Cerium oxide (ceria)-based materials in the nanoscale regime are of significant fundamental and technological interest. Nanoceria in pure and doped forms has current and potential use in solid oxide fuel cells, catalysis, UV- screening, chemical mechanical planarization, oxygen sensors, and bio-medical applications. The characteristic feature of Ce to switch between the +3 and + 4 oxidation states renders oxygen buffering capability to ceria. The ease of this transformation was expected to be enhanced in the nanoceria. This dissertation demonstrated that the manipulation of substances at the molecular or nanometer level can lead to the discovery and development of new materials with interesting properties and important applications. Chapter 1 describes the development of a nanoscale molecular thin film material for corrosion protection. By using a self-assembled monolayer film with a thickness of only about 1 nanometer as a linkage, a covalent bonding was achieved between a polyurethane top coating and an aluminum alloy substrate. Integration of living cells with novel microdevices requires the development of innovative technologies for manipulating cells. Chemical surface patterning has been proven as an effective method to control the attachment and growth of diverse cell populations. Patterning polyelectrolyte multilayers through the combination of layer-by-layer self-assembly technique and photolithography offers a simple, versatile and silicon compatible approach that overcomes chemical surface patterning limitations, such as short-term stability and low protein adsorption resistance. The present study evaluates two fetal individuals (B532 and B625) from the Kellis 2 cemetery (Roman period circa A.D. 50 A.D. 450), Dakhleh Oasis, Egypt, that display skeletal anomalies that may explain their death. Both individuals exhibit bowing of the long bones in addition to other skeletal deformities unique to each individual. To assess these pathologies a differential diagnosis based on the congenital occurrence of long bone bowing is developed. Long bone bowing is selected because it is the more prevalent abnormality in the paleopathological literature and the other abnormalities are not as easily identifiable in the literature. For the …

Hormonally-triggered regulatory hierarchies play a major role in organismal development. Disruption of a single member of such a hierarchy can lead to irregular development and disease. Therefore, knowledge of the members involved and the mechanisms controlling signaling through such pathways is of great importance in understanding how resulting developmental defects occur. Type II transmembrane serine proteases (TTSPs) make up a family of cell surface-associated proteases that play important roles in the development and homeostasis of a number of mammalian tissues. The objective of this study was to obtain a bioassessment using benthic macroinvertebrates to meet TMDL (Total Maximum Daily Load) criteria for an oligotrophic (Lake Seminary) and an eutrophic (Lake Jesup) freshwater system in Seminole County, Florida. Monthly sampling of the benthic macroinverterbrate communities provided important biological data necessary to construct TMDL protocol and trophic state. Escherichia coli and Serratia liquefaciens, two common microbial spacecraft contaminants known to replicate under low atmospheric pressures of 25 mb, were tested for growth and survival in simulated martian conditions. Stressors of high salinity, low temperature, and low pressure were screened alone and in combination to determine how they might affect microbial activity. Growth and survival of E. coli and S. liquefaciens under low temperatures (30, 20, 10, or 5 °C) with increasing concentrations (0, 5, 10, or 20 %) of three salts believed to be present on the surface of Mars (MgCl2, MgSO4, NaCl) were monitored over 7 d. …

