Genetic Engineering Of Plant Chloroplasts, Henry Daniell, Bruce Mcfadden
UCF Patents
Novel chimeric constructions and methods for their use are provided for expression of exogenous genes in a plant chloroplast. Particularly, expression is achieved by the use of a chloroplast or bacterial 5' untranslated region in the expression cassette. The expression cassette may be integrated into the chloroplast genome by the use of chloroplast DNA flanking sequences, or may replicate autonomously if provided with a chloroplast origin of replication. Plants and cells containing the transformed chloroplasts are also provided. The constructs may be used with both monocotyledenous and dicotyledenous chloroplasts.

The Effect Of Tourist Theft On Future Travel Decisions, Judith Lynn Holcomb

Past research has shown that theft is a prevalent crime against tourists. This study's purpose was to determine the effects of past incidents of personal theft on tourists' future decisions to travel by analyzing and comparing those who have experienced personal theft to those who heard of such incidents through personal accounts of friends or family. The findings, which were taken from a survey of 215 respondents, revealed that that experiencing personal theft, or knowing of someone who has, is not a deterrent for visiting a destination where the theft occurred. The role of Mycobacterium avium subsp paratuberculosis (MAP) in the etiology and pathogenesis of inflammatory bowel disease (IBD) including Crohn's Disease (CD), has been investigated. The fastidious characteristics and cross reactivity of MAP with other members in Mycobacteria have produced significant challenges in their detection and identification. In this two year pilot study, an array of three PCR molecular assays based on the detection of sequences from the16S rRNA, IS1245, and IS900 genes, belonging to members of the MAC, have been developed and optimized into a common protocol to be used as a rapid and accurate diagnostic tool regarding M. …

In the Indian River Lagoon, the nudibranch Okenia zoobotryon and its egg masses are found attached to the bryozoan Zoobotryon verticillatum throughout the year. Okenia zoobotryon is thought to live, feed, and reproduce exclusively on Z. verticillatum, which would make this a very specialized predator-prey interaction. The primary goal of my study was to document certain aspects of the ecological and chemical relationships between Z. verticillatum and O. zoobotryon. Specifically, I wanted to understand the cues used by the nudibranch to locate and remain on Z. verticillatum. Traumatic brain injury (TBI) induces a state of microglialactivation, which includes upregulation of macrophage functions and release inflammatory mediators such as certain inflammatory cytokines. Current literature suggests that interleukin-1Beta is an important cytokine mediator, which is dramatically increased after brain injury. Previous studies indicate that ATP is released by traumatically injured astrocytes and serves as a cell-to-cell mediator through purinergic receptors after in vitro injury. However, the mechanism of interleukin-1Beta release after traumatic brain injury remains poorly defined and is difficult to study using in vivo models. Mycobacterium avium subspecies paratuberculosis (map) is an intracellular pathogen that is known to parasitize macrophages and monocytes. Map infiltrates gastrointestinal tract host tissue where it is the known etiological agent of johne's disease in ruminants and implicated in the etiology of crohn's disease in humans. Map's ability to survive within macrophages enables it to disseminate throughout the rest of the host, possibly infecting other circulating blood leukocytes. To gain an understanding of the ecology of drift algae in the Indian River Lagoon system along the east coast of central Florida, four questions were addressed: 1) What is the composition and rate of accumulation of drift? 2) How much movement and turnover occurs within drift accumulations? 3) Do growth rates differ for drift versus attached algae? 4) Is there a difference in photosynthetic performance in drift versus attached algal species? Odor problems are a common complaint from residents living near landfills. Many compounds can cause malodorous conditions. However, hydrogen sulfide (h2s) has been identified as a principal odorous component from construction and demolition (c&d)debris landfills. Although several studies have reported the ambient concentrations of h2s near c&d landfills, few studies have quantified emission rates of h2s. The most widely used and proven technique for measuring gas emission rates from landfills is the flux chamber method. This thesis deals with some modifications of the original Sierpinski fractal patch antenna, providing additional parameters to optimize in order to get the required radiation characteristics. This dissertation consists of five papers concerning nitrification in chloraminated drinking water distribution systems in a one and a half year field study. Seven finished waters were produced from different treatment processes and distributed to eighteen pilot distribution systems (PDSs) that were made pipes taken from actual distribution systems. Unlined cast iron (UCI), galvanized steel (G), lined cast iron (LCI), and PVC pipes were used to build the PDSs. All finished waters were stabilized and chloraminated before entering the PDSs. Although modern medicine made great steps toward curing most diseases considered deadliest, yet cancer remains one of the major public health problems. The first part of this thesis investigates a new source of selective compounds that are potential candidates against cancer. Fifteen Russian compounds were tested in order to establish their efficiency against two types of cancer: human breast SK-Br-3 and colorectal carcinoma HT-29. The bioassay results show that seven of the new synthetic Russian compounds can be considered new drug leads, based upon their low toxicity and efficacy in slowing the growth of human breast cancer and colon cancer …

