In 2022, there were forty-two mass school shootings, a record that surpassed the previous years. As of April 2023, there have already been fourteen school shootings that have resulted in injury or death. Each year, the frequency of mass school shootings and the number of victims continue to rise. Regarding the shooters' characteristics, some have reported a history of mental illness, instability within the home, poor academic performance, and more. Cholera toxin (CT) is a classic A-B type protein toxin that has an A subunit (A1 + A2) and a pentameric B subunit. The catalytic A1 domain is linked to the A2 domain via a disulfide linkage. CTA1 must be dissociated from the rest of the toxin to cause a cytopathic effect. Protein disulfide isomerase (PDI) can reduce the CTA1/CTA2 disulfide bond, but disassembly of the reduced toxin requires the partial unfolding of PDI that occurs when it binds to CTA1. This unfolding event allows PDI to push CTA1 away from the rest of the toxin.

Farmworkers in Apopka, FL, have been subjected to overhead pesticide exposure since the 1940s. Pesticides including Paraquat (PQ), Metribuzin and Aldicarb were sprayed onto the field while farmworkers worked. In "Fed Up: The High Cost of Cheap Food," farmworkers recalled the physical toll these conditions took on their bodies, blaming pesticides for their diseases, such as chronic kidney disease (CKD). While established that pesticides, specifically PQ, may be involved in some forms of Parkinson's disease, no explicit connection has been identified for SLE, CKD, and other diseases experienced by farm workers. Polycystic Ovarian Syndrome (PCOS) is common among women of reproductive age (ages 15-49), and it can put patients at higher risk for diseases such as type II diabetes or coronary heart disease. While the mechanisms behind PCOS are poorly understood, it is diagnosed using two characteristics: hyperandrogenism and anovulation. Once diagnosed, many patients are also tested for insulin resistance (IR). While IR is thought of as a symptom of PCOS, this study will examine the effects of treatment of IR to manage symptoms of PCOS rather than treatment of hyperandrogenism. Simulation is a critical component of nursing education. It provides a safe environment to practice and evaluate skills, supplements clinical hours, and removes barriers to a well-rounded education. Nurses are expected to be educators as part of our scope of practice, and simulation can facilitate this. One topic of nursing importance is lactation education, as breastfeeding has been shown to have multiple benefits for both mother and baby. National rates of exclusive breastfeeding for the first 6 months do not currently meet CDC recommendations. Sexual violence (SV) is a pressing concern in the United States. SV (i.e., unwanted sexual contact, coercion, and wanted or unwanted penetration of another). Individuals with minoritized sexual, racial/ethnic, and gender identities experience worse psychosocial outcomes than their majority counterparts. People with multiple marginalized identities have been shown to experience traumatic events at greater rates and with significantly different outcomes compared to those with one minoritized identity. OBJECTIVE: Research has shown a clear relationship between perfectionism and other pathology including eating disorders, typically restrictive and purging disorders such as Anorexia Nervosa and Bulimia Nervosa. However, the relationship between perfectionism and loss of control eating disorder had not been explored. Similarly, we suspect there to be a relationship between perfectionism and self-discrepancy, and that shame might be a mediating variable in the relationship between these constructs and loss of control eating. This study engaged in a curricular redesign of an undergraduate biochemistry course undertaken by a collaborative action research group. The initial portion of this study investigated student conceptual understanding of foundational concepts via a rigorously tested concept inventory. the impact of the redesign on the student foundational conceptual understanding was determined by comparison with a baseline data set. Findings suggest that students enter and exit the biochemistry course with little understanding of key foundational concepts. Examination of student performance after the incorporation of creative exercises showed a significant impact of the curricular changes on two of the concepts of interest. …

