Genetic Variation For Fusarium Head Blight Resistance In Soft Red Winter Wheat, Marla Dale Hall Jan 2002

Genetic Variation For Fusarium Head Blight Resistance In Soft Red Winter Wheat, Marla Dale Hall

University of Kentucky Master's Theses

Fusarium graminearum, the causative agent of Fusarium head blight, is an economically important pathogen of wheat (Triticum aestivum). Breeding Fusarium head blight (FHB) resistant wheat requires knowledge of the underlying genetic control of FHB resistance.

Two nine-parent diallel analyses were completed in greenhouse and field environments. Combining abilities, variance component ratios, and narrow sense heritabilities for FHB resistance and deoxynivalenol levels were calculated. Significant general and specific combining ability effects were observed. Resistance to FHB seems to be mostly controlled by additive genetic effects with some dominance noted in the field. Resistance noted in the greenhouse environment …

Identification And Characterization Of Multiple Dna Loop Repair Pathways In Human Cells, Scott D. Mcculloch Jan 2002

Identification And Characterization Of Multiple Dna Loop Repair Pathways In Human Cells, Scott D. Mcculloch

University of Kentucky Doctoral Dissertations

The stability of DNA is a critical factor for several diseases, the most prevalent of which is cancer. Several neurodegenerative and accelerated aging diseases are also characterized by genomic instability. The number and complexity of DNA repair pathways that human cells possess underscores the importance of genomic stability. These pathways ensure that damaged DNA is repaired and that a cells complement of DNA remains stable upon cell division. How one particular type of DNA alteration, a DNA loop, is processed in human cells was the focus of this study. We have employed an in vitro system to study defined DNA …

Creation Of A Bacterial Mutagenicity Assay Highly Sensitive To Dialkylnitrosamines, Matthew Troy Cooper Jan 2002

Creation Of A Bacterial Mutagenicity Assay Highly Sensitive To Dialkylnitrosamines, Matthew Troy Cooper

University of Kentucky Doctoral Dissertations

Although dialkylnitrosamines are environmentally significant carcinogens, the use of short-term bioassays to assess the mutagenic potential of these compounds remains problematic. The Ames test, a mutagenicity assay based on the reversion of Salmonella typhimurium histidine auxotrophs, is the most widely used bioassay in genetic toxicology, but the traditional Ames tester strains are largely insensitive to dialkylnitrosamine mutagenicity. I have constructed several mutagenicity tester strains that co-express combinations of full-length human cytochrome P450 2E1, rat cytochrome P450 reductase, and human cytochrome b5 in S. typhimurium lacking ogt and ada methyltransferases (YG7104ER, ogt-; and YG7108ER, ogt-, ada-). These new strains are susceptible …

The Efficiacy Of Various Salmonella Intervention Methods Applied To Pork Carcasses During Slaughter, Nathan C. Clayton

University of Kentucky Master's Theses

Twenty-one market-age swine slaughtered in the University of Kentuckyabattoir were inoculated with fecal slurry containing two strains of nalidixic acidresistant Salmonella typhimurium on the ham, belly, and jowl regions on eachside of the carcass. Trial 1 revealed that a 10 s hot water spray was just aseffective as the 20 s spray in removing S. typhimurium, as there was nodistinguishable difference in population. The shorter flame singe (10 s) was aseffective as the 20 s application and the two chlorine solutions (100, 200 ppm)had similar results. The 2% lactic acid spray reduced S. typhimurium populationssignificantly more than the 1% treatment. …

Solvent-Resistant Nanofiltration Membranes: Separation Studies And Modeling, Dharmesh S. Bhanushali

University of Kentucky Doctoral Dissertations

The primary focus of the research is to extend the principles of Nanofiltration(NF) to non-aqueous systems using solvent-resistant NF membranes. Several differentlevels of interaction are introduced when organic solvents are used with polymericmembranes and thus quantification of polymer-solvent interactions is critical. Puresolvent permeation studies were conducted to understand the mechanism of solventtransport through polymeric membranes. Different membrane materials (hydrophilic andhydrophobic) as well as different solvents (polar and non-polar) were used for the study. For example, hexane flux at 13 bar through a hydrophobic silicone based NF membranewas ~ 0.6 x 10-4 cm3/cm2. s. and that through a hydrophilic aromatic polyamide based …

"That's Just The Way We Like It": The Children's Horror Film In The 1980'S, Christina Mitchell Bentley

University of Kentucky Master's Theses

The decade of the 1980s has often been considered a reactionary artistic wasteland in film studies, but it was nonetheless a period of volatile changes for the film industry. This period saw the decline of the mainstream horror film and the rise of the family film, two currents that reflect and illuminate the enormous changes in film production, marketing, and distribution. The hybrid genre of the childrens horror film, born in the 1980s, is particularly apt for discussing both the industry changes in this period and childrens relationship as viewers to the medium of film. The thesis defines the childrens …

A "Good Angry Man": Harry Caudill, The Formative Years, 1922-1960, Tylina Jo Mullins

University of Kentucky Master's Theses

After the publication of Night Comes to the Cumberland: A Biography of a Depressed Area, Harry Caudill became a spokesperson for Appalachia. Throughout the 1960s, Caudill continued to challenge the corrupt political system of the Cumberland Valley. His indictment of the coal industry as a leading factor in the continual depression of the area led scholars and reformers to a better understanding of the interrelated dynamics affecting the region. Even though with the passage of time, scholars have rejected many of Caudills ideas concerning the mountain people, few doubt that he led the challenge against the political, economic, and social …

The Effects Of Information Utilization On Corporate Decision-Making And Export Performance, Trond Breien Peersen Jan 2002

The Effects Of Information Utilization On Corporate Decision-Making And Export Performance, Trond Breien Peersen

University of Kentucky Master's Theses

This study investigates if companies that actively use export information have higher export growth rates or greater satisfaction with export performance measures than non-users. Organizational communication structures relating to information flow to decision makers is investigated to provide further insight into the role of export information. The study is based upon the knowledge utilization theory which states company/user characteristics are as important to information utilization as the characteristics of the specific piece of information. Bivariate analysis does not indicate a direct relationship between information use and reported higher export growth rates over the past four years. However, there are indications …

Chemistry Faculty Patents

Naked single-walled nanotube carbon metals and semiconductors were dissolved in organic solutions by derivatization with SOCl2 and octadecylamine charge. Both ionic (charge transfer) and covalent solution phase chemistry with concomitant modulation of the single-walled carbon nanotubes (SWNT) band structure were demonstrated. Solution phase near-IR spectroscopy was used to study the effects of chemical modifications on the band gaps of the SWNTs. Reaction of solubilized SWNTs with dichlorocarbene led to functionalization of the nanotube walls.

Resilient Modulus Of Kentucky Soils, Tommy C. Hopkins, Tony L. Beckham, Charlie Sun, Bixian Ni

Kentucky Transportation Center Research Report

In recent years, the American Association of State Highway Transportation Officials (AASHTO) has recommended the use of resilient modulus for characterizing highway materials for pavement design. This recommendation evolved as of a result of a trend in pavement design of using mechanistic models, which are based on the theory of elasticity (layered elastic analysis) or linear and non-linear, finite elements (and finite difference methods), or a combination of both of those theoretical approaches. Although much progress has been made in recent years in developing mathematical, mechanistic pavement design models, results obtained from those models are only as good as the …