The analysis of Gene Regulatory Networks (GRNs) is essential to understanding the complete process of embryo development. Elucidating every gene regulatory circuit from maternal regulatory inputs all the way to the activation of differentiation gene batteries is an important step in increasing our understanding of developmental biology. In this work I study the cis-regulatory architecture of a pigment cell differentiation gene, polyketide synthase (SpPks) in the sea urchin Strongylocentrotus purpuratus. By nature discrete solitons represent self-trapped wavepackets in nonlinear periodic structures and result from the interplay between lattice diffraction (or dispersion) and material nonlinearity. In optics, this class of self-localized states has been successfully observed in both one-and two-dimensional nonlinear waveguide arrays. In recent years such lattice structures have been implemented or induced in a variety of material systems including those with cubic (Kerr), quadratic, photorefractive, and liquid-crystal nonlinearities. While several measures to assess emotional expression of products have been developed, the source of the emotion rating of a product or system was often unclear. Research to date indicates that parental and cognitive variables play a role in stress responses and health outcomes. Although researchers are beginning to focus on developmental processes in stress/health outcomes, there is little research examining which parental behaviors are most predictive of stress/health and whether cognitive variables mediate this relationship. As a result, the current study examines the self-reports of 160 late adolescents regarding parental behaviors, cognitive variables, and stress/health outcomes. In addition, blood pressure reactivity to a stressful situation was collected as a physiological measure of stress. Prostasin is expressed at the apical surface of normal epithelial cells and suppresses in vitro invasion of cancer cells. Prostasin re-expression in the PC-3 prostate carcinoma cells down-regulated the epidermal growth factor receptor (EGFR) protein expression and EGF-induced phosphorylation of the extracellular signal-regulated kinases (Erk1/2). We report here that prostasin and its activating enzyme matriptase are capable of inducing proteolytic cleavages in the EGFR extracellular domain (ECD) when co-expressed in the FT-293 cells, generating two aminoterminally truncated fragments EGFR135 and EGFR110, at 135 and 110 kDa. This article studies the propagation of initial disturbance in a quadratic autocatalytic chemical reaction in one-dimensional slab geometry, where two chemical species A, called the reactant, and B, called the autocatalyst, are involved in the simple scheme A + B - > 2B. Experiments demonstrate that chemical systems for which quadratic or cubic catalysis forms a key step can support propagating chemical wavefronts. When the autocatalyst is introduced locally into an expanse of the reactant, which is initially at uniform concentration, the developing reaction is often observed to generate two wavefronts, which propagate outward from the initial reaction zone. Similarities between Mycobacterium tuberculosis phoP-phoR mutants and the attenuated laboratory strain M. tuberculosis H37Ra in terms of morphological and cytochemical properties, lipid content, gene expression and virulence attenuation prompted us to analyze the functionality of this two-component regulator in the latter strain. Sequence analysis revealed a base substitution resulting in a one-amino-acid change in the likely DNA-binding region of PhoP in H37Ra relative to H37Rv. Using gel-shift assays, we show that this mutation abrogates the ability of the H37Ra PhoP protein to bind to a 40-bp segment of its own promoter. The discus fishes of the genus Symphysodon are popular ornamental cichlids that occur in floodplain lakes and flooded forests of the lowland Amazon Basin. These habitats are characterized by extreme seasonal fluctuations in the availability of food, shelter and dissolved oxygen, and also the densities of predators and parasites. Most aspects of discus biology are influenced by these fluctuating conditions. This paper reports an autoecological study of the western Amazonian discus S. haraldi (until recently classified as S. aequifasciatus). This species feeds predominantly on algal periphyton, fine organic detritus, plant matter, and small aquatic invertebrates. At high water it forages …

Supercontinuum (SC) generation is among the most interesting nonlinear optical effects lately discovered, due to the complexity of the mechanisms responsible for its generation. This phenomenon, first demonstrated by Alfano and Shapiro using picosecond pulses in condensed phase, has found novel applications in optical pulse compression, timeresolved spectroscopy and material characterization among many others. Here, we demonstrate that picosecond generated SC white-light in water preserves the polarization state: linear, elliptical and circular of the pump source. Brachyhypopomus draco, new species, is described from central, southern and coastal regions of Rio Grande do Sul state, Brazil, and Uruguay. It is diagnosed from congeners by, among other characters, the shape of the distal portion of the caudal filament in mature males, which during the reproductive period forms a distinct paddle shape structure.

