Analysis Of The Mechanical, Microstructural, And Biocompatibility Properties Of Novel Magnesium-Based Hybrid Nanocomposite Materials For Orthopedic Applications, Thomas H. Mcgehee

Biomedical Engineering Undergraduate Honors Theses

Bone fractures and complete joint replacements are becoming more common, primarily because of accidents and age-related issues. Conventional solutions to treat these orthopedic injuries involve inert metal alloys to correct bone fractures until the healing process is complete, where a secondary revision surgery is then required to remove the implant. Issues regularly associated with these solutions include stress shielding, bone resorption, infection, inflammation, and implant loosening. Magnesium (Mg)-based nanocomposites offer promising potential as an alternative to commonly used metal alloys because of their favorable mechanical properties and biocompatibility. A particularly advantageous property of Mg is its natural biodegradation in vivo, …

Justification For The Biological Foundation Of Methane On Mars: Methanogens’ Capacity To Metabolize On Montmorillonite Clay And Basalt, Ashley Ezeldin May 2025

Justification For The Biological Foundation Of Methane On Mars: Methanogens’ Capacity To Metabolize On Montmorillonite Clay And Basalt, Ashley Ezeldin

Biological Sciences Undergraduate Honors Theses

The possibility of extraterrestrial life, particularly on Mars, remains a compelling question in astrobiology. Mars' harsh atmospheric conditions, dominated by carbon dioxide and extreme cold, prevent the survival of aerobic species. However, it still leaves the possibility of methanogens, an anaerobic species that can survive in extreme conditions, to inhabit this planet. Due to the discovery of methane on Mars, it is possible that methanogens may live and grow in the subsurface of this planet.

For this experiment, basalt and montmorillonite clay were chosen as substrates because they are believed to be found in the composition of Mars's subsurface. Methane, …

Characterizing The Medial Forebrain Bundle In Adolescents With Anhedonic Depression, Tina Nguyen May 2025

Characterizing The Medial Forebrain Bundle In Adolescents With Anhedonic Depression, Tina Nguyen

Biological Sciences Undergraduate Honors Theses

Adolescent depression has quickly grown in prevalence, yet the underlying neuropathology remains poorly understood. Anhedonia–the diminished ability to experience pleasure–is a core symptom of melancholic depression. Early identification of biomarkers associated with anhedonic depression could help mitigate the severity of the disorder by enhancing treatment efficacy. Blunted functionality of the nucleus accumbens (NAcc), the reward center of the brain, and abnormal dopaminergic activity in the ventral tegmental area (VTA) have both been linked to depression. Furthermore, glutamatergic input from the hypothalamus modulates the VTA which in turn, downregulates dopamine release in the NAcc. However, there is limited evidence supporting that …

The Effect Protein Supplementation On Waist-To-Hip Ratio, Bmi, Lipid Profile, And Resting Energy Expenditure In Postmenopausal Women, Caleb Flores

Biological Sciences Undergraduate Honors Theses

Postmenopausal women are a growing and at risk population that face increased risk for adverse changes in body composition, lipid profile, and overall cardiometabolic health due to hormonal changes associated with aging. Over 40% of postmenopausal women have obesity, which can be linked not just aging but the menopausal transition. Because of the negative effects of menopause on cardiometabolic, research into dietary interventions to combat these effects are needed. High-protein diets have shown promise in mitigating these effects, but sufficient research has not been done to determine the effects on postmenopausal women. This study aimed to evaluate the effects of …

Assessment Of Nitrogen Source Utilization During Nitrogen Fixation And Nitrogenase Regulation By Methanosarcina Acetivorans, Avery Carter May 2025

Assessment Of Nitrogen Source Utilization During Nitrogen Fixation And Nitrogenase Regulation By Methanosarcina Acetivorans, Avery Carter

Biological Sciences Undergraduate Honors Theses

Nitrogen is a crucial element in biomolecules such as proteins and DNA, essential for all cellular life. Biological nitrogen fixation, a process utilized by certain bacteria and archaea, reduces dinitrogen (N₂) to ammonia (NH₃), enabling growth in the absence of fixed nitrogen sources like NH₃. This process is driven by the metalloenzyme nitrogenase, which catalyzes the reaction: N₂ + 16ATP + 8e⁻ + 8H⁺ → 2NH₃ + H₂ + 16ADP + 16Pi. Due to its high energy demand, nitrogenase production and activity are tightly regulated. Among archaea, nitrogenase is found exclusively in methanogens, where its regulation remains poorly understood. We …

Activity-Based Protein Profiling On Nucleophilic Amino Acid Residues With Pyrocarbonates, James Van Horn

