Serum amyloid A (SAA) is a family of proteins, the plasma levels of which may increase >1000-fold in acute inflammatory states. We investigated the role of SAA in sepsis using mice deficient in all three acute-phase SAA isoforms (SAA-TKO). SAA deficiency significantly increased mortality rates in the three experimental sepsis mouse models: cecal ligation and puncture (CLP), cecal slurry (CS) injection, and lipopolysaccharide (LPS) treatments. SAA-TKO mice had exacerbated lung pathology compared to wild-type (WT) mice after CLP. Platelets express several surface receptors that could interact with different viruses. To understand the mechanisms of HIV-1′s interaction with platelets, we chose the EcoHIV model. While EcoHIV is an established model for neuroAIDS, its effects on platelets are ill-defined. Our results indicate that EcoHIV behaves differently from HIV-1 and is cleared from circulation after 48 h post- infection. The EcoHIV course of infection resembles an HIV-1 infection under the effects of combined antiretroviral therapy (cART) since infected mice stayed immunocompetent and the virus was readily detected in the spleen. Background and aims: Genome and epigenome wide association studies identified variants in carnitine palmitoyltransferase 1a (CPT1a) that associate with lipid traits. The goal of this study was to determine the role of liver-specific CPT1a on hepatic lipid metabolism.

Approach and results: Male and female liver-specific knockout (LKO) and littermate controls were placed on a low-fat or high-fat diet (60% kcal fat) for 15 weeks. Mice were necropsied after a 16 h fast, and tissues were collected for lipidomics, matrix-assisted laser desorption ionization mass spectrometry imaging, kinome analysis, RNA-sequencing, and protein expression by immunoblotting. The mosquito family Culicidae is divided into 2 subfamilies named the Culicinae and Anophelinae. Nix, the dominant male-determining factor, has only been found in the culicines Aedes aegypti and Aedes albopictus, 2 important arboviral vectors that belong to the subgenus Stegomyia. Here we performed sex-specific whole-genome sequencing and RNAseq of divergent mosquito species and explored additional male-inclusive datasets to investigate the distribution of Nix. Except for the Culex genus, Nix homologs were found in all species surveyed from the Culicinae subfamily, including 12 additional species from 3 highly divergent tribes comprising 4 genera, suggesting Nix originated at least 133 to …

Cell proliferation requires metabolic reprogramming to accommodate biosynthesis of new cell components, and similar alterations occur in cancer cells. However, the mechanisms linking the cell cycle machinery to metabolism are not well defined. Cyclin D1, along with its main partner cyclindependent kinase 4 (Cdk4), is a pivotal cell cycle regulator and driver oncogene that is overexpressed in many cancers. Here, we examine hepatocyte proliferation to define novel effects of cyclin D1 on biosynthetic metabolism. Metabolomic studies reveal that cyclin D1 broadly promotes biosynthetic pathways including glycolysis, the pentose phosphate pathway, and the purine and pyrimidine nucleotide synthesis in hepatocytes. CD8þ T cells from HLA-A2.1-transgenic mice, but not wild-type mice, immunized with the amino- terminus region (aa 41e152) of dense granule protein 6 (GRA6Nt) of Toxoplasma gondii secreted large amounts of perforin and granzyme B in response to GRA6Nt through antigen presentation by HLA-A2.1 in vitro. When those CD8þ T cells were transferred into chronically infected HLA-A2.1-expressing NSG mice deficient in T cells, cerebral cyst burden of the recipients of HLA-A2.1-transgenic T cells, but not of WT T cells, became significantly less than that of control mice with no cell transfer. Background: Magnetic resonance imaging (MRI) is a non-invasive imaging modality which, in conjunction with biopsies, provide a qualitative assessment of tumor response to treatment. Intravenous injection of contrast agents such as fluorine (19F) nanoemulsions labels systemic macrophages, which can, then, be tracked in real time with MRI. This method can provide quantifiable insights into the behavior of tumor-associated macrophages (TAMs) in the tumor microenvironment and macrophage recruitment during therapy.

A major challenge to integrating public metabolic resources is the use of different nomenclatures by individual databases. This paper presents md_harmonize, an open-source Python package for harmonizing compounds and metabolic reactions across various metabolic databases. The md_harmonize package utilizes a neighborhood-specific graph coloring method for generating a unique identifier for each compound via atom identifiers based on a compound’s chemical structure. The resulting harmonized compounds and reactions can be used for various downstream analyses, including the construction of atom-resolved metabolic networks and models for metabolic flux analysis. Dysregulated eEF2K expression is implicated in the pathogenesis of many human cancers, including triple-negative breast cancer (TNBC), making it a plausible therapeutic target. However, specific eEF2K inhibitors with potent anti-cancer activity have not been available so far. Targeted protein degradation has emerged as a new strategy for drug discovery. Droughts and overgrazing play a crucial role in the degradation of semi-arid rangelands. This is evident in the loss of palatable long-lived grass species and bush encroachment. Early warning indicators are needed to mitigate long-term degradation and decline in essential forage provision. Functional diversity provides valuable information on ecosystem health. Rangelands cover over 75% of Kuwait’s total land area. Most of these rangelands are severely degraded because of overgrazing, poor anthropic utilization, and mismanagement. In view of the general lack of analysis of the ecological restoration effects of underground soil in the desert grassland ecological restoration projects in Inner Mongolia, this study selected the most representative open-pit quarries, open-pit iron mines and open-pit coal mines in the desert steppe. Using high-throughput sequencing technology, nematode community composition, diversity, and function were analyzed to determine the response of nematodes to different ecological restoration methods in damaged areas of coal mines. A global network of service laboratories exists to test livestock feed materials (typically grass hay and pellets) for ergovaline, ergot alkaloids and lolitrem B to ensure ‘safe feeds’ are being given to livestock. These compounds are mycotoxins produced by endophytic fungi that naturally reside in feed material. They have been purposely bred into grass species, as they enhance the plant’s survival from drought and insect predation. Unfortunately, ergovaline and other ergot alkaloids also cause vasoconstrictive effects and reproductive difficulties in livestock, resulting in a $1 million annual loss in production for the cattle industry in the USA alone. Sustainable intensification of livestock systems implies greater efficiency in resource utilization resulting in greater output of products and other ecosystem services per unit of resource input. Strategies to improve resource use efficiency include diversification of plant and ruminant species with complementary resource use. Forages that have root systems with contrasting architecture and exploring different soil layers with complementary use of resource acquisition (e.g., nutrients, water) could enhance primary productivity. Monitoring and managing the soil seed bank is fundamental to land management as it constitutes the future generations of invasive plant communities. Invasive plants have traits that result in high recruitment through increased seed generation, short seed dormancy and phenotypic plasticity. Furthermore, invasive plants with growth forms that inhibit the growth and recruitment of other species can lead to monocultures and associated reduction in above-ground biodiversity, potentially negatively impacting the soil seed bank diversity and ecosystem functions and services.
