A Formative Evaluation Of Kentucky’S Environmental Compliance Assistance Program, Bonnie Wathen Jan 2008

A Formative Evaluation Of Kentucky’S Environmental Compliance Assistance Program, Bonnie Wathen

MPA/MPP/MPFM Capstone Projects

Kentucky’s Department for Environmental Protection’s Division of Compliance Assistance has the Environmental Compliance Assistance Program (CAP). CAP is a relatively new program that has had little to no evaluation and measurement. However, with limited state resources to be distributed to departments, it is imperative that programs be able to justify their existence. Due to this fact, I chose to evaluate the CAP for my capstone project.

Because inadequate data on outcomes is available at this time, I was unable to conduct a traditional summative evaluation or include any statistical analysis. Instead I chose to conduct a formative, or process, evaluation …

Computer Literacy, Access And Use Of Technology In The Family And Consumer Sciences Classroom, Dana Renee Jenkins

University of Kentucky Master's Theses

For years, schools across the nation have been joining the technology revolution. Today, students have at least some form of technology available to them in school (Roblyer, Castine, andamp; King, 1993; Croxall andamp; Cummings, 2000). This trend is not likely to change, so there is an increasing need for teachers who are literate in the use of the various types of technology. The purpose of this study was to determine if a relationship exists between computer literacy and use of technology, as well as if a relationship exists between teachers access to technology and their use of technology in Family …

Microbial Community Structure Dynamics In Ohio River Sediments During Reductive Dechlorination Of Pcbs, Andres Enrique Nunez Jan 2008

Microbial Community Structure Dynamics In Ohio River Sediments During Reductive Dechlorination Of Pcbs, Andres Enrique Nunez

University of Kentucky Doctoral Dissertations

The entire stretch of the Ohio River is under fish consumption advisories due to contamination with polychlorinated biphenyls (PCBs). In this study, natural attenuation and biostimulation of PCBs and microbial communities responsible for PCB transformations were investigated in Ohio River sediments.

Natural attenuation of PCBs was negligible in sediments, which was likely attributed to low temperature conditions during most of the year, as well as low amounts of available nitrogen, phosphorus, and organic carbon. Moreover, surface sediments were relatively oxidized, as indicated by the prevalence of aerobic bacteria such as beta- Proteobacteria, alpha-Proteobacteria, Sphingobacteria, and Nitrospira in 16S rRNA sediment …

For the very same reason SD cards are being widely used in space applications also. Using these SD Cards in space applications requires robust radiation hardened SD cards and Highly Reliable Fault Tolerant File Systems to handle them. The present …

The Role Of Stem Cell Antigen-1(Sca-1) In Muscle Aging, Sonia Angela Richards-Malcolm Jan 2008

The Role Of Stem Cell Antigen-1(Sca-1) In Muscle Aging, Sonia Angela Richards-Malcolm

University of Kentucky Master's Theses

Muscle aging is associated with a decrease in the number of satellite cells and their progeny, muscle progenitor cells (MPCs) that are available for muscle repair and regeneration. However, there is an increase in non-immuno-hematopoietic cells (CD45 negative) in regenerating muscle from aged mice characterized by high stem cell antigen -1(Sca-1) expression. In aged regenerating muscle, 14.2% of cells are CD45neg Sca-1pos while 7.2% of cells are CD45neg Sca-1pos in young adult muscle. In vitro, CD45neg Sca-1pos cells over express genes associated with fibrosis, potentially controlled by Wnt2. These cells are proliferative, non-myogenic and non-adipogenic, and arise in clonally-derived MPCs …

Being A Man In Kentucky: Perspectives Of Rural Migrant Workers, Mitchell Beam Snider Jan 2008

Being A Man In Kentucky: Perspectives Of Rural Migrant Workers, Mitchell Beam Snider

University of Kentucky Master's Theses

This thesis concerns identity constructions among rural migrant workers in Kentucky in relation to experiences and articulations of transnational spaces, networks, and identities. It was conducted through semi-structured interviews of migrant laborers on two rural Kentucky horse farms with 13 men. In this project, the men’s identities could be seen to have access to and utilize social, economic, cultural, and familial connections across national borders. These aspects of transnational identities were contrasted and compared to aspects of these men’s masculine identities to problematize popular representations of masculinities. This thesis shows how traditional notions of masculinities are questioned, reinforced, discarded, touted …

