Targeted photopolymerization is the basis for multiple diagnostic and cell encapsulation technologies. While eosin is used in conjunction with tertiary amines as a water-soluble photoinitiation system, eosin is not widely sold as a conjugate with antibodies and other targeting biomolecules. Here we evaluate the utility of fluorescein-labeled bioconjugates to photopolymerize targeted coatings on live cells. We show that although fluorescein conjugates absorb approximately 50% less light energy than eosin in matched photopolymerization experiments using a 530 nm LED lamp, appreciable polymer thicknesses can still be formed in cell compatible environments with fluorescein photosensitization. Smoothened (Smo), a GPCR family protein, plays a critical role in the reception and transduction of Hedgehog (Hh) signal. Smo is phosphorylated and activated on the cell surface; however, it is unknown whether Smo can be intracellularly activated. Here, we demonstrate that inactivation of the ESCRT-III causes dramatic accumulation of Smo in the ESCRT-III/MVB compartment, and subsequent activation of Hh signaling. In contrast, inactivation of ESCRTs 0–II induces mild Smo accumulation in the ESCRT-III/MVB compartment. We provide evidence that Kurtz (Krz), the Drosophila β-arrestin2, acts in parallel with the ESCRTs 0-II pathway to sort Smo to the multivesicular bodies and …

Evolution has been a paradox in the field of science, but, the study of evolutionary medicine applies both the evolutionary game theory and medicine. This study was conducted to explore evolution by analyzing two 19th century collections of medical formulations and prescriptions while compared to the trend of public health and pathogenic mechanisms. Analysis of organic structure in historical prescriptions, descriptive epidemiology in Kentucky, and the idea of the germ theory will be used explicitly to show the evolutionary change of health and disease. Previous studies have shown that adolescents raised in impoverished conditions are more likely to develop drug abuse in adulthood. In addition, both stress-inducing living conditions (impoverishment/isolation) and drugs of abuse may lead to an increase in the c-fos transcription factor in the reward circuit of the brain, particularly in the nucleus accumbens. The aim of the current study was to quantify the number of c-fos positive cells in the nucleus accumbens of enriched and isolated rats exposed to the opioid remifentanil. Volumetric muscle loss (VML) is a debilitating injury which results in full or partial loss of function. Current clinical options utilize tissue grafts and bracing to restore function. Tissue graft implantation oftentimes leads to serious complications, some of which end in graft rejection and thereby necessitate further surgeries and procedures. Polymeric scaffolds show promise as scaffolding systems due to their mechanical properties and overall degradation profiles. Scaffolds need appropriate mechanical properties, 10-60 kPa modulus, and overall degradation times, five days to two weeks, to initiate tissue regeneration. Background: East Liverpool, Ohio, the site of a hazardous waste incinerator and a manganese (Mn) processor, has had air Mn concentrations exceeding United States Environmental Protection Agency reference levels for over a decade. Save Our County, Inc., a community organization, was formed to address community environmental health concerns related to local industry. Researchers from the University of Cincinnati partnered with Save Our County to determine if air Mn had an impact on the neurocognitive function of children in the community.

A summary of 2,127 mouse genes linked to cilia by previous high-throughput approaches (reviewed in Inglis et al., 2006), published studies of individual gene products, or by McClintock et al. 2008. Each gene was viewed at Entrez Gene to verify and update the Gene ID and Gene Symbol. Each was also searched against PubMed (Gene Symbol AND (cili* OR flagel*)) for published evidence linking the mouse gene or putative orthologs to cilia or flagella. This study bridges the gap in literature about the impact of father absence on female adult attachment and current stress levels. A sample of 666 female college students between the ages of 18 and 22 at the University of Kentucky was recruited to complete an online survey about their experience with their fathers and the effects on their attachment and stress levels, while assessing the age of their first sexual experience. Father absence seems to be a significant predictor of earlier sexual activity among females and anxious attachment styles, along with higher stress levels. When the father is absent from …

Recurrent neural networks (RNNs) are state of the art sequential machine learning tools, but have difficulty learning sequences with long-range dependencies due to the exponential growth or decay of gradients backpropagated through the RNN. Some methods overcome this problem by modifying the standard RNN architecure to force the recurrent weight matrix W to remain orthogonal throughout training. The first half of this thesis presents a novel orthogonal RNN architecture that enforces orthogonality of W by parametrizing with a skew-symmetric matrix via the Cayley transform. Methylation at cytosine (5mC) is a fundamental epigenetic DNA modification recently associated with iAs-mediated carcinogenesis. In contrast, the role of 5-hydroxymethylcytosine (5hmC), the oxidation product of 5mC in iAs-mediated carcinogenesis is unknown. Here we assess the hydroxymethylome in iAs-transformed cells, showing that dynamic modulation of hydroxymethylated DNA is associated with specific transcriptional networks. Moreover, this pathologic iAs-mediated carcinogenesis is characterized by a shift toward a higher hydroxymethylation pattern genome-wide. At specific promoters, hydroxymethylation correlated with increased gene expression. Furthermore, this increase in hydroxymethylation occurs concurrently with an upregulation of ten-eleven translocation (TET) enzymes that oxidize 5-methylcytosine (5mC) in DNA. To …

