Highly Pathogenic Avian Influenza (HPAI) presents a substantial economic risk to the poultry industry. Domesticated fowl contract HPAI initially through exposure, direct or indirect with migratory waterfowl and outbreaks can result in significant economic losses to growers and the poultry industry at large. A HPAI outbreak occurred in Minnesota and Iowa and spread across over 13 other states in 2014 and 2015. This caused an estimated $1.6 billion in losses (CDC, 2016) and led to shortages of eggs and turkeys together with elevated prices (Anni et al. 2005). Lipins are a family of proteins that have critical functions in the control of fat storage and energy homeostasis. Biochemically, lipins have two functions. They provide an enzymatic activity (phosphatidate phosphatase or PAP activity) in the glycerol-3 phosphate pathway that leads to the production of storage fats (triacylglycerols). In addition, they play a role in the regulation of genes in the cell nucleus as transcriptional co-regulators. The PAP activity of lipins has been widely studied in a number of organisms. Microdialysis is a diffusion-based sampling method that can be useful for monitoring various biological systems. Matrix metalloproteinases are a class of enzymes responsible for remodeling the extracellular matrix that, when dysregulated, are linked to various diseases. The delivery method of microdialysis is of particular interest as a sampling technique for enzymatic reactions. Microdialysis was performed in vitro using a model enzyme, porcine pancreatic elastase, because it is a useful substitute for matrix metalloproteinases. A colorimetric substrate for elastase, succinyl-ala-ala-ala-p-nitroanilide, and its product p-nitroaniline were measured using a UV-Vis spectrophotometer. The objective of this thesis was to identify polymorphisms in the glucocorticoid receptor (GR) and interleukin-8 receptor (CXCR2) genes and to associate genotypes between the above mentioned polymorphisms and production traits in crossbred cattle. The hypothesis was that polymorphisms will exist for GR and CXCR2 genes and will be linked to production traits. Glucocorticoid receptors have been positively associated with higher milk yields, lactose content, feed intake, and feed conversion rates. Transposon mutagenesis is a technique that has been widely used for annotating gene function and has now been combined with next-generation sequencing (Tn-Seq) to assess mutant fitness on a genome wide basis. Ground cover management systems affect soil quality and health and thereby orchard growth and productivity. There have been few studies in the southern US on the effects of managed drive-rows using cover crops as part of a sustainable apple orchard management system. A field study used treatments of 1) seasonal legumes (cowpea [Vigna unguiculata] and crimson clover [Trifolium incarnatum]), 2) seasonal grasses (millet [Setaria italic] and annual rye [Lolium multiflorum]), or 3) unmanaged natural vegetation drive row plantings, with mowed vegetation blown into the tree row as mulch (mow/blow) nested variable. In this honors thesis, I focus on young adult novels that highlight rather than minimize the experience of having a mental disability, and I argue that this topic must be included in education and book discussions in order to create more inclusive classrooms and communities. I examine representations of depression, anxiety, bipolar disorder, and autism in popular young adult novels, paying special focus to the way characters represent the relationship between power and disability. Protein targeting is a fundamental cellular process that directs proteins from their site of synthesis to the site where they function. The signal recognition particle (SRP) dependent targeting pathway is conserved in both eukaryotes and prokaryotes where it co-translationally targets polypeptide chains emerging from ribosomes to the endoplasmic reticulum (eukaryotes) or cytoplasmic membrane (prokaryotes). A structurally unique form of SRP is found in chloroplasts where it functions to post-translationally bind and target a subset of integral thylakoid membrane proteins, the light harvesting chlorophyll binding proteins (LHCPs). Intratumoral heterogeneity is ubiquitously present within primary tumors and contributes to intractable behaviors such as metastasis and mutability spatiotemporally. Mounting evidence has shown that heterogeneous cell populations can adversely affect cell metabolism and metastatic potential. The cell’s only fluorescent molecules within the electron transport chain, flavin adenine dinucleotide (FAD) and nicotinamide adenine dinucleotide (NADH), can allow the quantitation of cell metabolism. We demonstrate the use of the optical redox ratio (FAD/(NADH+FAD)) to determine the metabolic behaviors of a heterogeneous panel of cells with varying metastatic programs at normal conditions and following acute hypoxia. In the CNS, glial scar formation often occurs following TBI, and astrocytes are widely believed to contribute to this