Five articles describing the impact of surface characteristics, and development of mass transfer models for diffusion controlled membrane applications are published in this dissertation. Article 1 (Chapter 3) describes the impact of membrane surface characteristics and NOM on membrane performance for varying pretreatment and membranes during a field study. Surface charge, hydrophobicity and roughness varied significantly among the four membranes used in the study. Membrane surface characteristics, NOM and SUVA measurements were used to describe mass transfer in a low pressure RO integrated membrane system. Biostability and related issues (e.g. nitrification) were investigated for 18 months in 18 pilot distribution systems, under various water quality scenarios. This study specifically investigated the impact of steady-state water changes on HPC levels in chlorinated and chloraminated distribution systems. Chlorination was more effective than chloramination in reducing HPC levels (1-2 log difference). There was a rapid increase in HPC corresponding to the change in steady-state water quality, which was observed in all PDS. Modeling effort demonstrated that HPC levels reached a maximum within five days after water quality change and return to initial level ten days after the change. …

Malaria kills at least one million people each year, mostly children - a death every 30 seconds. Almost one half of the world population is at risk from malaria. Antimalarial drugs are the only means for the treatment of about 500 million annual global malaria cases. Because of prevalent drug-resistance it is extremely urgent to identify new drug targets. Many proteins involved in eukaryotic signal transduction and cell cycle progression undergo post-translational lipid modification by a prenyl group. Presenilin mutations are responsible for most cases of autosomal dominant inherited forms of early onset Alzheimer disease. Presenilins play an important role in amyloid beta-precursor processing, NOTCH receptor signaling, and apoptosis. However, the molecular mechanisms by which presenilins regulate apoptosis are not fully understood. Here, we report that presenilin-1 (PS1) regulates the proteolytic activity of the serine protease Omi/HtrA2 through direct interaction with its regulatory PDZ domain. We show that a peptide corresponding to the cytoplasmic C-terminal tail of PS1 dramatically increases the proteolytic activity of Omi/HtrA2 toward the inhibitor of apoptosis proteins and beta-casein and induces cell death in …

The degenerate and nondegenerate two-photon absorption (2PA) spectra for a symmetric and an asymmetric fluorene derivative were experimentally measured in order to determine the effect of intermediate state resonance enhancement (ISRE) on the 2PA cross section delta. The ability to tune the individual photon energies in the nondegenerate 2PA (ND-2PA) process afforded a quantitative study of the ISRE without modifying the chemical structure of the investigated chromophores. Both molecules exhibited resonant enhancement of the nonlinearity with the asymmetric compound showing as much as a twentyfold increase in delta. Nonlinear photoinduced anisotropy in a bacteriorhodopsin film was theoretically and experimentally investigated and a broadband active optical limiter was demonstrated in the visible spectral range. A diode-pumped second harmonic yttrium aluminum garnet laser was used as a pumping beam and three different wavelengths at lambda=442, 532, and 655 nm from different lasers were used as probing beams. The pump and probe beams overlap at the sample. When the pumping beam is absent, the probing beam cannot transmit the crossed polarizers. With the presence of the pumping beam, a portion of the probing light is detected owing to the photoinduced anisotropy. …

Photoinduced birefringence in bacteriorhodopsin films was investigated using pump-probe method and its application for photonic switching explored. A diode-pumped second-harmonic YAG laser was used as a pumping beam and a diode laser at lambda=660 nm was used as a probing beam. The pump and probe beams overlap at the sample. Without the pumping beam, the probing light cannot transmit the analyzer to the detector. However, due to the photoinduced anisotropy, a portion of the probing light is detected when the pumping beam is present. In applications such as airport operations, military simulations, and medical simulations, conducting simulations in accurate and realistic settings that are represented by real video imaging sequences becomes essential. This paper surveys recent work that enables visually realistic model constructions and the simulation of synthetic objects which are inserted in video sequences, and illustrates how synthetic objects can conduct intelligent behavior within a visual augmented reality.

Telomere homeostasis, a process that is essential for continued cell proliferation and genomic stability, is regulated by endogenous telomerase and a collection of associated proteins. In this paper, a protein called KIP ( previously reported as a protein that binds specifically to DNA-dependent protein kinase), has been identified as a telomerase-regulating activity based on the following pieces of evidence.
