Examination Of Hippocampal N-Methyl-D-Aspartate Receptors Following Chronic Intermittent Ethanol Exposure In Vitro, Anna R. Reynolds
Examination Of Hippocampal N-Methyl-D-Aspartate Receptors Following Chronic Intermittent Ethanol Exposure In Vitro, Anna R. Reynolds
Theses and Dissertations--Psychology
Chronic intermittent ethanol exposure (CIE) is associated with degeneration of hippocampal neurons. The present study used hippocampal cultures to examine the loss of NeuN immunoreactivity, a relaible marker or neuronal density, after 1, 2, or 3 cycles of 5 days EtOH exposure (50 mM), followed by a 24-hour period of EWD or continuous EtOH exposure. NeuN immunoreactivity was decreased by 13%, 19%, and 16% in the CA1, CA3, and dentate gyrus after 3 cycles of CIE respectively; thionine staining confirmed significant cellular losses within each hippocampal subregion. Two cycles of CIE in aged tissue cultures resulted in significant decreases in …

Psychiatry Faculty Publications
The treatment recommendations in obsessive-compulsive disorder (OCD) after lack of response to selective serotonin reuptake inhibitors (SSRIs) include augmentation with other drugs, particularly clomipramine, a more potent serotonin reuptake inhibitor (SRI), or antipsychotics. We present two cases of response to lamotrigine augmentation in treatment-refractory OCD; each received multiple SRI trials over a >10-year period. The first patient had eleven years of treatment with multiple combinations including clomipramine and SSRIs. She had a >50% decrease of Y-BOCS (from 29 to 14) by augmenting paroxetine (60 mg/day) with lamotrigine (100 mg/day). The second patient had 22 years of treatment with multiple combinations, …
The Cellular Nucleic Acid Binding Protein In Aging And Disease, Robin Webb
The Cellular Nucleic Acid Binding Protein In Aging And Disease, Robin Webb
Theses and Dissertations--Molecular and Cellular Biochemistry
The ZNF9 gene on chromosome 3 encodes the cellular nucleic acid binding protein (CNBP), a ubiquitously expressed, 177 amino acid (≈19.5kDa) protein that is highly conserved among vertebrates. The function of the protein is largely unknown, however an expansion in the first intron of the protein results in myotonic dystrophy type 2 (DM2), a multisystemic disease featuring cardiac arrhythmia, muscle wasting, cataracts, and a range of neuropathologies. Remarkably, we recently discovered that CNBP is involved in regulating the activity of β-secretase, the enzyme that produces the first cleavage event in the generation of the amyloid-β peptide (Aβ). The progressive fibrillization …
The Disordered Regulation Of Calcineurin: How Calmodulin-Induced Regulatory Domain Structural Changes Lead To The Activation Of Calcineurin, Victoria B. Dunlap
The Disordered Regulation Of Calcineurin: How Calmodulin-Induced Regulatory Domain Structural Changes Lead To The Activation Of Calcineurin, Victoria B. Dunlap
Theses and Dissertations--Molecular and Cellular Biochemistry
Calcineurin (CaN) is a highly regulated Ser/Thr protein phosphatase that plays critical roles in learning and memory, cardiac development and function, and immune system activation. Alterations in CaN regulation contribute to multiple disease states such as Down syndrome, cardiac hypertrophy, Alzheimer’s disease, and autoimmune disease. In addition, CaN is the target of the immunosuppressant drugs FK506 and cyclosporin A. Despite its importance, CaN regulation is not well understood on a molecular level. Full CaN activation requires binding of calcium-loaded calmodulin (CaM), however little is known about how CaM binding releases CaN’s autoinhibitory domain from the active site. Previous work has …

Factors Influencing The Establishment And Survival Of Native Hardwood Tree Seedlings Of The Kentucky Inner Bluegrass Blue Ash-Oak Savanna-Woodland, James D. Shaffer
Theses and Dissertations--Biology
Historically, the Kentucky Inner Bluegrass blue ash-oak savanna-woodland was the primary ecosystem of the Inner Bluegrass Region (IBR) of Kentucky. After European settlement, the majority (>99%) of Bluegrass savanna was converted to agricultural and urban land uses. Currently remnant savanna tree species are failing to recruit. Therefore, a long-term restoration ecology project researching competition and disturbance on seedling establishment, survival, and growth has been established at Griffith Woods (the largest remaining savanna in Kentucky) in Harrison Co., KY. Fourteen native hardwood tree species (a total of 6,168 seedlings) have been experimentally planted. Light, soil, surrounding vegetation, and herbivory, factors …

An Attitude Determination System With Mems Gyroscope Drift Compensation For Small Satellites, Maxwell Bezold
An Attitude Determination System With Mems Gyroscope Drift Compensation For Small Satellites, Maxwell Bezold
Theses and Dissertations--Electrical and Computer Engineering
This thesis presents the design of an attitude determination system for small satellites that automatically corrects for attitude drift. Existing attitude determination systems suffer from attitude drift due to the integration of noisy rate gyro sensors used to measure the change in attitude. This attitude drift leads to a gradual loss in attitude knowledge, as error between the estimated attitude and the actual attitude increases.