Effects Of Fescue Herbicides Plateau® And Cimarron® On Pregnancy Maintenance In Broodmares And On Alkaloid Concentrations In Endophyte Infected Tall Fescue, Kathleen Scarlett Black

University of Kentucky Master's Theses

Ingestion of endophyte infected (E+) fescue by pregnant mares can cause significant reproductive problems. Plateau® and Cimarron® herbicides suppress fescue while leaving desired forages unharmed. To determine if these herbicides are harmful to pregnant mares, they were allowed to graze pastures treated with Plateau®, Cimarron®, or vehicle carrier. Pregnancies were monitored via ultrasonography, blood chemistry, and hematology. Of the components measured only creatinine differed among treatments over time (P=0.0003) and that increase was only significant in one of four studies.

Two additional experiments were conducted to determine the effect of the herbicides on alkaloids within E+ fescue. A greenhouse experiment …

Endothelial Cell Dysfunction By Environmental Contaminants, Elizabeth Grace Oesterling Jan 2008

Endothelial Cell Dysfunction By Environmental Contaminants, Elizabeth Grace Oesterling

University of Kentucky Doctoral Dissertations

Within the last few decades, epidemiological evidence has linked exposure to air pollution, both its particles and its organic components, with cardiovascular disease (CVD) progression. CVD is a life long disease with the disruption of the endothelium being the inaugural event in this inflammatory process. The vascular endothelium is extremely susceptible to environmental insults given its tremendous surface area and that it is in constant contact with blood and components circulating within the blood, including xenobiotics. The endothelium is important as a barrier from blood constituents however, dysfunction of this barrier leads to the influx of lymphocytes and granulocytes that …

Metabolite Analysis Of Clostridium Thermocellum Using Capillary Electrophoresis Based Techniques, Anup P. Thakur Jan 2008

Metabolite Analysis Of Clostridium Thermocellum Using Capillary Electrophoresis Based Techniques, Anup P. Thakur

University of Kentucky Doctoral Dissertations

Clostridium thermocellum is a thermophilic bacterium that converts biomass to ethanol directly; however, high sensitivity of this bacterium toward ethanol limits its commercial utility. To elucidate the effect of ethanol on the growth of this bacterium a metabolite analysis of C. thermocellum was performed. The hypothesis of the project was that exogenous ethanol alters the metabolite profile of C. thermocellum. For metabolite analysis, capillary electrophoresis-electrospray ionization-mass spectrometry method (CE-ESI-MS) was developed due to highly polar and charged nature of metabolites. To increase the sensitivity of CE-ESI-MS, several parameters at the ESI interface were optimized. The application of 50% isopropanol …

Implications For The Hsf2/Prc1 Interaction And Regulation Of Condensin By Phosphorylation During Mitosis, Lynea Alene Murphy Jan 2008

Implications For The Hsf2/Prc1 Interaction And Regulation Of Condensin By Phosphorylation During Mitosis, Lynea Alene Murphy

University of Kentucky Doctoral Dissertations

At the beginning of mitosis, chromosomes are condensed and segregated to facilitate correct alignment later in cytokinesis. Condensin is the pentameric enzyme responsible for this DNA compaction and is composed of two structural maintenance of chromosomes (SMC) subunits and three non-SMC subunits. Condensin mutations generate chromosomal abnormalities due to improper segregation, leading to genome instability and eventual malignant transformation of the cell. Cdc2 phosphorylation of the non-SMC subunits, CAP-G, CAP-D2, and CAP-H, has been demonstrated to be important for condensin supercoiling activity and function. While these subunits are thought to be phosphorylated by Cdc2, the exact sites have not yet …

Toward The Construction Of "Kentucky Food" In The Twenty-First Century: Food Localism And Commodification Of Place Identity Under Post-Tobacco Agricultural Restructuring, 1990-2006., Taro Futamura

