Lexington Humane Society: Analysis Of Volunteers, D'Arcy Robb Jan 2011

Lexington Humane Society: Analysis Of Volunteers, D'Arcy Robb

MPA/MPP/MPFM Capstone Projects

The Lexington Humane Society depends substantially on volunteer labor yet suffers from a high volunteer attrition rate. Using data from the organization’s database and a survey of volunteers, this project paints a demographic picture of the volunteer population and identifies traits that affect volunteer longevity.

The evidence shows that the organization’s volunteers are overwhelmingly likely to be pet-owning women with no children at home who are motivated to volunteer because it allows them to act on their values. Middle-age individuals are more likely than those under 25 or over 65 to become long term volunteers, and individuals who have recently …

Elucidating The Biochemical Wizardry Of Triterpene Metabolism In Botroycoccus Braunii, Thomas Daniel Niehaus

Theses and Dissertations--Plant and Soil Sciences

B. braunii is a green alga that has attracted attention as a potential renewable fuel source due to its high oil content and the archeological record of its unique contribution to oil and coal shales. Three extant chemotypes of B. braunii have been described, namely race A, race B, and race L, which accumulate alkadienes and alkatrienes, botryococcene and squalene and their methylated derivatives, and lycopadiene, respectively. The methylated triterpenes, particularly botryococcenes, produced by race B can be efficiently converted to high quality combustible fuels and other petrochemicals; however, botryococcene biosynthesis has remained enigmatic.

It has been suggested that botryococcene …

Rit Gtpase Signaling Mediates Oxidative Stress Resistance And Survival Of Adult Newborn Neurons After Traumatic Brain Injury, Weikang Cai Jan 2011

Rit Gtpase Signaling Mediates Oxidative Stress Resistance And Survival Of Adult Newborn Neurons After Traumatic Brain Injury, Weikang Cai

Theses and Dissertations--Molecular and Cellular Biochemistry

The small GTPases function as molecular switches to control diverse signaling cascades. The mammalian Rit and Rin, along with Drosophila Ric, comprise an evolutionarily conserved subfamily of the Ras-related GTPases. Previous studies using cultured cell models suggested that Rit was involved in the control of cell proliferation, transformation, neuronal differentiation, morphogenesis, and cell survival, but the principal physiological function of Rit remained uncharacterized.

To address this outstanding question, we employed a genetic approach, engineering a Rit knockout mouse. Using this animal model, we demonstrate a central role of Rit in governing cell survival in a p38-dependent fashion. Primary mouse embryonic …

Trichloroethylene Exposure And Traumatic Brain Injury Interact And Produce Dual Injury Based Pathology And Pioglitazone Can Attenuate Deficits Following Traumatic Brain Injury, Andrew David Sauerbeck Jan 2011

Trichloroethylene Exposure And Traumatic Brain Injury Interact And Produce Dual Injury Based Pathology And Pioglitazone Can Attenuate Deficits Following Traumatic Brain Injury, Andrew David Sauerbeck

University of Kentucky Doctoral Dissertations

The development of Parkinson's disease (PD) in humans has been linked to genetic and environmental factors for many years. However, finding common single insults which can produce pathology in humans has proved difficult. Exposure to trichloroethylene (TCE) or traumatic brain injury (TBI) has been shown to be linked to PD and it has also been proposed that multiple insults may be needed for disease development.

The present studies show that exposure to TCE prior to a TBI can result in pathology similar to early PD and that the interaction of both insults is required for impairment in behavioral function, and …

Methionine-35 Of Aβ(1-42): Importance For Oxidative Stress In Alzheimer Disease, D. Allan Butterfield, Rukhsana Sultana