Background: Many protein regions and some entire proteins have no definite tertiary structure, existing instead as dynamic, disorder ensembles under different physiochemical circumstances. Identification of these protein disorder regions is important for protein production, protein structure prediction and determination, and protein function annotation. A number of different disorder prediction software and web services have been developed since the first predictor was designed by Dunker's lab in 1997. Chelonid fibropapillomatosis-associated herpesvirus (CFPHV) is an alphaherpesvirus believed to cause marine turtle fibropapillomatosis (FP). A serodiagnostic assay was developed for monitoring sea turtle populations for CFPHV exposure. CFPHV glycoprotein H (gH) expressed in recombinant baculovirus was used in an enzyme-linked immunosorbent assay (ELISA) to detect virus-specific 7S turtle antibodies. Using captive-reared green turtles (Chelonia mydas) with no history of virus exposure as "known negatives" and others with experimentally induced FP as "known positives," the assay had 100% specificity but low sensitivity, as seroconversion was detected in only half of the turtles bearing experimentally induced tumors. Background: Social insects dominate ecological communities because of their sophisticated group behaviors. However, the intricate behaviors of social insects may be exploited by social parasites, which manipulate insect societies for their own benefit. Interactions between social parasites and their hosts lead to unusual coevolutionary dynamics that ultimately affect the breeding systems and population structures of both species. In this paper we report the development of nanosecond-pulsed fiber laser technology for the next generation EUV lithography sources. The demonstrated fiber laser system incorporates large core fibers and arbitrary optical waveform generation, which enables achieving optimum intensities and other critical beam characteristics on a laser-plasma target. Experiment demonstrates efficient EUV generation with conversion efficiency of up to 2.07% for in-band 13.5-nm radiation using mass-limited Sn-doped droplet targets. This result opens a new technological path towards fiber laser based high power EUV sources for high-throughput lithography steppers.

In measurement-based quantum computation, quantum algorithms are implemented via sequences of measurements. We describe a translationally invariant finite-range interaction on a one-dimensional qudit chain and prove that a single-shot measurement of the energy of an appropriate computational basis state with respect to this Hamiltonian provides the output of any quantum circuit. The required measurement accuracy scales inverse polynomially with the size of the simulated quantum circuit. This shows that the implementation of energy measurements on generic qudit chains is as hard as the realization of quantum computation. Considering the increased incidence of bacterial infections and the emergence of multidrug resistant bacteria at the global level, we designed superparamagnetic iron oxide nanoparticles as nanosensors for the assessment of antimicrobial susceptibility through magnetic relaxation. In this report, we demonstrate that iron oxide nanosensors, either dextran-coated supplemented with Con A or silica-coated conjugated directly to Con A, can be used for the fast (1) quantification of polysaccharides, (2) assessment of metabolic activity and (3) determination of antimicrobial susceptibility in blood. Background: Reelin and Notch-I signaling pathways have been recently found to be necessary to induce the expression of brain lipid binding protein ( BLBP) and to promote the process extension and the maturation of the neuronal progenitors, the radial glial cells. In this study, we report the cross talk between these two pathways. Results: Both in vitro Reelin treatment and overexpression of Notch-I intracellular domain (NICD) induced BLBP expression and a radial glial phenotype in an immortalized human neural progenitor (HNP) cell line, isolated from the cortex of 14 weeks old fetus. Central to the pathogenesis of Alzheimer disease is the aberrant processing of the amyloid precursor protein (APP) to generate amyloid beta-peptide (A beta), the principle component of amyloid plaques. The cell fate determinant Numb is a phosphotyrosine binding domain (PTB)-containing endocytic adapter protein that interacts with the carboxyl-terminal domain of APP. The physio-logical relevance of this interaction is unknown. Mammals produce four alternatively spliced variants of Numb that differ in the length of their PTB and proline-rich region. In the current study, we determined the influence of the four human Numb isoforms on the intracellular trafficking and processing of APP. …

Maturation of neuronal synapses is thought to involve mitochondria. Bcl-X-L protein inhibits mitochondria-mediated apoptosis but may have other functions in healthy adult neurons in which Bcl-X-L is abundant. Here, we report that overexpression of Bcl-X-L postsynaptically increases frequency and amplitude of spontaneous miniature synaptic currents in rat hippocampal neurons in culture. Bcl-X-L, overexpressed either pre or postsynaptically, increases synapse number, the number and size of synaptic vesicle clusters, and mitochondrial localization to vesicle clusters and synapses, likely accounting for the changes in miniature synaptic currents.
