The tumor microenvironment plays an important role in regulating cancer cell behavior. The tumor microenvironment describes the cancer cells, and the surrounding endothelial cells, fibroblasts, and mesenchymal stem cells, along with the extracellular matrix (ECM). The tumor microenvironment stiffens as cancer undergoes malignant progression, providing biophysical cues that promote invasive, metastatic cellular behaviors. The optimal defense hypothesis predicts that the allocation of plant defenses across plant organs is proportional to the importance of a given organ to plant fitness. Despite this, much less work has been devoted to the study of reproductive defenses in plants relative to vegetative structures like leaves. This study examines the apparancy hypothesis and the resource availability hypothesis using a phylogenetic comparative approach within the genus Cornus . Although previous research has identified a relationship between insecure attachment styles and symptoms of depression and anxiety, evidence regarding the mechanisms of action driving this relationship has been lacking. Consequently, the current study examined the mediating role of resilience in the relationship between insecure attachment styles (i.e. anxious-avoidant, anxious-ambivalent, helpless-disorganized and frightened-disorganized) and symptoms of depression and anxiety. The current study included a sample of 182 participants (i.e., 87 men and 95 women) who completed six questionnaires that assessed each participants' relationship with their caregivers during their childhood, present symptoms of depression and anxiety, and their resilience. The causality dilemma between dysbiosis and cancer has given rise to numerous studies both exploring the mechanisms behind cancer progression and the associative shifts in the microbiota upon carcinogenesis. Aside from the hallmark study of Dr. Barry Marshall in establishing the true causal relationship between Helicobacter pylori and gastric adenocarcinoma, studies have only been successful in adding associative links of carcinogenesis mediated by bacteria to the literature. Phenylethanolamine N-methyltransferase (Pnmt) is the enzyme that N-methylates norepinephrine to produce the stress hormone/neurotransmitter, epinephrine, which is abundantly expressed in adrenal glands. Developmental studies have also identified Pnmt expression in the embryonic heart and several areas of the brain, including brainstem, cerebellum, and hypothalamus. Thus, we hypothesize that selective ablation of Pnmt+ cells will have detrimental effects on cardiovascular, neuromuscular, and metabolic processes. To uncover the importance of Pnmt+ cells in vivo, we generated a novel Diphtheria Toxin A (DTA) suicide model (Pnmt+/Cre; R26+/DTA) to selectively ablate Pnmt-expressing (Pnmt+) cells in mice. AB-type protein toxins have a catalytic A subunit attached to a cell-binding B subunit. Ricin, Shiga toxin (Stx), exotoxin A, and diphtheria toxin are AB toxins that act within the host cytosol and kill the host cell through pathways involving the inhibition of protein synthesis. Our overall goal is to help elucidate the cellular basis of intoxication for therapeutic development. According to the current model of intoxication, the effect of AB toxins is irreversible. Atherosclerosis (ATH) is an inflammation-mediated disease in which cell death underlies the formation of lesions along the intima layer of vascular walls resulting in vessel narrowing, decreased blood flow, and increased risk of lesion rupture leading to myocardial infarction and stroke. The current study was undertaken to investigate whether inflammation in ATH can induce pyroptosis in vascular smooth muscle cells (SMC's). The Florida manatee (Trichechus manatus latirostris) was recently downlisted federally from "endangered" to "threatened" despite acknowledgments of remaining threats to long term population persistence. Challenges to future manatee conservation include, but are not limited to, increases in frequency of harmful algal blooms, intensifying anthropogenic disturbance, and loss of warm-water habitat. Understanding Sediment Biogeochemistry And The Role Of Juvenile Oysters On Recently Restored Eastern Oyster Reefs, Bryan Locher

In recent decades, goals for the restoration of eastern oyster (Crassostrea virginica) populations along the eastern coast of the United States have shifted from increasing harvestable oyster fisheries to enhancing the range of ecosystem services provided by oyster reefs. By filtering large volumes of water and releasing nutrient-rich feces and pseudofeces, oysters can locally enhance sediment biogeochemical cycling compared to that of unstructured benthic environments. An ongoing restoration program in Mosquito Lagoon, FL was leveraged to assess the immediate impacts ( < 1 year) of restoration on sediment biogeochemical properties of oyster reefs. The first study measured both short-term and long-term pools of carbon, nitrogen and phosphorus on dead, natural and restored reefs periodically over one year. The second study investigated one of the contributions to sediment nutrient pools by comparing feeding and feces/pseudofeces nutrient content of juvenile and older oysters. Results show that inorganic nitrogen and phosphorus pools can change within weeks after restoration and total nutrient pools by 6 months post-restoration. Restored reefs experienced a 136 % increase in ammonium, 78 % increase in total nitrogen, 46 % increase in total phosphorus, and 75 % increase in organic matter concentrations after 12 months of restoration. These nutrient increases were all positively correlated with oyster density, shell length and reef height measured on each reef. When standardized to grams of dry tissue weight, juvenile oysters showed significantly higher rates of chlorophyll-a removal, release of ammonium, and biodeposits with higher concentrations of dissolved organic carbon, nitrite + nitrate, and ammonium. The short-term changes to biogeochemical cycling on eastern oyster reefs within the first year of restoration are important to managers seeking to monitor ecosystem service recovery and overall coastal ecosystem health.