Chemistry Faculty Publications

Alzheimer disease (AD) is an age-related progressive neurodegenerative disorder. This devastating disease is characterized by the presence of senile plaques (SP), neurofibrillary tangles (NFTs), and loss of synapses. Amyloid beta-peptide 1–42 (Aβ(1–42)) is the main component of SP and is pivotal to AD pathogenesis. Brain of subjects with AD and arguably its earliest manifestation, mild cognitive impairment (MCI), demonstrate increased levels of oxidative stress markers. Our laboratory combined these two aspects of AD and MCI and proposed the Aβ(1–42)-associated free radical oxidative stress hypothesis to explain oxidative stress under which the MCI and AD brain exist …

Synthesis And Characterization Of Polymeric Antioxidant Delivery Systems, Paritosh P. Wattamwar

Theses and Dissertations--Chemical and Materials Engineering

Even though the role of oxidative stress in a variety of disease states is known, strategies to alleviate this oxidative stress by antioxidants have not been able to achieve clinical success. Particularly, treatment of oxidative stress by small molecule antioxidants has not received due attention because of the challenges associated with its delivery. Antioxidant polymers, where small molecule antioxidants are incorporated into the polymer backbone, are an emerging class of materials that can address some of these challenges.

In this work, biodegradable polymers incorporating phenolic antioxidants in the polymer backbone were synthesized. Antioxidant polymers were then characterized for their in …

Opioid Codrugs For Pain Management, Ujjwal Chakraborty Jan 2011

Opioid Codrugs For Pain Management, Ujjwal Chakraborty

Theses and Dissertations--Chemistry

Pain is an unpleasant sensory and emotional experience associated with actual or potential tissus damage or described in terms of such damage. Opioids are effective in treating moderate to severe pain, but opioid alone therapy is associated with several adverse effects, development of tolerance and addiction potential. One way to solve these problems is to administer opioids with adjuvant drugs. In this project several opioid molecules were combined with other adjuvant drugs in a single chemical entity to form a codrug.

A series of codrugs were prepared by conjugation of an opioid with S-(-)-nornicotine, ketamine, norketamine and gabapentin. Several …

Role Of Angiotensin Converting Enzyme 2 (Ace2) In Obesity-Associated Hypertension, Manisha Gupte

University of Kentucky Doctoral Dissertations

The purpose of this research was to determine whether adipocytes express ACE2 and its role in obesity-associated hypertension with diet-induced obesity.

To determine if ACE2 was expressed in adipose tissue and its regulation in the setting of diet-induced obesity, we fed male mice either a low fat (LF) or high fat (HF) diet acutely (1 week) or chronically ( 4 months). We demonstrated that ACE2 was regulated specifically in adipose tissue with consumption of a HF diet. However, with chronic HF feeding adipose ACE2 was dysregulated resulting in activation of the systemic RAS and increased blood pressure.

To determine the …

Disjuncture Among Classic Period Cultural Landscapes In The Tuxtla Mountains, Southern Veracruz, Mexico, Wesley Durrell Stoner Jan 2011

Disjuncture Among Classic Period Cultural Landscapes In The Tuxtla Mountains, Southern Veracruz, Mexico, Wesley Durrell Stoner

University of Kentucky Doctoral Dissertations

Teotihuacan was the most influential city in the Classic Mesoamerican worldsystem. Like other influential cities in the ancient world, however, Teotihuacan did not homogenously affect the various cultural landscapes that thrived in Mesoamerica during the Classic period (300-900 CE). Even where strong central Mexican influences appear outside the Basin of Mexico, the nature, extent, and strength of these influences are discontinuous over time and space. Every place within the Classic Mesoamerican landscape has a unique Teotihuacan story. In the Tuxtla Mountains of southern Veracruz, Mexico, Matacapan, located in the Catemaco Valley, drew heavily upon ideas and symbols fostered at Teotihuacan, …

Beef Cattle Grazing Preference Of Tall Fescue As Affected By Endophyte, Herbert Troye Owens Iii