A-102395 is a member of the capuramycin family of antibiotics which was isolated from the culture broth of Amycolatopsis sp. SANK 60206. A-102339 is structurally classified as a nucleoside antibiotic, which like all members of the capuramycin family, inhibits bacterial MraY (translocase I) with IC50 of 11 nM which is the lowest among the capuramycin family. A semisynthetic derivative of capuramycin is currently in clinical trials as an antituberculosis antibiotic, suggesting high potential for using A-102395 as a starting point for new antibiotic discovery. Stereotypes surrounding race and socioeconomic status often have overlapping attributes. That is, we tend to stereotypically associate African Americans and poor individuals with being incompetent. Further, people automatically associate African Americans with the concept of poor. The current research examined people’s mental representations of a homeless person, a poor person, and a person with a home, to see if people’s mental representation of a homeless varied from that of a poor person. Results from Study 1 (N = 524), using a bi-racial base image indicate that people, on average, mentally represent the poor and homeless in a similar manner. …

Students who are good self-regulators have higher motivation and achievement than those who are not. The beliefs students hold influence the goals they set, how they regulate learning, their motivation, and their subsequent actions. Beliefs about one’s own willpower (the capacity to exert self-control in everyday life) have been shown to affect individuals’ self-regulation. Willpower has been conceptualized as a limited resource that is easily depleted in demanding situations. However, some researchers have shown that individuals’ beliefs about willpower capacity (i.e., as finite or abundant), and not their actual willful acts, are more predictive of self-regulated behavior. Classroom-based physical activity is a newly explored avenue for providing physical activity opportunities to children within the school, but it is one that is showing academic gains in areas such as on-task behavior. Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by social and communication deficits, as well as restricted, repetitive, and stereotyped behaviors, interests, and activities (American Psychiatric Association, 2013). Although important relationships have been shown and described among the issues of challenging behavior, parent stress, and parenting sense of competence for families of children with ASD, there is a shortage of intervention programs appropriately suited for families which target these issues. The current study aimed to uncover processes and experiences that led individuals to critically engage in racial justice activism, specifically the Black Lives Matter movement. A constructivist grounded theory approach was utilized under critical-ideological and Black feminist paradigms in order to build a practical theory related to developing critical consciousness about oppression facing the Black community. Black activists in the movement between the ages of 23 and 60 (N=12) participated in intensive individual interviews. The reversible oxidation of cysteine by reactive oxygen species (ROS) is both a mechanism for cellular protein signaling as well as a cause of cellular injury and death through the generation of oxidative stress. The study of cysteine oxidation is complicated by the methodology currently available to isolate and enrich oxidized-cysteine containing proteins. We sought to simplify this process by reducing the time needed to process samples and reducing sample loss and contamination risk.

Uterine cancer is the nation’s most common gynecologic malignancy but is understudied in the geographically and socioeconomically diverse state of Kentucky (KY). This study assessed the frequency, distribution, and survival of uterine corpus malignancies in KY, and specifically the differences between Appalachia (AP) and non-Appalachia (NAP).
This study utilizes SEER and Kentucky Cancer Registries to study uterine corpus malignancy between January 1, 2000 and December 31, 2014. The analysis looks at incidence between diagnoses in AP and NAP. Systemic acquired resistance (SAR) is a type of plant defense mechanism that is induced after a localized infection and confers broad-spectrum immunity against related or unrelated pathogens. During SAR, a number of chemical signals and proteins generated at the site of primary infection travel to the uninfected tissues and are thought to alert the distal sites against secondary infections. Glycerol-3-phosphate (G3P) is one of the chemical signals that play an important role in SAR. The Coal Combustion Residual (CCR) and Effluent Limitations Guidelines (ELGs) rules pose significant technical and economic challenges for ash management and for treatment and discharge of flue gas desulfurization (FGD) wastewater at coal-fired power plants. Beneficial use is always a priority for ash, but only about half is being reused on a national basis, and the rest is destined for disposal. The U.S. Environmental Protection Agency identified biological treatment as the baseline treatment technology for FGD wastewater in the ELGs. Gene regulatory networks are composed of interconnected regulatory nodes created by regulatory factors of multiple types. All organisms finely tune gene expression in order to adapt to and survive within their current niche. Obligate parasitic bacteria are under extreme pressure to quickly and appropriately adapt their gene regulatory programs in order to survive within their given host. Borrelia burgdorferi is one such organism and persists in nature by alternating between two hosts; Ixodes spp. ticks and small vertebrate animals. Emerald ash borer (EAB), Agrilus planipennis (Coleoptera: Buprestidae), is an aggressive invader from Asia that has killed millions of trees in North America. Recently EAB has been documented developing in a novel host, white fringetree, Chionanthus virginicus. I evaluated larval performance in two common ash species and white fringetree by infesting excised bolts with emerald ash borer eggs. In addition I evaluated several plant characteristics to determine which most influence larval development. Changes to traditional taxonomic methods to incorporate new technologies and methods have already improved the quality of species hypotheses, but more work can be done to improve the speed of new species documentation. The mitochondrial COI DNA barcode has been successfully used to identify species with high accuracy since the early 2000s, and has been used in conjunction with morphological examinations and other DNA markers to discover and delimit new species. This thesis explores the application of DNA barcodes as the primary data for delimitation and diagnosis of new species of ichneumonoids.

Signaling scaffolds are critical for the correct spatial organization of enzymes within the ERK1/2 signaling pathway and proper transmission of intracellular information. However, mechanisms that control molecular dynamics within scaffolding complexes, as well as biological activities regulated by the specific assemblies, remain unclear.
The scaffold protein Shoc2 is critical for transmission of the ERK1/2 pathway signals. Shoc2 accelerates ERK1/2 signaling by integrating Ras and RAF-1 enzymes into a multi-protein complex. Germ-line mutations in shoc2 cause Noonan-like RASopathy, a disorder with a wide spectrum of developmental deficiencies.
