Transportation agencies are increasingly more challenged by the effects of extreme weather and national hazards on transportation assets. Severe weather events in Kentucky have become more frequent and more costly. These extreme events damage transportation assets, increase maintenance costs and strain transportation budgets. Climate models predict changing conditions to occur over the next century that differ from historical data. In Kentucky, models predict a warmer and wetter environment. The goal of this research is to help KYTC plan for extreme weather and natural hazards events, as well as absorb, recover from, and adapt to the resulting damage. Researchers determined where …

This review considers fully three-dimensional biomaterial environments of varying complexity as these pertain to research on the placenta. The developments in placental cell sources are first considered, along with the corresponding maternal cells with which the trophoblast interact. We consider biomaterial sources, including hybrid and composite biomaterials. Properties and characterization of biomaterials are discussed in the context of material design for specific placental applications. The development of increasingly complicated three-dimensional structures includes examples of advanced fabrication methods such as microfluidic device fabri- cation and 3D bioprinting, as utilized in a placenta context. Introduction: Attention-deficit/hyperactivity disorder (ADHD) is a neurodevelopmental disorder characterized by attention problems, excessive activity, and impulsivity - occurring in approximately 13% of children 12– 17 years of age, and as many as 16% of older adolescents 18–22 years of age, with a greater prevalence in boys than girls. ADHD frequently co-occurs with specific learning disorder (LD), a neurodevelopmental disorder characterized by difficulties learning and using academic skills, such as reading, writing, and mathematics. Taken together, ADHD and/or LD are common among high school students and college students and can influence performance on computerized tests used in concussion management. However, normative …

Unsupervised learning, particularly clustering, plays a pivotal role in disease subtyping and patient stratification, especially with the abundance of large-scale multi-omics data. Deep learning models, such as variational autoencoders (VAEs), can enhance clustering algorithms by leveraging inter-individual heterogeneity. However, the impact of confounders—external factors unrelated to the condition, e.g. batch effect or age—on clustering is often overlooked, introducing bias and spurious biological conclusions. In this work, we introduce four novel VAE-based deconfounding frameworks tailored for clustering multi-omics data. These frameworks effectively mitigate confounding effects while preserving genuine biological patterns. The deconfounding strategies employed include (i) removal of latent features correlated …

Purpose: To examine whether time of year (relative to hibernation emergence) influences the retinal degenerative effects of intravitreal injection of adenosine triphosphate (ATP) in the 13-lined ground squirrel (13-LGS). Methods: Eighteen (9 male, 9 female) 13-LGS in three experimental cohorts (early season, mid-season, late season) (n = 6 each) underwent baseline imaging using scanning light ophthalmoscopy (SLO) and optical coherence tomography (OCT). Animals then received a 10-μL intravitreal injection of 0.723 M ATP, followed by OCT and SLO imaging at 3, 10, and 21 days. Adaptive optics SLO (AOSLO) was performed in animals without retinal damage after the 21-day follow-up. …

This article describes the design of a single instructional module on writing and critiquing the Specific Aims (SA) section of a grant proposal for a biomedical research study. It begins by discussing the goals and learning objectives for the module. Then, it discusses the typical structure of a SA section to orient readers to the core content. This method is rooted in using an established framework for scientific writing, which is presented in brief, along with examples of how it applies to writing the SA section. OBJECTIVE: We sought to determine whether clonidine, a non-opioid a―2-adrenergic agonist, would effectively treat neonatal opioid withdrawal syndrome (NOWS). METHODS: This was an intention-to-treat randomized clinical trial. Enrollment criteria included prenatal opioid exposure, age #7 days, gestational age $35 weeks, no other medical condition, and need for pharmacotherapy. Primary outcomes were length of treatment and neurobehavioral performance. RESULTS: A total of 1107 patients were screened for enrollment (645 ineligible, 91 parents or staff unavailable, 216 declined, 155 consented). Comparative genomic studies of social insects suggest that changes in gene regulation are associated with evolutionary transitions in social behavior, but the activity of predicted regulatory regions has not been tested empirically. We used self-transcribing active regulatory region sequencing, a high-throughput enhancer discovery tool, to identify and measure the activity of enhancers in the socially variable sweat bee, Lasioglossum albipes. We identified over 36,000 enhancers in the L. albipes genome from 3 social and 3 solitary populations. Many enhancers were identified in only a subset of L. albipes populations, revealing rapid divergence in regulatory regions within this species. Population-specific enhancers …

