Convolutional Neural Networks (CNNs) have been at the frontier of the revolution within the field of computer vision. Since the advent of AlexNet in 2012, neural networks with CNN architectures have surpassed human-level capabilities for many cognitive tasks. As the neural networks are integrated in many safety critical applications such as autonomous vehicles, it is critical that they are robust and resilient to errors. This thesis focuses on two important computational problems in genomics and metagenomics with the public available next-generation sequencing data. One is about gene regulation, for which we explore how distal regulatory elements may interact with the proximal regulatory elements. The other is about metagenomics, in which we study how to reconstruct bacterial strain genomes from shotgun reads. Studying gene regulation, especially distal gene regulation, is important because regulatory elements, including those in distal regulatory regions, orchestrate when, where and how much a gene is activated under every experimental condition. Muscular dystrophy is a genetically linked myopathy with no cure available. The lack of a cure makes early detection and treatment of muscular dystrophy imperative. When reviewing protocols examining muscular fatigue at submaximal isometric contractions, proper warm-up appeared to be absent and could have caused skewed results and conclusions. This study examines the effects of implementing a warm-up protocol before fatiguing trials. In this study, 10 adult subjects conducted fatiguing protocols with the right rectus at submaximal isometric contractions. The warm-up period included a light walk along with contractions at 20% and 33% of maximal voluntary isometric contraction (MVIC) levels. …

Hyperglycemia is a diabetic condition with abnormally high levels of glucose in the bloodstream. Identifying bacterial strains is important not only for human health research but also for various environmental and industrial applications. The plethora of available shotgun metagenomic datasets provide an unprecedented opportunity to discover novel bacterial strains in different environmental niches. In this thesis, we evaluated the existing novel-strain-based tools and showed that their performance is still suboptimal. This study explores the relationships among parent, peer, and romantic attachment, identity processing style, and emotion regulation. Previous studies have revealed potential associations among each of the aforementioned factors (e.g., between identity and attachment as well as between attachment and emotion regulation), but this is the first study to examine the three factors together, putting emphasis on romantic attachment. It was predicted that attachment will vary depending on the interaction between identity processing style and emotion regulation. Huntington disease (HD) is a fatal neurodegenerative disease caused by CAG tract expansion in the huntingtin (HTT) gene, which results in production of mutant huntingtin (mtHTT) protein. Although mtHTT is expressed throughout life, onset of HD symptoms typically begins in mid-life, around 35 to 50 years of age. Characteristic HD symptoms include motor, cognitive, and psychiatric abnormalities. The emergence of symptoms in adulthood suggests that aging may play a role in HD pathogenesis. Prostate cancer is one of the leading causes of death for men in America as approximately 1 in 41 men will have prostate cancer. In this research, we focus on enzalutamide-resistant prostate cancer cells as cell resistance to enzalutamide is a prevalent obstacle in treating prostate cancer. We tested a novel compound library at different doses and observed each compound's efficacy in inducing apoptosis in enzalutamide-resistant cells. Furthermore, we analyzed the mechanism by which apoptosis was induced in compounds that showed a high efficacy at lower doses. This thesis examines psychological war trauma nurses experienced during the First World War. Psychological war trauma, or shell shock, as it was commonly known during the war, has largely been identified as a male affliction. In this thesis, I demonstrate that women too, suffered trauma and we can better understand nurses' trauma by applying some of the same analytical techniques that scholars have previously used to examine male combatant trauma. A survey of the first two decades of the French Communist Party's propaganda reveals a wide range of female imagery, from the androgynous, Soviet-style militant of the 1920s to the fashionable, feminine figure of the 1930s. Earlier scholars noting this discrepancy argued that the Party first adopted the Soviet "new woman," based on the Marxist principle of absolute gender equality but rejected it just over a decade later in order to broaden their appeal to the French masses. This dissertation expands on scholarship that examines efforts at incorporating a social justice framework into pedagogy, specifically, the role of technical and professional communication (TPC) faculty in addressing discriminatory digital technologies within a social-justice-oriented tech ethics pedagogy. I observe that despite the thousands of computer technology students who graduate from their programs every year in the US with the latest training, there seems to be no change to the adverse tech culture that produces discriminatory and racist work environments and digital technologies. Actin filament assembly and mechanics are crucial for muscle contraction, maintenance of cell structure, motility, and division. Actin filament assembly occurs in a crowded intracellular environment consisting of various types of molecules, including small organic molecules known as osmolytes. Ample evidence highlights the protective functions of osmolytes such as trimethylamine-N-oxide (TMAO), including their effects on protein stability and their ability to counteract cellular osmotic stress. This thesis investigates the representation of asexual individuals in the works of Murata Sayaka, Kawakami Mieko, and Kamatani Yuki, all of whom are contemporary Japanese writers that portray near–suffocating social environments in their depictions of modern-day Japan. Their texts illustrate the augmented demands Japanese society places upon a cross-section of asexual and neurodivergent individuals. Despite the thematic and character–related similarities in their works, I argue that each author presents a unique interpretation of how these asexual individuals interact with—and try to integrate into—wider Japanese society and mainstream culture.
