Investigation Of Subsurface Thermal Radiation Phenomena In Fibrous Thermal Protection Systems, Colby L. Gore
Investigation Of Subsurface Thermal Radiation Phenomena In Fibrous Thermal Protection Systems, Colby L. Gore
Theses and Dissertations--Science, Technology, Engineering, and Mathematics (STEM) Education
Atmospheric re-entry induces intense thermal stress upon vehicles through a high-enthalpy boundary layer that encompasses the craft. To protect these vehicles, numerous insulative methods, known as thermal protection systems (TPS), are deployed to protect the craft during re-entry. The TPS materials insulate the craft from intense thermal transmission that would terminate a mission. In hypersonic environments, vast temperature gradients exist, enforcing the exponential effect of thermal radiation. However, TPS materials are commonly low-density fibrous materials that allow for the potential to be penetrated by thermal radiation. A series of arc-jet tests was completed to replicate enhanced radiative conditions in a …

Background: Solid tumors vary by the immunogenic potential of the tumor microenvironment (TME) and the likelihood of response to immunotherapy. The emerging literature has identified key immune cell populations that significantly impact immune activation or suppression within the TME. This study investigated candidate T-cell populations and their differential infiltration within different tumor types as estimated from mRNA co-expression levels of the corresponding cellular markers.
Methods: We analyzed the mRNA co-expression levels of cellular biomarkers that define stem-like tumor-infiltrating lymphocytes (TILs), tissue-resident memory T-cells (TRM), early dysfunctional T-cells, late dysfunctional T-cells, activated-potentially anti-tumor (APA) T-cells and Butyrophilin 3A (BTN3A) isoforms, utilizing …

Auditory hair cells form precise and sensitive staircase-like actin protrusions known as stereocilia. These specialized microvilli detect deflections induced by sound through the activation of mechano-electrical transduction (MET) channels located at their tips. At rest, a small MET channel current results in a constant calcium influx which regulates the morphology of the actin cytoskeleton in the shorter ‘transducing’ stereocilia. However, the molecular mechanisms involved in this novel type of activity-driven plasticity in the stereocilium cytoskeleton are currently unknown. Here, we tested the contribution of myosin XVA (MYO15A) isoforms, given their known roles in the regulation of stereocilia dimensions during hair …

Oleate hydratase (OhyA), a flavoenzyme that catalyzes the hydration of unsaturated fatty acids, has been identified in various Bacillales organisms, including those in the Listeria, Lysinibacillus, Paenibacillus, and Staphylococcus genera. In this study, we combine structural biology with molecular and phylogenetic analyses to investigate the evolutionary dynamics of the OhyA protein family within the Bacillales order. Our evolutionary analysis reveals two distinct OhyA clades (clade I and clade II) within Bacillales that, while sharing catalytic function, exhibit significant genomic and structural differences. Our findings suggest that these OhyA clades originated from independent evolutionary processes through convergent evolution rather than gene …

Winter Cold Temperate Rangelands: Improvement Strategies For Intensive Rangeland Ecosystems, B E. Allan, D Scott
IGC Proceedings (1977-2023)
Transfer of developed grassland technologies to rangelands is discussed in terms of environmental gradients and the options and constraints they impose. Winier feed limitation can be overcome through conserved feeds and various in situ systems involving special purpose pastures and feed banks. General animal nutrition can be improved through intensive development, low input systems, better control of range utilisation and, where appropriate, correction of mineral deficiencies. The emphasis in seeking quick and easy solutions to rangeland management problems is questioned, A more informed, environmentally sound and sustainable approach to rangeland fanning atlitudes and management is proposed. A stepwise process to …

Both topography and wildfire can exert significant influences on ecosystem processes and functions during boreal forest successions. However, their impacts on ecosystem multifunctionality (EMF) remain unclear. A mega-fire burned an area of 8700 hectares in the Great Xing’an Mountains in 2000, creating a wide range of fire severity levels across various topographic positions. This provided a unique opportunity to explore the impacts of mixed-severity fire disturbance in boreal forests. We evaluated the effect pathways of wildfire and topography on aboveground multifunctionality (AEMF), soil multifunctionality (SEMF), and overall multifunctionality (OEMF). We found that high-severity burning resulted in lower AEMF, SEMF, and …

