Living at the northern limits of its geographic range, the Florida manatee is particularly susceptible to cold stress-related mortality during the winter months, with most deaths occurring in the lower two-thirds of the state. Contributing to this cold stress susceptibility is the manatee's limited physiological and behavioral responses available when thermally stressed. While capable of migrating south in response to falling water temperatures, manatees must still find warm water when ambient river temperature drops below 20°C for more than a few days. This is in part due to the species low metabolic rate, limited capacity for thermogenesis, and limited ability …

Cardiovascular disease is responsible for the loss of one life every 38 seconds and accounts for 26.6 percent of all infants that die of congenital birth defects. Adrenergic hormones are critically important regulators of cardiovascular physiology in embryos and adults. They are key mediators of stress responses and have profound stimulatory effects on cardiovascular function, and dysregulation of adrenergic function has been associated with many adverse cardiac conditions, including congenital malformations, arrhythmias, ischemic heart disease, heart failure, and sudden cardiac death. The glycine riboswitch is a structured RNA found upstream of genes in mRNA transcripts in many bacteria, functioning as a biofeedback gene regulator. Upon binding glycine, a complete RNA transcript including gene sequences is transcribed, effectively turning on gene expression. In an effort to understand the intricacies of its functioning, many mutants of the riboswitch were made and characterized during Ph. D. work, resulting in discovery of a P0 duplex/kink-turn motif involving a few nucleotides upstream of the established glycine riboswitch sequence which changed its ligand binding characteristics (Chapter 1). A problem representation is a cognitive structure created by the solver in correspondence to the problem. Sketching representative diagrams in the domain of physics encourages a problem solving strategy that starts from 'envisionment' by which one internally simulates the physical events and predicts outcomes. Research studies also show that sketching representative diagrams improves learner's performance in solving physics problems. Alasdair MacIntyre argues in favor of a historicist Thomism in ethics and political philosophy. In his theory, sociological categories take up much of the space traditionally occupied by metaphysics. This peculiar feature of MacIntyre's Thomism, and its merits and demerits, is already a subject that has been taken up by many critics. In this thesis, these criticisms are supplemented and unified by identifying what is perhaps the most fundamental difficulty with MacIntyre's ethics: his version of Thomism is problematic because it treats epistemology as first philosophy. High-risk alcohol consumption remains a primary public health concern for students on college campuses. In response to this concern the National Advisory Council of the National Institute on Alcohol Abuse and Alcoholism created a task force to identify and recommend strategies to aid college administrators in implementing effective alcohol programming at their institutions. While most administrators report being aware of these recommendations, many have not successfully implemented empirically supported interventions on their campuses. We propose a ratio estimator to determine population estimates using capture-recapture sampling. It's different than traditional approaches in the following ways: (1) Ordering of recaptures: Currently data sets do not take into account the "ordering" of the recaptures, although this crucial information is available to them at no cost. (2) Dependence of trials and cluster sampling: Our model explicitly considers trials to be dependent and improves existing literature which assumes independence. (3) Rate of convergence: The percentage sampled has an inverse relationship with population size, for a chosen degree of accuracy. Understanding factors limiting population growth is crucial to evaluating species persistence in changing environments. I used Integral Projection Models (IPMs) to elucidate the role of biotic interactions and disturbance on population growth rate in two plants: Helianthemum squamatum, a perennial endemic to gypsum habitats in central Spain, and Liatris ohlingerae, a long-lived perennial endemic to the Lake Wales Ridge of central Florida. In H. squamatum, there was a strong positive effect of trampling in the site with the highest plant density and moderate positive effects of seed addition in the site with the lowest plant density. The focus of this research was on the effect of school schedules on student achievement for ninth-grade students in a Florida school district. Data were collected from two central Florida high schools from the 2011-2012 and 2012-2013 school years. Five one-way analyses of covariance (ANCOVA) were performed to ascertain if there was any interaction between school schedules and student achievement. Examined were the interactions (a) between schedule and schools, (b) schedule and male students, (c) schedule and female students, (d) schedule and Black students, and (e) schedule and Hispanic students. This dissertation in practice presents a research-based model for staff development utilizing the elements of a professional learning community. The focus of this problem of practice was determined through an analysis of one high school's reading data indicating that 36% of the student body was reading below grade level according to the state assessment test for reading. Plaques of amyloid β peptide (Aβ) are a hallmark trait of Alzheimer’s disease (AD). However, the precise