scar formation. While the role of astrocytes in extracellular matrix (ECM) production is known, the exact mechanism(s) for this event remain unclear. As the body ages, physical and cognitive declines can result in balance and mobility deficits, but research has shown that proper nutrition and exercise can help maintain physical and mental capacity. The purpose of this study was to analyze the relationship between habitual dietary protein intake and dual task performance in sedentary (SED), recreationally active (RA), and masters athletes (MA). It is well known that arginine vasotocin (AVT) in birds is involved in physiological homeostasis such as cardiovascular, osmotic regulation as well as reproductive functions. Pertinent to these physiological functions, AVT immunoreactive (-ir) neurons in the hypothalamus have been found associated with hemorrhage, dehydration, oviposition and other physiological regulation. Evidence, however, suggests that AVT also plays significant roles in modulating behavior, memory, stress, and food intake. Designed transmembrane peptides were employed for investigations of histidine residues within the hydrophobic environment of the lipid bilayer by means of oriented solid-state deuterium NMR spectroscopy. Using the model peptide GWALP23 sequence (GGALW(LA)6LWLAGA) as a host framework, the effects of single and double histidine mutations were explored. Replacement of leucine residue 12 to polar neutral histidine had little influence on the peptide average orientation, however under strongly acidic pH conditions in DOPC bilayers, the histidine becomes positively charged (pKa 2.5) and the GWALP23-H12 peptide exits the membrane and adopts a surface-bound orientation. Due to the ecological importance of crayfish and the increasing vulnerability of freshwater systems due to habitat loss, invasive species, and hydrologic alteration, understanding crayfish-environment relationships is crucial in the context of aquatic species conservation. I sought to examine the influence of hydrologic variation among intermittent and permanent streams on crayfish occupancy, abundance, predation risk, and potential vulnerability to invasive species effects. I conducted crayfish and environmental data sampling during two consecutive summers across 20 Ozark streams of differing permanence levels (10 intermittent, 10 permanent). Monochamus (Coleoptera: Cerambycidae) are a genus of longhorn beetles commonly known as pine sawyers. They have a worldwide distribution coincident with pines (Pinaceae) and are vectors of the pinewood nematode (Bursaphelenchus xylophilus). In the United States, there are eight known Monochamus species and they have a sympatric distribution with at least one other Monochamus species throughout their range. Monochamus are known to attack stressed, dead, and dying conifers especially pines. This thesis applies southern literary theory to contemporary young adult literature (YAL) in order to analyze constructions and representations of the South and adolescence(ts) in the texts discussed. Arguing that Harper Lee’s To Kill a Mockingbird acts a master narrative for popular conceptions of southern adolescent identity, this project proposes that Lee’s 2015 novel Go Set a Watchman along with contemporary publications in YAL disrupts the master narrative Mockingbird and Scout Finch present readers. Weed-resistance management has become a topic of concern for modern agriculture. Cost related to herbicide usage has increased greatly due to evolution and proliferation of resistant weeds. A sacramental poetics requires a particular mode of being-in-the-world. Religiously-minded poets, from Dante and Milton to Donne and Herbert, have long considered how the individual becomes attuned with creation and God’s will. But what happens when modernity and secularization challenge long-held assumptions about the universe and how humankind fits into it? A reevaluation is then needed. Rails (Family: Rallidae) are among the least studied birds in North America, in large part due to their elusive nature. As a wetland-dependent species, understanding the timing of their migration and their habitat needs during migration is especially important since management needs to be timed to balance the needs of many species. I developed and verified a new distance sampling based nocturnal ATV spotlight survey because traditional call-broadcast surveys are not effective during autumn migration because of the drop off in call rate after the breeding season. Interpersonal hate, the affective state of one individual experiencing hate toward another, is widely experienced. However, to date there is no experimental evidence of interpersonal hate. This phenomenon was explored, as was its place in the triad of hostile emotions. It was theorized that interpersonal hate would be differentiated from the hostile emotions (contempt, anger, and disgust) by appraisal of violation type and by behavioral response, and that the hostile emotions could be recognized by these characteristics, as well.
