Ybor City is a historical neighborhood in Tampa, Florida, and a tourist attraction known for its immigrant roots and once-thriving cigar industry. This thesis places Ybor City into the context of the burgeoning heritage tourism market, examining how cities financially reliant on tourism often sanitize their public historical narrative. I identify the main actors involved in Ybor City's marketing and preservation by investigating contemporary newspaper articles and multiple National Park Service documents, thereby uncovering the motivations and decisions that led to Ybor's cultural image of a bustling, relatively peaceful early 20th-century "Latin" community. Vitamin D, folate, and cobalamin (vitamin B12) are crucial micronutrients in human physiology that are necessary for healthy calcium, phosphorus, and single-carbon metabolism. Recent studies have indicated that these vitamins also affect the inflammatory response in ways unrelated to their well-characterized deficiencies. Accordingly, analysis of their effect on chronic inflammatory diseases like Crohn's disease (CD) is warranted. This investigation examines the effects of vitamin deficiency on macrophages and intestinal epithelial cells upon exposure to Mycobacterium avium paratuberculosis (MAP,) a pathogen capable of triggering CD, to model the inflammatory response in clinical CD patients. Neuroblastoma is an aggressive pediatric cancer that is poorly responsive to traditional cancer therapies. Oncolytic viral (OV) vectors such as Zika virus (ZIKV) and Parainfluenza virus type 5 (P/V virus) are promising neuroblastoma therapeutics, but the role of innate immune responses in the effectiveness of OV killing is not well understood. Previous studies showed the neuroblastoma cell line SK-N-AS (expressing low CD24) had low permissivity for ZIKV infection, and this restriction was relieved by ectopic CD24 expression (CD24-high cells). Compared to permissive CD24-high cells, the non-permissive CD24-low cells had elevated basal levels of IRF-1, NF?B and phosphorylated STAT1; these cells …

In the face of unprecedented anthropogenic change, we increasingly turn to emergent technologies and extensive data sets for solutions that complement much needed systemic changes in our societies. These technological solutions, however, must be approached with care. We must recognize and address biases in the way data has been accumulated. In demographic studies, dormant life stages, such as seed banks, and other cryptic factors have often been neglected. Interdisciplinary research has demonstrated that learning and problem solving with advanced learning technologies (ALTs) such as intelligent tutoring systems, simulations, hypermedia, serious games, and virtual reality can promote and foster the development of 21st century skills (e.g., collaboration, problem solving, self-regulated learning) by measuring and using the interactions between cognitive, affective, metacognitive, motivational, and social (CAMMS) processes. Virtual reality (VR) games and educational systems commonly employ interaction cues to provide information on how to take appropriate actions at particular moments. Interaction cues can be employed for different purposes, such as informing the user to look, go, pick, and operate. Additionally, different types of interaction cues can directly affect usability and user experiences. Avicenna argues that the celestial spheres each have a soul, termed the motive soul, which is emanated by the first celestial intellect––a body of knowledge which knows itself. Despite outlining the powers of the motive soul, Avicenna does not formally investigate the psychology of the spheres nor their volition. Rather, he presents their volition as a mystery and leaves it to posterity to solve. The intent of this thesis is to explore the concept of creativity as it appears in the major works of Karl Marx and to attempt to discern its role in the emancipatory political project that Marx proposes. Contemporary understandings of creativity still rely upon notions of the artist-genius, locating the "true" expression of creative freedom in the work of art. This study investigates the impact of an exogenous and unexpected shock (COVID-19) on the wine tourism business from the winery's perspective. A sample of 146 Portuguese wineries was surveyed. The econometric results show that the share of wine tourism sales, the amount of dependence on exports and the assertiveness of brand recognition have a structural effect on direct-to-consumer tasting room wine sales, even when the winery's business is disrupted by a shock that degrades the dynamics and flows of international trade. E-cigarettes have recently become increasingly popular, especially amongst middle and high school students. Although they are marketed as safer alternatives to tobacco cigarettes, they produce toxic metals and carcinogenic nitrosamines. This thesis studies the effects of e-cigarette aerosol on the growth and proliferation of oral epithelial cells because the consequences of vaping, including a potential risk for aberrant growth leading to cancer, are not well understood. Cells were grown in matrigel, causing the formation of three-dimensional spheroids modeling the physiological architecture of the oral epithelium. The COVID-19 outbreak highlighted the important role that diagnostic tests play in the healthcare system. To reduce the impact of infectious disease outbreaks, the development of rapid and cost-effective point-of-care-tests (POCTs) is crucial. With the dissemination of the Monkeypox (Mpox) virus, it became a necessity to produce POCTS that are inexpensive and easy to use. This essay analyzes how the films Less Than Zero and The Doom Generation intersect with their overlapping themes regarding sex, sexuality, and death. The intended goal of the readings in this essay is to examine how queerness was represented in film during the AIDS crisis, specifically through the feature of a sexually fluid trio of friends. Friendship is not always magical, as the protagonists learn in "The Miss-Adventures of Molly & Sage." In this buddy-comedy set in a magical fantasy world, the titular characters are two teenage girls who go on epic quests and cause mischief. Many aspects of the story went through changes during the production of this hand-drawn animated short film. However, the main characters and their chaotic friendship remained consistent throughout the animation process. This research uses GIS methods to contextualize a Florida Cracker community in the Ocala National Forest. My case study is drawn from two seasons of fieldwork and supporting desktop surveys associated with Pat's Island, the home of multiple Cracker families. Cracker culture is historically categorized by its efforts to avoid modernization and prevent the commercialization of traditional lifeways in the quickly shifting society of late-19th and early 20th-century Florida. The geospatial expressions of these Cracker values are an emphasis on semi-remote living, adaptations to a unique environmental context, and the development of a semi-self-sufficient community. Sexualization happens when agency is taken away from the individual being sexualized, where one is seen, depicted, and treated like a sexual object rather than a person. This thesis aims to explore celebrities' reactions to their own public sexualization. To do this, an inductive approach was used when analyzing first-person narratives (written and video) and interviews (podcast, magazine and newspaper) by those whose sexualization is a notable feature of their career in the United States.