University of Kentucky Doctoral Dissertations

This study examines the concept of local food and the discourses surrounding the concept, both of which have played a significant role during Kentuckys agricultural restructuring. Since the mid-1990s, Kentucky farmers who were dependent on tobacco production began to struggle financially after the substantial reduction of quota allotments, and they were encouraged to diversify their agricultural production. Subsequently, practices of producing, marketing, and consuming locally-grown food were implemented. Drawing primarily on qualitative data, this study investigates the meanings of Kentuckys local food discourse development in four dimensions: 1) the political economy of tobacco production and the structural change of Kentuckys …

This dissertation examines rural families responses to global and local situations that have made earning a livelihood as a farmer very difficult. Drawing from original research, including a household livelihood census of 195 households, interviews with 72 people, participant observation, and archival research, the dissertation explores how rural families have responded to declines in domestic agricultural markets fueled by global and national forces and local environmental change. It asks: what impact will small farming families responses to these forces of change have on peoples identities as peasants? I argue that while great change is underway in the countryside, peasant identity …

The Inheritance Of Cold Tolerance In A Seeded Bermudagrass (Cynodon Dactylon L. Pers.) Population, Thomas Richard Stefaniak Jan 2008

The Inheritance Of Cold Tolerance In A Seeded Bermudagrass (Cynodon Dactylon L. Pers.) Population, Thomas Richard Stefaniak

University of Kentucky Doctoral Dissertations

One of the principle factors that limits the areas to which seeded bermudagrass can be adapted is low temperature. Therefore, increasing the winter tolerance of seeded bermudagrass cultivars has been a goal of turfgrass breeders for many years. Design of an efficient breeding method for developing cultivars with increased tolerance to cool and freezing temperatures could be enhanced by having heritability estimates for cold tolerance traits. Additionally, the identification of correlated traits can be useful in improvement of cultivar development. Heritability estimates for winter tolerance can be obtained from cold treatments imposed artificially or from observations made in the field. …

Innovative Techniques For Digitizing And Restoring Deteriorated Historical Documents, George V. Landon, Jr.

University of Kentucky Doctoral Dissertations

Recent large-scale document digitization initiatives have created new modes of access to modern library collections with the development of new hardware and software technologies. Most commonly, these digitization projects focus on accurately scanning bound texts, some reaching an efficiency of more than one million volumes per year. While vast digital collections are changing the way users access texts, current scanning paradigms can not handle many non-standard materials. Documentation forms such as manuscripts, scrolls, codices, deteriorated film, epigraphy, and rock art all hold a wealth of human knowledge in physical forms not accessible by standard book scanning technologies. This great omission …

Loline Alkaloid Biosynthesis Gene Expression In Epichloe Endophytes Of Grasses, Dong-Xiu Zhang

University of Kentucky Doctoral Dissertations

Loline alkaloids (LA) are secondary metabolites produced by Epichloandamp;euml; (anamorph, Neotyphodium) grass endophytes. They are toxic and deterrent to a broad range of herbivorous insects but not to livestock. This protective bioactivity has spurred considerable research into the LA biosynthetic pathway. LOL, the gene cluster containing nine genes, is required for LA biosynthesis. The regulation of LOL genes during LA production in culture and in symbio is of interest. In this study, coordinate regulation between LOL gene expression and LA production level was investigated in both MM culture and symbiota. Results showed that expression of LOL genes in N. uncinatum …

Targeting The Metal Chelator D-Penicillamine To Exploit The Elevated Copper And Oxidative Stress Associated With Cancer, Anshul Gupte Jan 2008

Targeting The Metal Chelator D-Penicillamine To Exploit The Elevated Copper And Oxidative Stress Associated With Cancer, Anshul Gupte

University of Kentucky Doctoral Dissertations

The significantly increased copper and oxidative stress levels are characteristic hallmarks of cancer cells. These differences provide a unique opportunity for selective targeting of cancer cells. D-penicillamine (D-pen) has been proposed to generate reactive oxygen species (ROS) in presence of copper. Therefore, these studies were aimed at investigating the potential application of a currently marketed copper chelator, D-pen, as a novel cytotoxic anti-cancer agent. D-pen was shown to produce ROS, specifically hydrogen peroxide (H2O2), in the presence of cupric sulfate through a copper catalyzed oxidation reaction. During this process D-pen was converted to D-pen disulfide. The experimental proof of the …