role of Aβ aggregates is not well understood. Recent studies have identified that naturally occurring N-terminal truncation and pyroglutamylation of Aβ significantly increases its neurotoxicity by an unknown mechanism. Content of pyroglutamylated Aβ (pE-Aβ) in AD brains has been shown to reach up to 50% of total Aβ. Modified pE-Aβ co-aggregates with Aβ by a seeding mechanism and forms structurally distinct and highly toxic oligomers. Ecological interactions within a community shape the structure of ecosystems and influence ecosystem function. Plant-pollinator interactions exist as mutualistic exchange networks that may collapse as habitat loss occurs, thereby threatening the overall health of an ecosystem. Understanding the impacts of human-mediated habitat disturbance on ecological interactions is therefore crucial for conservation efforts. As such, the common prerequisites serve as minimum admission requirements for schools of business in the state of Florida. Lyme disease, the most common tick borne disease in United States, is caused by the bacterial pathogen Borrelia burgdorferi. In nature, B. burgdorferi exists in an enzootic infectious cycle between an arthropod vector and mammalian hosts. Identification and characterization of the genes essential for B. burgdorferi survival throughout its infectious cycle is an important step toward understanding the molecular mechanisms involved in B. burgdorferi pathogenesis. Effective teacher questioning during whole group instruction remains an important pedagogy in science education, especially the importance of helping novice teachers to guide student thinking using effective questions. This study examined how novice secondary science teachers’ understand the relationship between student thinking and teacher questioning. The sample was seven novice secondary science teachers’ enrolled in the University of Central Florida’s job embedded Resident Teacher Professional Preparation Program (RTP3 ). Generation of foam cells, an essential step for reverse cholesterol transport (RCT) studies, uses the technique of receptor dependent macrophage loading with radiolabeled acetylated Low Density Lipoprotein (Ac-LDL). In this study, we used the ability of a biologically relevant detergent molecule, Lysophosphatidylcholine (Lyso PtdCho), to form mixed micelles with cholesterol or cholesteryl ester (CE) to generate macrophage foam cells. Fluorescent or radiolabelled cholesterol / Lyso PtdCho mixed micelles were prepared and incubated with RAW 264.7 or mouse peritoneal macrophages. Results showed that such micelles were quite stable at 4°C and retained the solubilized cholesterol during one month storage. Macrophages incubated …

Some grasshoppers and locusts are capable of expressing an extreme form of density-dependent phenotypic plasticity, known as locust phase polyphenism. The single largest cause of compound attrition during drug development is due to inadequate tools capable of predicting and identifying protein interactions. Several tools have been developed to explore how a compound interferes with specific pathways. However, these tools lack the potential to chronically monitor the time dependent temporal changes in complex biochemical networks, thus limiting our ability to identify possible secondary signaling pathways that could lead to potential toxicity. Identification of osseous materials is generally established on gross anatomical factors; however, highly fragmented or taphonomically altered materials are often problematic and alternative methods, such as biological, histological, or chemical analysis, must be utilized. Recently, chemical methods have been proposed to sort unknown materials according to their Ca/P ratios. Ubelaker and colleagues (2002) proposed using SEM/EDX to achieve this distinction and Christensen and colleagues (2012) have validated X-ray Fluorescence Spectrometry (XRF) for this application. The Baltic region was a vibrant center of power and economic prosperity in medieval Europe; Lithuania in particular. Until now, little stable isotopic analysis has been utilized to assess diet in this region during this time period. The aim of this study was to undertake a preliminary assessment of the composition of diet at late medieval Alytus (late 14th to early 18th centuries) from bone (N=35) and dentin (N=38) collagen samples. A quantitative study was conducted to determine if the amount of formulaic language influenced ESL teachers' perceptions (n=102) of non-native writing skill, as evidenced by composite and sub-scale scores on the ESL Composition Profile (Jacobs et al., 1981). Formulaic language was operationalized as 25 three-word strings sampled from the writing sub-list of the Academic Formulas List (Simpson-Vlach & Ellis, 2010) and further validated as frequent in the Michigan Corpus of Upper Level Student Papers. Since their recent introductions into Florida waters, three nonnative species [Perna viridis Linnaeus, 1758 (Asian green mussel), Mytella charruana d'Orbigny, 1846 (charru mussel) and Megabalanus coccopoma Darwin, 1854 (pink titan acorn barnacle)] have expanded both north and south along the Atlantic coast. Cholera toxin (CT) moves from the cell surface to the endoplasmic reticulum (ER) where the catalytic CTA1 subunit separates from the holotoxin and unfolds due to its intrinsic thermal instability. Unfolded CTA1 then moves through an ER translocon pore to reach its cytosolic target. Due to the instability of CTA1, it must be actively refolded in the cytosol to achieve the proper conformation for modification of its G protein target. The cytosolic heat shock protein Hsp90 is involved with the ER-to-cytosol translocation of CTA1, yet the mechanistic role of Hsp90 in CTA1 translocation remains unknown.