This study examined the influence of adverse childhood experiences (ACEs) on Black sexual minority women's mental health and substance use disorders in adulthood. Secondary data analysis was conducted using N = 149 Black sexual minority women's reports from the Generations Study. Study variables included psychological distress, chronic strains, stressful life events, ACEs, and substance use disorders, including alcohol use disorder and drug use disorder. Quantitative data analysis consisted of descriptives and bivariate correlations. Participants' total ACE scores were positively significantly correlated with increased alcohol misuse, chronic life strains, and stressful life events. Artificial superlattices composed of perovskite oxides serves as an essential platform for engineering coherent phonon transport by redefining the lattice periodicity, which strongly influences the lattice-coupled phase transitions in charge and spin degrees of freedom. However, previous methods of manipulating phonons have been limited to controlling the periodicity of superlattice, rather than utilizing complex mutual interactions that are prominent in transition metal oxides. Colorectal cancer (CRC) is highly prevalent, accounting for approximately one-tenth of cancer cases and deaths globally. It stands as the second most deadly and third most common cancer type. Although the gut microbiota has been implicated in CRC carcinogenesis for the last several decades, it remains one of the least understood risk factors for CRC development, as the gut microbiota is highly diverse and variable. Many studies have uncovered unique microbial signatures in CRC patients compared with healthy matched controls, with variations dependent on patient age, disease stage, and location. Objective: Evaluate hemoglobin concentration (HbC) in sows and their offspring over consecutive parities. Materials and methods: Twenty-three females were monitored for HbC during parities 1, 2, and 3 at 7 timepoints (30 [± 2], 60 [± 2], 90 [± 2], and 112 days of gestation, 2 and 16 [± 1] days of lactation, and 5 [± 1] days post weaning). Piglet HbC was measured within 18 hours after birth and at 16 (± 1) days of age. Pigs were classified as anemic (HbC < 10 g/dL) or nonanemic (HbC ≥ 10 g/dL) at each timepoint. Results: On gestation day 90, 71.1% of sows were anemic across parities. In parity 1, HbC was less on lactation day 16 than all gestational timepoints (P < .001). In parity 2, HbC on lactation days 2 and 16 was less than all gestational timepoints (P < .001). In parity 3, HbC on lactation days 2 and 16 was less than gestation days 30 and 60 (P = .015). Piglet anemia prevalence at 1 day of age was 55.8%, 36.3%, and 46.1% for parity 1, 2, and 3, respectively (X2 < .001). Piglet anemia prevalence at 16 days of age was 35.6%, 18.7%, and 15.9% for parity 1, 2, and 3, respectively (X2 < .001). Implications: Decreasing sow HbC over the reproductive cycle and lack of post-weaning recovery in parity 3 indicates iron declines with advancing parity and may impact long-term health. Piglet anemia prevalence declined with advancing parity, suggesting a need to reevaluate piglet iron supplementation in litters from younger females.

Background: Black race, inadequate health literacy, and poor perceived control are predictors of increased cardiovascular disease (CVD) risk. The purpose of this study was to explore the relationships among race, health literacy, perceived control, and CVD risk while controlling for known risk factors in incarcerated men. Methods: We included data from 349 incarcerated men to examine race and CVD risk (Framingham Risk Score) using a serial mediation model with health literacy and perceived control using 95% confidence intervals (CIs) from 5000 bootstrap samples. The recent destruction brought upon the Appalachian region by Hurricane Helene has emphasized the importance of the Journal of Appalachian Health. As an open-access peer-reviewed source of information, the Journal plays a critical role in not only facilitating public health research about Hurricane Helene, but also combating misinformation regarding the event. In the days following Hurricane Helene, misinformation about the federal government’s response, along AI generated images, have left many in the region confused and misled about what had happened. Impacts Of The Covid-19 Pandemic On Intimate Partner Violence And Child Maltreatment Services In Rural Northwest North Carolina, Elisabeth G. Galphin, Adam Hege, Amy Dellinger Page

The current study examined the impact of the COVID-19 pandemic on IPV and child maltreatment services in rural northwestern North Carolina. Qualitative interviews were conducted with eight professionals representing six service organizations across four counties. The findings highlighted challenges these agencies faced throughout the pandemic, new risks for the clients served, and positive outcomes. In addition, it has been a useful learning experience as public health and social service agencies learn to serve their communities more effectively moving forward. This is especially relevant for rural communities, as it has put public health preparedness at the forefront.