Zinc Deficiency And Mechanisms Of Endothelial Cell Dysfunction, Huiyun Shen Jan 2008

Zinc Deficiency And Mechanisms Of Endothelial Cell Dysfunction, Huiyun Shen

University of Kentucky Doctoral Dissertations

Atherosclerosis is a chronic inflammatory disease thought to be initiated by endothelial cell dysfunction. Research described in this dissertation is focused on the role of zinc deficiency in endothelial cell activation with an emphasis on the function of the transcription factors nuclear factor-κB (NF-κB), peroxisome proliferator activated receptor (PPAR), and the aryl hydrocarbon receptor (AhR), which all play critical roles in the early pathology of atherosclerosis. Cultured porcine aortic vascular endothelial cells were deprived of zinc by the zinc chelator TPEN and/or treated with the NF-κB inhibitor CAPE or the PPARγ agonist rosiglitazone, followed by measurements of PPARα expression, cellular …

The Metastasis Suppressor Nm23-H1 Is Required For Dna Repair, Mengmeng Yang Jan 2008

The Metastasis Suppressor Nm23-H1 Is Required For Dna Repair, Mengmeng Yang

University of Kentucky Doctoral Dissertations

NM23-H1 represents the first identified metastasis suppressor, exhibiting reduced expression in breast carcinoma and melanoma, and an ability to inhibit metastatic growth without significant impact on the transformed phenotype. Although its molecular mechanism of action is not fully understood, NM23-H1 possesses at least three enzymatic activities that may mediate metastasis suppressor function. It catalyzes nucleoside diphosphate kinase (NDPK) activity, as well as protein histidine kinase and 3’-5’ exonuclease activities. As 3’-5’ exonucleases are generally required for maintenance of genomic integrity, this activity represents a plausible mediator to underlie the metastasis suppressor function of NM23-H1 protein. To investigate the relevant activity …

Selective Regulation Of Cardiomyocyte Signaling By Rgl2, Leah M. Allen Jan 2008

Selective Regulation Of Cardiomyocyte Signaling By Rgl2, Leah M. Allen

University of Kentucky Doctoral Dissertations

A key cardiovascular signaling molecule involved in both physiologic and pathologic regulation of cardiomyocytes is the small molecular weight G-protein, Ras. Differential effects of Ras are mediated by multiple effector molecules, including the RalGEFs which activate Ral. Studies performed in cardiomyocytes have indicated a role for Ral in cardiac hypertrophic signaling and the RalGEF family member, Rgl2, was shown to specifically interact with Ras in the heart. Therefore, I hypothesized that Rgl2 was an important Ras effector that would regulate cardiomyocyte signaling.

To elucidate the potential importance of Rgl2 in regulating cardiomyocyte signaling, a gain-of-function approach was utilized in which …

Induction Of The Heat Shock Response To Protect Against Polyglutamine Diseases And The Role Of Protein Sumoylation In Laminopathies And Alzheimer's Disease, Yu-Qian Zhang Jan 2008

Induction Of The Heat Shock Response To Protect Against Polyglutamine Diseases And The Role Of Protein Sumoylation In Laminopathies And Alzheimer's Disease, Yu-Qian Zhang

University of Kentucky Doctoral Dissertations

Heat shock proteins function as molecular chaperones which help protein folding and prevent protein aggregation. My study shows that celastrol, a pharmacological compound capable of up-regulating the levels of heat shock proteins, inhibits cell death and protein aggregation caused by expanded polyglutamine containing protein, and the protective effects of celastrol are dependent on heat shock factor 1. These results suggest the potential of celastrol as a therapeutic agent in the treatment of polyglutamine diseases.

Sumoylation is a protein modification which plays diverse roles in regulating the target proteins. My study shows that lamin A is a target of protein sumoylation, …