University of Kentucky Master's Theses

Many factors control, contribute to, stimulate and limit forage selection. It is apparent that cattle prefer certain cultivars compared to others. This study sought to test if cattle displayed preferences for certain cultivars over a two-year period in 2008 and 2009. Determining the effect of endophyte status on preference was another objective. Previous research showed rapid increase in the selection of preferred cultivars, i.e., diet learning. We attempted to replicate those results. Forty cultivars (34 tall fescue (Lolium arundinaceum (Schreb.) Darbysh.) cultivars), two festuloliums (Festuca pratensis x Lolium perenne), two meadow fescue (Lolium pratense) and one meadow …

Going On Otor: Disaster, Mobility, And The Political Ecology Of Vulnerability In Uguumur, Mongolia, Daniel J. Murphy Jan 2011

Going On Otor: Disaster, Mobility, And The Political Ecology Of Vulnerability In Uguumur, Mongolia, Daniel J. Murphy

University of Kentucky Doctoral Dissertations

Post-socialist states have increasingly adopted rural governance and resource management policies framed around the concepts of decentralization, devolution, and de-concentration in which formerly central state powers are transferred to lower, more local levels of governance. In more recent incarnations, these policies have become inspired by neo-liberal discourses of minimal government, self-rule, and personal responsibility. Increasingly, the social science literature has argued that such forms of neo-liberal governance lead to a variety of unforeseen and diverse consequences. This dissertation attempts to understand the impact of these political transformations on household vulnerability in the context of hazardous events called zud. I …

Pollution Knowledge And Urban Water Politics In The Ganges River Basin (India), Sya Buryn Kedzior Jan 2011

Pollution Knowledge And Urban Water Politics In The Ganges River Basin (India), Sya Buryn Kedzior

University of Kentucky Doctoral Dissertations

Millions of people rely upon the Ganges River as a source of water provision and a site of disposal for sewage, solid waste, agricultural runoff and industrial effluent. The river is also a goddess in the Hindu pantheon who is worshipped for her purificatory powers, despite water quality levels that fall far short of standards for use in bathing, washing, and drinking. In recent years, a number of non-governmental organizations (NGOs) have formed to oppose both pollution of the river and the failure of state-run pollution abatement programs. They are joined by an increasingly frequent number of seemingly spontaneous protests …

Circadian And Homeostatic Regulation Of Sleep In Cast/Eij And C57bl/6j Mice, Peng Jiang

University of Kentucky Doctoral Dissertations

Sleep is essential for mammals and possibly for all animals. Advancing our knowledge of sleep regulation is crucial for the development of interventions in sleep-related health and social problems. With this aim, this study utilizes laboratory mice to explore sleep regulatory mechanisms at behavioral, molecular, and genetic levels.

Sleep is regulated by the interaction of circadian and homeostatic processes. The circadian clock facilitates sleep to occur at a favorable time of the day. Normal mice, such as the C57BL/6J (B6) strain, sleep mostly during the day and initiate activities at dark onset. Here, I show mice of the CAST/EiJ (CAST) …

Regulation Of Abcg5 And Abcg8 Sterol Transporters In Biliary Cholesterol Elimination, Reverse Cholesterol Transport And Dyslipidemia, Nadezhda Steliyanova Sabeva Jan 2011

Regulation Of Abcg5 And Abcg8 Sterol Transporters In Biliary Cholesterol Elimination, Reverse Cholesterol Transport And Dyslipidemia, Nadezhda Steliyanova Sabeva

University of Kentucky Doctoral Dissertations

ATP-binding cassette transporters ABCA1 and ABCG1 initiate reverse cholesterol transport generating HDL particles, whereas ABCG5/G8 promote biliary cholesterol secretion thereby facilitating the last step of reverse cholesterol transport. Mutations in the leptin axis result in obesity and dyslipidemia in ob/ob and db/db mice. These mice have defective HDL clearance, increased plasma cholesterol and decreased biliary cholesterol elimination. My studies demonstrate that ABCG5/G8 protein is low in these animals and can be restored with caloric restriction or leptin replacement. To directly test whether ABCG5/G8 alone is able to correct reverse cholesterol transport defect, liver specific ABCG5/G8 expression was achieved in db/db …
