New breeding methods have provided scientists with opportunities to improve traits in a wide range of crops, however, there remains resistance to foods that are produced from these crops, and mandates on labels used to describe such processes continue to be a source of policy debate. Here we focus on gene editing and examine (i) whether consumer acceptance varies when the technology is applied to different ingredients (unrefined versus highly refined ingredients) and (ii) the impact of two different claims related to gene editing (health-focused claim versus an environment-focused claim). Our results show that consumers are less likely to purchase …

Computational Discovery Of Microstructured Composites With Optimal Stiffness-Toughness Trade-Offs, Beichen Li, Bolei Deng, Wan Shou, Tae-Hyun Oh, Yuanming Hu, Yiyue Luo, Liang Shi, Wojciech Matusik

Mechanical Engineering Faculty Publications and Presentations

The conflict between stiffness and toughness is a fundamental problem in engineering materials design. However, the systematic discovery of microstructured composites with optimal stiffness-toughness trade-offs has never been demonstrated, hindered by the discrepancies between simulation and reality and the lack of data-efficient exploration of the entire Pareto front. We introduce a generalizable pipeline that integrates physical experiments, numerical simulations, and artificial neural networks to address both challenges. Without any prescribed expert knowledge of material design, our approach implements a nested-loop proposal-validation workflow to bridge the simulation-to-reality gap and find microstructured composites that are stiff and tough with high sample efficiency. …

Vaccine Immunogenicity Versus Gastrointestinal Microbiome Status: Implications For Poultry Production, Chrysta N. Beck, Jiangchao Zhao, Gisela F. Erf Feb 2024

Vaccine Immunogenicity Versus Gastrointestinal Microbiome Status: Implications For Poultry Production, Chrysta N. Beck, Jiangchao Zhao, Gisela F. Erf

Poultry Science Faculty Publications and Presentations

At the turn of the 21st century, the importance of maintaining a balanced microbiome was brought to the forefront of the microbiology, immunology, and physiology research fields. Exploring the complex interactions between vaccine administration, mucosal microbiome, oral tolerance, and enteric inflammation in health and disease is challenging since environmental factors (such as diet and sanitation) have major influences on gut microbiota composition. High enteric pathogen load has been shown to contribute to dampened cell-mediated and humoral immune responses to vaccines in human case studies, either through elevated enteric inflammation or increased tolerance to environmental microbes. Although antibiotic and probiotic interventions …

Antimicrobial Tolerance In Salmonella: Contributions To Survival And Persistence In Processing Environments, Tomi Obe, Aaron S. Kiess, Ramakrishna Nannapaneni

Poultry Science Faculty Publications and Presentations

Salmonella remains a top bacterial pathogen implicated in several food-borne outbreaks, despite the use of antimicrobials and sanitizers during production and processing. While these chemicals have been effective, Salmonella has shown the ability to survive and persist in poultry processing environments. This can be credited to its microbial ability to adapt and develop/acquire tolerance and/or resistance to different antimicrobial agents including oxidizers, acids (organic and inorganic), phenols, and surfactants. Moreover, there are several factors in processing environments that can limit the efficacy of these antimicrobials, thus allowing survival and persistence. This mini-review examines the antimicrobial activity of common disinfectants/sanitizers used …

Molecular Genomic Analyses Of Enterococcus Cecorum From Sepsis Outbreaks In Broilers, Douglas D. Rhoads, Jeff Pummill, Adnan Ali Khalaf Alrubaye

Poultry Science Faculty Publications and Presentations

Extensive genomic analyses of Enterococcus cecorum isolates from sepsis outbreaks in broilers suggest a polyphyletic origin, likely arising from core genome mutations rather than gene acquisition. This species is a normal intestinal flora of avian species with particular isolates associated with osteomyelitis. More recently, this species has been associated with sepsis outbreaks affecting broilers during the first 3 weeks post-hatch. Understanding the genetic and management basis of this new phenotype is critical for developing strategies to mitigate this emerging problem. Phylogenomic analyses of 227 genomes suggest that sepsis isolates are polyphyletic and closely related to both commensal and osteomyelitis isolate …

Bacterial wilt (BW), caused by Ralstonia solanacearum, is one of the devastating diseases in tomatoes (Solanum lycopersicum L.). The use of resistant cultivars and breeding for genetic resistance is the most effective, economical, and environmentally friendly management strategy for this disease. It is necessary to screen diverse germplasm and cultivated genotypes to identify resistant resources and to develop resistant cultivars in tomatoes to combat the changing pathogen isolates. This study evaluated 40 United States Department of Agriculture (USDA) tomato accessions for their BW resistance to the R. solanacearum isolate P822 under greenhouse conditions. The tomato plants were inoculated …

Phosphorus (P) fertilizers with low water solubility, like struvite (MgNH4PO4·6H2O), have been identified to possibly reduce nutrient losses in furrow-irrigated cropping systems. However, there is a lack of research on the impacts of P and nitrogen (N) fertilization on greenhouse gas [GHG; i.e., methane (CH4), nitrous oxide (N2O), and carbon dioxide (CO2)] production in furrow-irrigated rice (Oryza sativa). The objective of this study was to evaluate the effects of electrochemically precipitated struvite (ECST), chemically precipitated struvite (CPST), triple superphosphate (TSP), diammonium phosphate (DAP), environmentally smart nitrogen …