In this thesis a Kalman filter is used to complete sensor fusion which combines sensor observations with a projected attitude based on the dynamics of the satellite. The system proposed in this thesis also utilizes …

Evaluation Of Depression Screening Practices For College Women In A Primary Care University Health Clinic, Sarah E. Lester
DNP Projects
Mental disorders, including depression, are increasing in frequency and intensity in the college student population. College-aged women appear to be particularly vulnerable to depression. Primary care providers play an important role in addressing this issue, as they are the principal health care contacts for more than 50% of patients with mental illnesses. Guidelines from the 2009 United States Preventative Services Task Force recommend screening all adults (age 18+) for depression in primary care when depression care supports are in place. However, current screening rates for depression in the primary care setting from are estimated at only 1.6 to 3.3% (United …

Nitrate Reduction Coupled To Iron(Ii) And Manganese(Ii) Oxidation In An Agricultural Soil, Stephanie Pyzola
Nitrate Reduction Coupled To Iron(Ii) And Manganese(Ii) Oxidation In An Agricultural Soil, Stephanie Pyzola
Theses and Dissertations--Plant and Soil Sciences
New evidence shows iron(II) oxidation is strongly coupled to nitrate reduction under anaerobic conditions in freshwater sediments and agricultural soils. However, the contribution of iron(II) oxidation to nitrate reduction is unknown. Furthermore, oxidation of manganese(II) by nitrate has been largely overlooked. This study investigated nitrate-dependent iron(II) and manganese(II) oxidation in an agricultural soil (Sadler silt loam) using stirred-batch kinetic techniques with native soil organic carbon (SOC) as the electron donor and included addition of amendments (hydrogen gas and wheat residue). In the presence of native SOC, nitrate-dependent Fe(II) and Mn(II) oxidation occurred at early stages of the reaction while organic …
Calendula Officinalis Growth And Production Of Secondary Compounds In Greenhouse And Soil-Based Herbal Organic Production Systems, Victoria M. Anderson
Calendula Officinalis Growth And Production Of Secondary Compounds In Greenhouse And Soil-Based Herbal Organic Production Systems, Victoria M. Anderson
Theses and Dissertations--Plant and Soil Sciences
Calendula officinalis is a useful model crop because calendula flowers are used both ornamentally and medicinally. Organic production systems have many challenges; among these is the synchrony of nitrogen mineralization with the requirements of the crop. Organic greenhouse substrates have significantly different initial nitrogen levels which influence the performance of calendula, the highest initial N and plant performance was found in the organic compost peat substrate (OCP). The addition of supplemental nitrogen improved performance, but only OCP performed as well as the conventionally fertilized peat-based substrate. The nitrogen mineralization patterns in a soil-based greenhouse pot experiment showed that highly processed …
Ecophysiological Responses Of Tall Fescue Genotypes To Endophyte Infection And Climate Change, Marie Bourguignon
Ecophysiological Responses Of Tall Fescue Genotypes To Endophyte Infection And Climate Change, Marie Bourguignon
Theses and Dissertations--Plant and Soil Sciences
Tall fescue is a widely used forage grass in the eastern USA and can form a symbiosis with a fungal endophyte, which can be beneficial for the plant but can cause livestock health issues. Little is known regarding the symbiotic response to predicted climate change. To address this knowledge gap, I analyzed tall fescue variety trial data collected throughout the U.S., exploring relationships between climate variables and yield for two different fescue cultivars that were either endophyte-free or infected. This study showed no endophyte or cultivar effect on fescue yield, but identified temperature, precipitation and location as significant predictors of …