Captive breeding can reduce the pressure on wild populations caused by overcollection, but if not properly monitored, wildlife laundering can lead to further population declines. Ultraviolet advanced oxidation processes were compared using sodium perborate (UV/NaBO3 AOP) or hydrogen peroxide (UV/H2O2 AOP) for 1,4-dioxane removal from tertiary wastewater effluent. Both UV/H2O2 and UV/NaBO3 AOPs were also tested with the addition of acetic acid. Results revealed that sodium perborate performed similarly to hydrogen peroxide – the UV/NaBO3 AOP with 6 milligrams per liter (mg/L) as H2O2 resulted in 43.9 percent 1,4-dioxane removal, while an equivalent UV/H2O2 AOP showed 42.8 percent removal. Although the oxidants performed similarly, NaBO3 is an average of 3.3 times more expensive than H2O2. However, the solid form of NaBO3 can provide a major …

The purpose of this dissertation was to investigate the intersectional awareness of preservice general education teachers as experienced through an internship in an urban, Title 1 funded school. Intersectionality Theory (Boveda, 2016; Crenshaw, 1990; Dill & Zambrana, 2009; Jones & Wijeyesinghe, 2011) was used as a theoretical framework to inform this study. A descriptive phenomenological design (Creswell, 2013; Moustakas, 1994) was used to examine preservice teachers' (PSTs)lived experiences within their teacher preparation program courses and internship. Endothelial cells, which form the inner layer of the vasculature, interact with the extracellular matrix (ECM) by exerting mechanical contractile cell-substrate stress called tractions and cell-cell stresses called intercellular stresses. This cellular mechanical behavior involves many fundamental biological processes, including cell migration, differentiation, angiogenesis, and wound healing. Also, the inner surface of the vasculature, where endothelial cells reside, is constantly exposed to various fluid flows. The most variable fluid flow regimes occur primarily within branching regions of the vasculature. Endothelial cells residing within these regions experience disturbed flow, which consists of irregular flow patterns and extreme wall shear stress gradients. …

The Migration period (4th-7th century AD) represents a time of major sociopolitical change, but relatively little is known about this time in Lithuania's history (Bliujien?, 2013; Jankauskas & Kozlovskaya, 1999). The focus of this dissertation is to understand the lived experiences of individuals interred in the Migration Period cemetery at Plinkaigalis, Lithuania, through integrated analyses of stable isotope, statistical and spatial methods to assess 253 human bone/tooth samples. This thesis investigates DNA dynamics and translocation through nanopores using Brownian dynamics (BD) simulations, offering insights into sequencing technologies, DNA marker detection, and accurate barcoding utilizing solid-state nanopore platforms. First, we in silico study the intricate process of capture and translocation in a single nanopore. Our simulation reveals a high probability of hairpin loop formation during the capture process. However, attaching a charged tag to one end of DNA improves multi-scan rates and enhances unidirectional translocations. Many Best Management Practices (BMPs) have been developed to reduce excessive nutrients in stormwater runoff and mitigate harmful algal blooms in downstream receiving water bodies. This study demonstrates a new BMP by comparing two green sorption media (i.e., specialty adsorbents) for nutrient removal in cascade upflow biofiltration systems operated in parallel. The proposed filtration technology can control hydraulic gradients, prevent clogging and settlements, and increase hydraulic loading while removing more nutrients in an integrated physicochemical and microbiological treatment process. Solid State Nuclear Magnetic Resonance (ssNMR) spectroscopy can be a powerful tool for investigating the atomic-level structures and dynamics of biological macromolecules, including proteins. In this dissertation, I present an ssNMR study of three diverse proteins, revealing insights into their respective secondary structures, conformational variations, and intermolecular interactions. Additionally, I introduce novel computational methods to facilitate the assignment of chemical shifts of ssNMR spectra. The first of the proteins is the capsid protein of the Rous Sarcoma Virus. Research describing conservation agencies has generally been exploratory and limited in the use of theory. Similarly, organizational theories can provide an understanding for how organizations change and adapt to their given environments, but they have not been applied to conservation agencies with unique challenges and strains from other public bureaucracies or private corporations.
