports across Appalachia have implicated herbivory in the hindrance and failure of reforestation efforts, yet empirical evaluation of herbivory impacts on planted seedling vitality in this region remains relatively uninitiated. Pathogenic fungi are increasingly associated with epidemics in wildlife populations and represent a significant threat to global biodiversity. Snake fungal disease is an emerging disease caused by the fungus, Ophidiomyces ophiodiicola, and appears to be widespread in the eastern United States. Yet an evaluation of field diagnostics, and an understanding of the population-level consequences of the disease, are lacking. The martial eagle (Polemaetus bellicosus) is a vulnerable species that is declining throughout large portions of its range. There is an urgent need to improve understanding of this species’ ecology to inform its conservation. I equipped 20 adult martial eagles with global positioning system backpack transmitters to characterize diet and space use of the species in the Maasai Mara region of Kenya. The resulting high-resolution transmitter data sets allowed for the rapid location of kills and provided a means to estimate home range size. Many insects form close relationships with microbial symbionts. Insect symbionts can provide novel phenotypes to their hosts, including influencing dietary breadth. In the polyphagous cowpea aphid, Aphis craccivora, the facultative symbiont Arsenophonus improves aphid performance on one host plant (locust), but decreases performance on other plants. The goal of my thesis was to investigate the mechanism by which Arsenophonus facilitates use of locust. First, I assembled an Aphis craccivora-Arsenophonus-Buchnera reference transcriptome to conduct RNAseq analysis, comparing gene expression in aphids feeding on locust and fava, with and without Arsenophonus infection. Cucurbits (i.e. squash, melons, pumpkins, gourds) are high value crops of global importance. Insect pests in these systems are often controlled by chemical insecticides, which are not always effective and can be damaging to the environment. Many integrated pest management (IPM) techniques have been developed for the control of pests in these systems, with a goal of improving system stability and reducing chemical inputs. The primary goal of this paper is to demonstrate the application of a relatively esoteric and interdisciplinary technique, called spectral analysis, to dyadic social sciences data. Spectral analysis is an analytical and statistical technique, commonly used in engineering, that allows times series data to be analyzed for the presence of significant regular/periodic fluctuations/oscillations. Public awareness of declining pollinator populations has increased interest in creating “bee–friendly” urban landscapes. I quantified bee visitation and assemblages of 72 species of flowering woody plants common in urban landscapes. I found strong plant species effects and variation in seasonal activity of particular bee taxa but no overall differences in bee visitation or genus diversity between native versus nonnative species or trees versus shrubs. Analysis of pollen from a subset of these plants revealed small but statistically significant differences in total and essential amino acids between native and nonnative species and trees and shrubs, although each group had species …

Although the rates of delinquent behavior have been decreasing since the 1990s, adolescent delinquent behavior continues to take a great toll on society as well as on perpetrators themselves. In this way, it is essential to understand the process of delinquency development. The current dissertation is comprised of three studies that analyzed the predictors and the development of delinquency using genetically-informed designs. The sample used for all studies comes from the Add Health dataset, a nationally-representative data on adolescents followed across 14 years.

The University of Kentucky Superfund Research Center held the Appalachian Community Health and Well-being Forum at the Letcher County Cooperative Extension Office in Eastern Kentucky. A four-member panel consisted of two local health officials, a nutrition expert, and a federal scientist; answered questions from community members. Although links between sleep patterns and internalizing problems and externalizing or problem behaviors in children and adolescents have been established in literature, several gaps remain in this research. To explore relational processes of couple’s satisfaction this study drew on the relational spirituality framework (Mahoney, 2010) in order to test a relational religiosity model to evaluate the effects of public, private, ideological, intellectual, and experiential religiosity that were mediated by relational virtues of commitment, sacrifice, forgiveness, and sanctification and relational equality on couple’s satisfaction in two cultures. Data for this component used convenience samples of English-speaking respondents (hereafter American sample; n = 1,529) and Russian-speaking respondents (hereafter Russian sample; n = 529). Background

The rabies virus is a zoonotic pathogen that is nearly 100 percent fatal once symptoms occur. Because it is still active in wildlife populations in Kentucky, the need for preventive measures and surveillance is critical. Temperature and precipitation have been shown to affect rabies. This study examined rabies submissions in Kentucky, weather and geographical distribution to both describe and predict cases.

Methods

This review focuses on the role of Aβ in AD pathogenesis in Down syndrome and current approaches for imaging Aβ in vivo. We will describe how Aβ deposits with age, the posttranslational modifications that can occur, and detection in biofluids. Three unique case studies describing partial trisomy 21 cases without APP triplication, and the occurrences of low level mosaic trisomy 21 in an early onset AD patient are presented. Purpose—Increased vascularity is a hallmark of renal cell carcinoma (RCC). Microvessel density (MVD) is one measurement of tumor angiogenesis; however, its utility as a biomarker of outcome is unknown. ECOG-ACRIN 2805 (E2805) enrolled 1,943 resected high-risk RCC patients randomized to adjuvant sunitinib, sorafenib, or placebo. We aimed to determine the prognostic and predictive role of MVD in RCC.

Experimental Design—We obtained pretreatment primary RCC nephrectomy tissues from 822 patients on E2805 and constructed tissue microarrays. Hydrothermal liquefaction is a thermochemical technique for obtaining crude bio-oil from lignocellulosic biomass with moderate temperature and pressure. The crude bio-oil can then be upgraded to various biofuels and bioproducts. Hydrothermal liquefaction is amenable to use of biomass feedstocks that have high-moisture. The overall goal of this research is to demonstrate the effectiveness of white rot fungus (WRF) as a pretreatment option in the production of bio-oil from switchgrass through hydrothermal liquefaction. Ground-level and overhead images provide complementary viewpoints of the world. This thesis proposes methods which leverage dense overhead imagery, in addition to sparsely distributed ground-level imagery, to advance traditional computer vision problems, such as ground-level image localization and fine-grained urban mapping. Our work focuses on three primary research areas: learning a joint feature representation between ground-level and overhead imagery to enable direct comparison for the task of image geolocalization, incorporating unlabeled overhead images by inferring labels from nearby ground-level images to improve image-driven mapping, and fusing ground-level imagery with overhead imagery to enhance understanding. Over the past decade, the evolution of next-generation sequencing technology has considerably advanced the genomics research. As a consequence, fast and accurate computational methods are needed for analyzing the large data in different applications. The research presented in this dissertation focuses on three areas: RNA-seq read mapping, large-scale data query, and metagenomics sequence classification.