Chemistry & Biochemistry Undergraduate Honors Theses

The complexity of the proteome requires strategies that efficiently characterize protein functions. Activity-Based Protein Profiling (ABPP) is a method that identifies enzyme activity by targeting key catalytic residues. This study focused on histidine, a residue involved in enzymatic catalysis, metal coordination, and protein-protein interactions. Despite its importance, histidine-specific chemical probes are limited due to its variable reactivity. This work developed and characterized a histidine-specific probe using a pyrocarbonate-based mechanism. Singleresidue assays confirmed selective reactivity of the probe with histidine over other nucleophilic amino acids. Further testing on enzymatic proteins and lysed cell proteomes showed that the probe modified histidine residues …

Quantifying Freshwater Salinization Impacts On Nitrate Removal Via Denitrification In Urban Streams, Brynnen Beck May 2025

Quantifying Freshwater Salinization Impacts On Nitrate Removal Via Denitrification In Urban Streams, Brynnen Beck

Crop, Soil and Environmental Sciences Undergraduate Honors Theses

As the urban world expands, the construction of impervious surfaces increases erosion and runoff into nearby streams. Urbanization also adds waste from human activity into the environment. One of these wastes is runoff from road salt application in winter months, which can increase stream salinization and interfere with other important ecological processes, such as denitrification. Therefore, the objective of this study was to compare nitrate removal via denitrification between two urban streams to understand the role of road salts in freshwater salinization. We compared conductivity measurements between the streams to see how background conductivity (a proxy for salinity) influences resilience …

Over-Imitation In Adolescents With Autism Compared With Typically Developing Peers: An Expanded Literature Review, Elizabeth Floeck

Communication Disorders and Occupational Therapy Undergraduate Honors Theses

Abstract

Background:

Over-imitation, the act of imitating both relevant and irrelevant actions, is a behavior commonly found in human learning and social development. It is often observed in typically developing (TD) children and is believed to play a role in both social and learning development. While the exact motivations behind over-imitation remain a mystery, over-imitation appears to be a socially motivated behavior. Children may over-imitate when they know certain actions are unnecessary, suggesting this behavior stems from social acceptance and norms. In individuals with autism spectrum disorder (ASD), social and communicative behaviors are drastically different from TD individuals. In individuals …

Identifying Novel Molecular Targets To Induce Ferroptosis In Drug-Tolerant Cancer Cells Through Computational Modeling, Haris J. Rana May 2025

Identifying Novel Molecular Targets To Induce Ferroptosis In Drug-Tolerant Cancer Cells Through Computational Modeling, Haris J. Rana

Biological Sciences Undergraduate Honors Theses

Ferroptosis is an iron-dependent, reactive oxygen species (ROS)-driven form of regulated cell death in which the cell membrane is degraded by lipid peroxidation. It is regulated by multiple enzymes, such as glutathione peroxidase 4 (GPX4), which prevents membrane degradation by reducing lipid peroxides and protecting cells from oxidative damage. When inhibitors like RSL3 target GPX4, lipid peroxides build up, and toxic ROS compromises membrane integrity. Traditional anticancer treatments, such as most chemotherapies, aim to eliminate tumor cells by damaging DNA and triggering apoptosis. However, resistance to apoptosis is a “hallmark of cancer,” often resulting in treatment failure. As a distinct …

Examining The Effects Of Quercetin On Phenotypic Characteristics Of Human Mesenchymal Stem Cells, Thomas M. Needy

Biomedical Engineering Undergraduate Honors Theses

Introduction A significant obstacle to mesenchymal stem cell (MSC) potency and therapeutic utility is in vitro senescence, an irreversible cessation of replication associated with age-related complications. Senolytic drugs, such as quercetin, may be helpful in selectively culling senescent cells while leaving non-senescent cells unaffected, thereby increasing potency of high-passage MSCs.

Methods The phenotypic, genotypic, and immunomodulatory effects of quercetin were assessed using in vitro models. Senescent cells, created through repeated subculturing of MSCs in vitro, and non-senescent cells were treated with 10 μM quercetin, differentiated into osteocytes, adipocytes, and chondrocytes, and analyzed to observe the effect of quercetin.

Results Quercetin …

Atomic Level Characterization For The Transport Cycle Conformational Pathways Of Multidrug Resistance Protein 1 (Mrp1), Samuel Wamwere Mwatha

Graduate Theses and Dissertations

The multidrug resistance-associated protein 1 (MRP1/ABCC1) is an ATP-binding cassette (ABC) transporter that mediates the cellular efflux of endogenous and xenobiotic substrates, including therapeutic drugs. Its overexpression is a major contributor to multidrug resistance in cancer, which tends to result in negative clinical results. Even with considerable progress made in structural biology, the intricate details of how membrane lipid composition affects the conformational changes and functional switches of MRP1 still lack explanation.

This thesis employs long-timescale, all-atom molecular dynamics (MD) simulations to study the influence of various lipids on MRP1 lipid bilayer environments. Both inward-facing and outward-facing conformations of MRP1 …