Abiotic Nitrate And Nitrite Reactivity With Iron Oxide Minerals, Prakash Dhakal
Abiotic Nitrate And Nitrite Reactivity With Iron Oxide Minerals, Prakash Dhakal
Theses and Dissertations--Plant and Soil Sciences
Under iron (Fe3+)-reducing conditions where aqueous Fe2+ and unreduced solid Fe3+-oxides commonly coexist, soil Fe2+ oxidation has been shown to be coupled with nitrate (NO3-) reduction. One possible secondary reaction is the involvement of NO3- and nitrite (NO2-) with Fe-oxide minerals found in many natural environments. Yet, spectroscopic measurements and kinetic data on reactivity of NO3- and NO2- with Fe-containing oxide minerals such as goethite (a-FeOOH), and magnetite (Fe3O4) are not found in the literature. The reactivity of …
Reduced Tillage And Living Mulches For Organic Vegetable Production, Alex G. Hessler
Reduced Tillage And Living Mulches For Organic Vegetable Production, Alex G. Hessler
Theses and Dissertations--Plant and Soil Sciences
Reduced tillage and living mulches are alternative management strategies that can improve soil quality by minimizing disturbance and building soil organic matter. Weed suppression by these two practices alone is often insufficient to avoid crop yield losses, but their performance in an integrated system is not well understood. This project investigated the production of bell pepper (Capsicum annuum L.) and dry bean (Phaseolus vulgaris L.) in conventional tillage and strip tillage with a living mulch of teff (Eragrostis tef Zucc.) and Korean lespedeza (Kummerowia stipulacea Maxim.). Yields of pepper and bean were …
Mass Selection With An Optical Sorter For Head Scab Resistance In Soft Red Winter Wheat, Hussein M. Khaeim
Mass Selection With An Optical Sorter For Head Scab Resistance In Soft Red Winter Wheat, Hussein M. Khaeim
Theses and Dissertations--Plant and Soil Sciences
Fusarium head blight (FHB) or head scab, caused by Fusarium graminearum Schwabe [telomorph: Gibberella zeae Schwein.(Petch)], is one of the most destructive diseases of wheat (Triticum aestivum L.) worldwide. Numerous strategies for scab resistance breeding are in use, including phenotypic selection for low severity and marker-assisted selection for resistance QTL. The most destructive consequences of scab are evidenced through a reduction in grain quality, and the presence of mycotoxins, the most common of which is deoxynivalenol (DON). Thus, there is great interest among breeders in selecting for resistance to both of these traits. To this end, a study was …
Control Of Erwinia Tracheiphila In Cucumis Melo, John R. Caudle
Control Of Erwinia Tracheiphila In Cucumis Melo, John R. Caudle
Theses and Dissertations--Plant and Soil Sciences
Currently there is no control of bacterial wilt disease, Erwinia tracheiphila, in susceptible cucurbit crops, once infection of the plant occurs. Conventional and organic production systems rely on insecticide applications to kill the vectors, striped and spotted cucumber beetles, Acalymma vittatum and Diabrotica undecimpunctata, respectively, prior to transmission of the pathogen which indirectly controls the disease to some extent. Physical barriers such as row covers are used to exclude the vectors from plants prior to flowering; however, pollination requirements expose plants to potential infection. Experimental field plots were developed to test various enhanced organic production systems in an …
Doxorubicin-Induced, Tnf-Α-Mediated Brain Oxidative Stress, Neurochemical Alterations, And Cognitive Decline: Insights Into Mechanisms Of Chemotherapy Induced Cognitive Impairment And Its Prevention, Jeriel T. Keeney
Theses and Dissertations--Chemistry