A key challenge in the development of organic mixed ionic-electronic conducting materials (OMIEC) for high performance electrochemical transistors is their stable performance in ambient. When operating in aqueous electrolyte, potential reactions of the electrochemically injected electrons with air and water could hinder their persistence, leading to a reduction in charge transport. Here, the impact of deepening the LUMO energy level of a series of electron-transporting semiconducting polymers is evaluated, and subsequently rendering the most common oxidation processes of electron polarons thermodynamically unfavorable, on organic electrochemical transistors (OECTs) performance. Employing time resolved spectroelectrochemistry with three analogous polymers having varying electron affinities …

Improving Transportation Resiliency, Ben Blandford, Erin Lammers-Staats, Emma Gregory
Kentucky Transportation Center Research Report
Transportation agencies are increasingly more challenged by the effects of extreme weather and national hazards on transportation assets. Severe weather events in Kentucky have become more frequent and more costly. These extreme events damage transportation assets, increase maintenance costs and strain transportation budgets. Climate models predict changing conditions to occur over the next century that differ from historical data. In Kentucky, models predict a warmer and wetter environment. The goal of this research is to help KYTC plan for extreme weather and natural hazards events, as well as absorb, recover from, and adapt to the resulting damage. Researchers determined where …

Unsupervised learning, particularly clustering, plays a pivotal role in disease subtyping and patient stratification, especially with the abundance of large-scale multi-omics data. Deep learning models, such as variational autoencoders (VAEs), can enhance clustering algorithms by leveraging inter-individual heterogeneity. However, the impact of confounders—external factors unrelated to the condition, e.g. batch effect or age—on clustering is often overlooked, introducing bias and spurious biological conclusions. In this work, we introduce four novel VAE-based deconfounding frameworks tailored for clustering multi-omics data. These frameworks effectively mitigate confounding effects while preserving genuine biological patterns. The deconfounding strategies employed include (i) removal of latent features correlated …

Comparative genomic studies of social insects suggest that changes in gene regulation are associated with evolutionary transitions in social behavior, but the activity of predicted regulatory regions has not been tested empirically. We used self-transcribing active regulatory region sequencing, a high-throughput enhancer discovery tool, to identify and measure the activity of enhancers in the socially variable sweat bee, Lasioglossum albipes. We identified over 36,000 enhancers in the L. albipes genome from 3 social and 3 solitary populations. Many enhancers were identified in only a subset of L. albipes populations, revealing rapid divergence in regulatory regions within this species. Population-specific enhancers …

Dynamics And Interactions Of Carbon, Nitrogen, Phosphorus And Sulphur Cycling In Grazed Pasture, A K. Metherell, C V. Cole, W J. Parton
IGC Proceedings (1977-2023)
There are close links between the carbon (C), nitrogen (N), phosphorus {P) and sulphur (S) cycles. This is especially evident in legume-based pasture in which ahe application of P and S fertilisers are required to achieve saaisfactory levels of N2 fixation and pasture production The dynamics of the nutrient cycles are controlled largely by the rates of pasture production and consumption, and by the rate of decomposition of organic matter. The CENTURY soil organic matter model integrates climate, soil and management driving variables to predict C, N, P and S dynamics. Simulated effecls of P and S fertiliser on nutrient …

Variations in speleothem calcium isotope ratios (δ44Ca) are thought to be uniquely controlled by prior carbonate precipitation (PCP) above a drip site and, when calibrated with modern data, show promise as a semi‐quantitative proxy for paleorainfall. However, few monitoring studies have focused on δ44Ca in modern cave systems. We present a multi‐year comparative study of δ44Ca, carbon isotopes (δ13C), and trace elemental ratios from cave drip waters, modern calcite, and host rocks from two cave systems in California—White Moon Cave (WMC) and Lake Shasta Caverns (LSC).