Examining diversity over multiple spatial and temporal scales affords the opportunity to develop a mechanistic understanding of the factors influencing community diversity dynamics, and how these may shift in a changing world. This thesis first examines multi-decadal fish community diversity metrics across a coastal biogeographic transition zone to quantify changes in species assemblages, assess relationships between fish community diversity and the abiotic environment, and capture potential shifts in the location of a putative biogeographic break. Smart drug-design for antibody and nanomaterial-based therapies allows for optimization of drug efficacy and more efficient early-stage pre-clinical trials. The ideal drug must display maximum efficacy at target tissue sites, but to track and predict distribution to these sites, one must have a mechanistic understanding of the kinetics involved with the individual cells of the tissue itself. Florida Scrub-Jays (Aphelocoma coerulescens, FLSJ; federally Threatened) are cooperatively breeding birds endemic to Florida and dependent on fire-maintained xeric oak scrub. FLSJs are year-round residents, highly territorial, and rarely disperse far from their natal territory. Lifetime reproductive success is highest among individuals breeding in early-successional habitat, usually less than 9 to 10 years post-fire. However, because scrub burns infrequently such early-successional, high-quality habitat is extremely limited and competition for it as breeding space is likely intense. Because some birds live long enough to experience habitat succession, FLSJs also occupy later-successional overgrown scrub, even though both survival and fecundity decline. Although …

Tumor Necrosis Factor alpha antagonists (anti-TNFα) have been extensively used for Crohn's disease (CD) treatment. Even though they may control CD symptoms initially, treatment response varies among patients, which seems to depend on single nucleotide polymorphisms (SNPs) in TNFα receptors superfamily 1A and 1B (TNFRSF1A/B). Ideally, when a neural network makes a wrong decision or encounters an out-of-distribution example, its predictive confidence should be as low as possible. Three primary contributions in this dissertation address this challenge. The first two contributions are new approaches to mitigate overconfident predictions in modern neural networks. In the first (1), called competitive overcomplete output layer neural networks, several classifiers, as part of the same output layer, are trained simultaneously and later their consensus produces more reliable predictions. North American amphibians have recently been impacted by two major emerging pathogens, the fungus Batrachochytrium dendrobatidis (Bd) and iridoviruses in the genus Ranavirus (Rv). Environmental, seasonal and host factors may play important roles in disease dynamics, but few studies incorporate these components into their analyses. Nutrient-laden stormwater runoff causes environmental and ecological impacts on receiving water bodies. Biosorption Activated Media (BAM) composed of the sand, tire crumb, and clay have been implemented in stormwater best management practices due to its ability to efficiently remove nutrients from stormwater runoff, such as in roadside linear ditches, via unique chemophysical and microbiological processes. It has applications in video surveillance, human computer interaction, video sports analysis, virtual and augmented reality, video retrieval etc. In this dissertation, we address four important tasks in video understanding, namely action recognition, temporal action localization, spatial-temporal action detection and video object/action segmentation. The ability of human beings to physically touch our surroundings has had a profound impact on our daily lives. Young children learn to explore their world by touch; likewise, many simulation and training applications benefit from natural touch interactivity. As a result, modern interfaces supporting touch input are ubiquitous. Typically, such interfaces are implemented on integrated touch-display surfaces with simple geometry that can be mathematically parameterized, such as planar surfaces and spheres; for more complicated non-parametric surfaces, such parameterizations are not available. Here is a war-haunted Korean vet, brainwashed Nazi brides, a neurotic Soviet ballerina, and a re-imagined Ethan Frome. In these stories, anxiety waits in the wings: will Ethan's wife discover his affair? Will the brides acknowledge the dark truth behind their training? Will a mother recover her kidnapped baby? A vast scholarship has been dedicated to examining the discrimination racial and ethnic minorities endure in the United States. Research has also shown racial and ethnic minorities work to resist discrimination via various social processes. One ethnic minority group absent from the literature is the American Roma, more commonly known as Gypsies. Therefore, this study aimed to gain insights into the situation of the American Roma, including the discrimination they endure and the resistance tactics they employ.