The works presented in this dissertation provide insights into the mechanisms of chemotherapy-induced cognitive impairment (CICI or “ChemoBrain”) and take steps toward outlining a preventive strategy. CICI is now widely recognized as a complication of cancer chemotherapy experienced by a large percentage of cancer survivors. Approximately fifty percent of existing FDA-approved anti-cancer drugs generate reactive oxygen species (ROS). Doxorubicin (Dox), a prototypical ROS-generating chemotherapeutic agent, produces the reactive superoxide radical anion (O2-•) in vivo. Dox treatment results in oxidation of plasma proteins, including ApoA-I, leading to TNF-α-mediated oxidative stress in plasma and brain. TNF-α elevation in brain …
Oxidative Damage To Dna In Alzheimer's Disease, Sony Soman
Oxidative Damage To Dna In Alzheimer's Disease, Sony Soman
Theses and Dissertations--Chemistry
Previous studies from our laboratory and others show a significant increase in levels of both nuclear and mitochondrial DNA and RNA oxidation in vulnerable brain regions in the progression of Alzheimer’s disease (AD). Although total DNA oxidation is increased in AD it remains unclear whether oxidative damage is widespread throughout the genome or is concentrated to specific genes. To test the hypothesis that specific genes are more highly oxidized in the progression of AD, we propose to quantify the percent oxidative damage in genes coding for proteins shown to be altered in the progression of AD using quantitative/real-time polymerase chain …
Understanding Multidrug Resistance In Gram-Negative Bacteria -- A Study Of A Drug Efflux Pump Acrb And A Periplasmic Chaperone Sura, Meng Zhong
Theses and Dissertations--Chemistry
Multiple drug resistance (MDR) has been a severe issue in treatment and recovery from infection.Gram-negative bacteria intrinsically exhibit higher drug tolerance than Gram-positive microbes. In this thesis, two proteins involved in Gram-negative bacterial MDR were studied, AcrB and SurA.
Resistance-nodulation-cell division pump AcrAB-TolC is the major MDR efflux system in Gram-negative bacteria and efficiently extrudes a broad range of substances from the cells. To study subtle conformational changes of AcrB in vivo, a reporter platform was designed. Cysteine pairs were introduced into different regions in the periplasmic domain of the protein, and the extents of disulfide bond formation were …
Structural And Functional Characterization Of Mulitdrug Resistance Transporter And Regulator, Linliang Yu
Structural And Functional Characterization Of Mulitdrug Resistance Transporter And Regulator, Linliang Yu
Theses and Dissertations--Chemistry
Drug resistant bacteria pathogen poses a severe threat to human health. Bacterial drug efflux pumps are transporter proteins involved in the export of antibiotics out of cells. Efflux by transporters is one of the major drug resistant mechanisms. Multidrug efflux pumps can transport multiple classes of antibiotics and are associated with bacteria multiple drug resistance (MDR). Overproduction of these pumps reduces susceptibility of bacteria to a variety of antibiotics. MDR regulators are cytoplasmic proteins that control the expression level of MDR transporters in response to the cellular concentration of antibiotics. This thesis research focuses on three main directions in the …

The Role Of Bridging Therapy In Hepatocellular Carcinoma, Roberto Galuppo, Angie Mccall, Roberto Gedaly
Surgery Faculty Publications
Hepatocellular carcinoma (HCC) is the most common primary malignancy of the liver accounting for 7% of all cancers worldwide. Most cases of HCC develop within an established background of chronic liver disease. For that reason, liver resection is only possible in selected patients. Liver transplantation has become the treatment of choice in patients with HCC, end-stage liver disease, and significant portal hypertension. Shortage of organ donors has resulted in overall increase of waiting list time with increased risk of dropout due to tumor progression. Neoadjuvant therapies have emerged as an alternative to control tumor growth in patients while waiting. The …