Herbicide-resistant barnyardgrass and weedy rice control, without crop injury, is a challenge for rice producers in the United States. Herbicides not initially labeled for rice, such as oxyfluorfen, are now being evaluated as new tools for weed control. The ROXY® trait allows for the use of oxyfluorfen in rice for weed control preemergence and postemergence. Experiments were initiated in 2021 and 2022 to evaluate (1) the effectiveness of preemergence- and postemergence-applied oxyfluorfen on barnyardgrass and weedy rice, (2) the sensitivity of oxyfluorfen-resistant rice to oxyfluorfen as a function of application timing, and (3) the influence of soil moisture on oxyfluorfen-resistant …

Pig gastrointestinal tracts harbor a heterogeneous and dynamic ecosystem populated with trillions of microbes, enhancing the ability of the host to harvest energy from dietary carbohydrates and contributing to host adipogenesis and fatness. However, the microbial community structure and related mechanisms responsible for the differences between the fatty phenotypes and the lean phenotypes of the pigs remained to be comprehensively elucidated. Herein, we first found significant differences in microbial composition and potential functional capacity among different gut locations in Jinhua pigs with distinct fatness phenotypes. Second, we identified that Jinhua pigs with lower fatness exhibited higher levels of short-chain fatty …

In this study, we have successfully constructed a comprehensive database of metagenome-assembled genomes (MAGs) pertaining to the gut microbiota of the giant panda. Through our analysis, we have identified significant reservoirs of antibiotic resistance genes (ARGs), namely Escherichia coli, Citrobacter portucalensis, and Klebsiella pneumoniae. Furthermore, we have elucidated the primary contributors to ARGs, including Streptococcus alactolyticus and Clostridium SGBP116, in both captive and wild pandas. Additionally, our findings have demonstrated a higher prevalence of ARGs in the metagenome, with notable expression of the RPOB2 gene in S. alactolyticus. Crucially, 1217 ARGs shared homology with human gut …

The underrepresentation of women and the perpetuation of gender stereotypes in children's literature has long been a concern. This study examined the portrayal of women in children's picture books and the potential of interactive read-alouds to promote gender equity in early childhood education, with the aim of shedding light on gender inequality. This study used critical discourse analysis, theoretically informed by Rosenblatt's transactional theory and gender schema theory, to analyse 19 books and interactive read-aloud sessions between preservice teachers and children in six preschools. The results showed that books on women's inequality are discussed in three categories: the absence of …

Mo-Doped Titanate Nanofibers From Hydrothermal Syntheses For Improving Bone Scaffold, Yang Tian Jan 2024

Mo-Doped Titanate Nanofibers From Hydrothermal Syntheses For Improving Bone Scaffold, Yang Tian

Materials Science and Engineering Faculty Publications and Presentations

A longstanding interest in bone tissue engineering is the development of new bio- scaffolds that can be manufactured on a large scale with high throughput at low cost. Here, we report a low-cost and systematically optimized hydrothermal synthesis for producing Mo- doped potassium titanate nanofibers with high structural purity. This new nanosynthesis is based on bone tissue growth on an undoped titanate nanowires-entangled scaffold, as previously reported by our team. The morphological and structural characterization data suggest that the crystal structure of Mo-doped titanate nanofibers closely resembles that of the undoped ones. This resemblance is potentially valuable for assessing the …

Turtle Species And Ecology Drive Carapace Microbiome Diversity In Three Seasonally Interconnected Wetland Habitats, Matthew Parks, Jun Sheng Lee, Kassandra Camua, Ethan Hollender

Biological Sciences Faculty Publications and Presentations

Different species of freshwater turtles exhibit primary behaviours ranging from aerial basking to benthic bottom-walking, cycle between wet and dry conditions at different time intervals, and undertake short-distance overland movements between aquatic habitats. These behaviours in turn may impact the accumulation of microbes on external shell surfaces of turtles and provide novel niches for differentiation of microbial communities. We assessed microbial diversity using 16S and 18S rRNA metabarcoding on carapace surfaces of six species of freshwater turtles residing in three adjacent and seasonally interconnected wetland habitats in southeast Oklahoma (United States). Communities were highly diverse, with nearly 4200 prokaryotic and …

Deciphering The Double-Edged Action Of Sting Pathway In Cancer, Neelabh Datta Jan 2024

Deciphering The Double-Edged Action Of Sting Pathway In Cancer, Neelabh Datta

Inquiry: The University of Arkansas Undergraduate Research Journal

Cancer cells have unstable chromosomes and damaged DNA, which can activate the cellular alarm system cGAS-STING. This mechanism detects DNA in the incorrect location (the cytosol) and produces type I interferons, which can recruit immune cells to combat cancer. However, certain tumours can bypass or use this mechanism for their own benefit. This short article examines how cGAS-STING influences the different cells of the tumour microenvironment (TME), and how it can have both anti-tumour and pro-tumour effects. It also demonstrates how additional STING signals can cause inflammation and contribute to cancer growth. To employ cGAS-STING against cancer, it is critical …
