The Role Of Activin A Signaling In Gastric Reflux-Related Diseases And The Progression To Esophageal Adenocarcinoma, Cedric J. Roudebush

Honors Undergraduate Theses

Gastroesophageal reflux disease (GERD), or acid reflux, affects 6-9 million people in the United States. It is characterized by a reflux of gastric acid and bile salts from the stomach into the esophagus, causing injuries to the esophagus known as Barrett's esophagus (BE). BE is the main risk factor for the development of esophageal adenocarcinoma (EAC), a devastating cancer in the esophagus whose molecular roots remain poorly understood. In recent years, evidence points to the esophageal epithelium itself as responsible for causing and promoting inflammation upon injury by gastric reflux, namely via an increase in inflammatory cytokine secretion. This project …

Unraveling Pdi And Its Interaction With Ab Toxins, Jessica Guyette Jan 2019

Unraveling Pdi And Its Interaction With Ab Toxins, Jessica Guyette

Electronic Theses and Dissertations

Protein disulfide isomerase (PDI) is an essential endoplasmic reticulum (ER) protein that acts as both an oxidoreductase and chaperone. It exhibits substantial flexibility and undergoes cycles of unfolding and refolding in its interaction with cholera toxin (Ctx), which is a unique property of PDI. This unfolding allows PDI to disassemble the Ctx holotoxin, which is required for Ctx activity. Here, we investigated the unfolding and refolding property of PDI and how this affects its interaction with bacterial toxins. PDI showed remarkable redox-linked conformational resilience that allows it to refold after being thermally stressed. Deletion constructs of PDI showed that both …

Investigation Of The Cell-Cycle Dependent Activity Of The Brca1-Rbbp8 Complex For Homologous Recombination, Jil Shah

Honors Undergraduate Theses

When cells undergo mitosis, they must replicate all six billion base pairs of DNA within the nucleus. With the sheer volume of information, it is impossible to replicate with 100% accuracy each time. Homologous recombination (HR) is one of many mechanisms the body has developed to correct and repair replication errors to DNA. HR is specific to double-stranded breaks to DNA, and it requires a sister chromatid to preserve the genetic code. BRCA1 and Rbbp8 interact to form a complex that is heavily involved in this process. Although there is a strong consensus about the involvement of these proteins in …
