Microbiota Analysis Of Chickens Raised Under Stressed Conditions, Rabindra Kumar Mandal, Tieshan Jiang, Robert Wideman Jr, Troy Lohrmann, Young Min Kwon

Poultry Science Faculty Publications and Presentations

A substantial progress has been made toward understanding stress-associated gut and extraintestinal microbiota. However, a comprehensive understanding of the extraintestinal microbiota of chickens raised under stressed conditions is lacking. In this study, chickens were raised on a wire-floor model to induce stress, and the microbiota in the gut (ceca) and extraintestinal sites (blood, femur, and tibia) were characterized at different ages (1, 17, and 56 days) using 16S rRNA gene microbiota profiling. Open reference OTU picking showed extraintestinal sites had a significantly higher number of unassigned OTUs compared to ceca across all ages of chickens. Extraintestinal sites of all ages, …

Tall fescue (Schedonorus arundinaceus) is a cool-season perennial grass that is widely used as a forage for many livestock species including sheep. An endophyte (Neotyphodium coenophialum) in tall fescue produces ergot alkaloids that enhance plant survival but produce toxicosis in animals. The objective of this study was to investigate the rumen microbiota from gestation and lactation in ewes grazing tall fescue pastures with high (HA) or moderate (MA) levels of endophyte infection, and their relationship with serum parameters. Data were collected at the beginning of the study (d1), the week before initiation of lambing (d51), and at …

Methanosarcina Acetivorans Contains A Functional Isc System For Iron-Sulfur Cluster Biogenesis, Thomas M. Deere, Divya Prakash, Faith H. Lessner, Evert C. Duin, Daniel J. Lessner

Biological Sciences Faculty Publications and Presentations

Background

The production of methane by methanogens is dependent on numerous iron-sulfur (Fe-S) cluster proteins; yet, the machinery involved in Fe-S cluster biogenesis in methanogens remains largely unknown. Methanogen genomes encode uncharacterized homologs of the core components of the ISC (IscS and IscU) and SUF (SufBC) Fe-S cluster biogenesis systems found in bacteria and eukaryotes. Methanosarcina acetivorans contains three iscSU and two sufCB gene clusters. Here, we report genetic and biochemical characterization of M. acetivorans iscSU2.

Results

Purified IscS2 exhibited pyridoxal 5′- phosphate-dependent release of sulfur from L-cysteine. Incubation of purified IscU2 with IscS2, cysteine, and iron (Fe2+) resulted …

Minimal Structural Elements Required For Midline Repulsive Signaling And Regulation Of Drosophila Robo1, Haley E. Brown, Timonthy A. Evans

Biological Sciences Faculty Publications and Presentations

The Roundabout (Robo) family of axon guidance receptors has a conserved ectodomain arrangement of five immunoglobulin-like (Ig) domains plus three fibronectin type III (Fn) repeats. Based on the strong evolutionary conservation of this domain structure among Robo receptors, as well as in vitro structural and domain-domain interaction studies of Robo family members, this ectodomain arrangement is predicted to be important for Robo receptor signaling in response to Slit ligands. Here, we define the minimal ectodomain structure required for Slit binding and midline repulsive signaling in vivo by Drosophila Robo1. We find that the majority of the Robo1 ectodomain is dispensable …

Chlorophyll content is an important indicator of the growth status of japonica rice. The objective of this paper is to develop an inversion model that can predict japonica rice chlorophyll content by using hyperspectral image of rice canopy collected with unmanned aerial vehicle (UAV). UAV-based hyperspectral remote sensing can provide timely and cost-effective monitoring of chlorophyll content over a large region. The study was based on hyperspectral data collected at the Shenyang Agricultural College Academician Japonica Rice Experimental Base in 2018 and 2019. In order to extract the salient information embedded in the high-dimensional hyperspectral data, we first perform dimension …

Crop, Soil and Environmental Sciences Faculty Publications and Presentations

Pollen-mediated gene flow (PMGF) refers to the transfer of genetic information (alleles) from one plant to another compatible plant. With the evolution of herbicide-resistant (HR) weeds, PMGF plays an important role in the transfer of resistance alleles from HR to susceptible weeds; however, little attention is given to this topic. The objective of this work was to review reproductive biology, PMGF studies, and interspecific hybridization, as well as potential for herbicide resistance alleles to transfer in the economically important broadleaf weeds including common lambsquarters, giant ragweed, horseweed, kochia, Palmer amaranth, and waterhemp. The PMGF studies involving these species reveal that …

Berries are a rich source of phytochemicals, especially phenolics well known for protective activity against many chronic diseases. Berries also contain a complex mixture of volatile compounds that are responsible for the unique aromas of berries. However, there is very limited information on the composition and potential health benefits of berry volatiles. In this study, we isolated phenolic and volatile fractions from six common berries and characterized them by HPLC/HPLC-MS and GC/GC-MS, respectively. Berry phenolic and volatile fractions were evaluated for an anti-inflammatory effect using lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells by measuring levels of pro-inflammatory cytokines and the nuclear factor-kappa …

Tuning Network Dynamics From Criticality To An Asynchronous State, Jingwen Li, Woodrow L. Shew Sep 2020

Tuning Network Dynamics From Criticality To An Asynchronous State, Jingwen Li, Woodrow L. Shew

Physics Faculty Publications and Presentations

According to many experimental observations, neurons in cerebral cortex tend to operate in an asynchronous regime, firing independently of each other. In contrast, many other experimental observations reveal cortical population firing dynamics that are relatively coordinated and occasionally synchronous. These discrepant observations have naturally led to competing hypotheses. A commonly hypothesized explanation of asynchronous firing is that excitatory and inhibitory synaptic inputs are precisely correlated, nearly canceling each other, sometimes referred to as ‘balanced’ excitation and inhibition. On the other hand, the ‘criticality’ hypothesis posits an explanation of the more coordinated state that also requires a certain balance of excitatory …

Sodium Butyrate Reduces Salmonella Enteritidis Infection Of Chicken Enterocytes And Expression Of Inflammatory Host Genes In Vitro, Anamika Gupta, Mohit Bansal, Basanta Raj Wagle, Xiaolun Sun, Narayan Rath, Annie Donoghue, Abhinav Upadhyay

Poultry Science Faculty Publications and Presentations

Salmonella Enteritidis (SE) is a facultative intracellular pathogen that colonizes the chicken gut leading to contamination of carcasses during processing. A reduction in intestinal colonization by SE could result in reduced carcass contamination thereby reducing the risk of illnesses in humans. Short chain fatty acids such as butyrate are microbial metabolites produced in the gut that exert various beneficial effects. However, its effect on SE colonization is not well known. The present study investigated the effect of sub-inhibitory concentrations (SICs) of sodium butyrate on the adhesion and invasion of SE in primary chicken enterocytes and chicken macrophages. In addition, the …

The tips in the tree of life serve as foci for conservation and management, yet clear delimitations are masked by inherent variance at the species–population interface. Analyses using thousands of nuclear loci can potentially sort inconsistencies, yet standard categories applied to this parsing are themselves potentially conflicting and/or subjective [e.g., DPS (distinct population segments); DUs (Diagnosable Units-Canada); MUs (management units); SSP (subspecies); ESUs (Evolutionarily Significant Units); and UIEUs (uniquely identified evolutionary units)]. One potential solution for consistent categorization is to create a comparative framework by accumulating statistical results from independent studies and evaluating congruence among data sets. Our study illustrates …

Crop, Soil and Environmental Sciences Faculty Publications and Presentations

Alternative water management practices for rice (Oryza sativa) production have been developed for water conservation purposes, such as the furrow-irrigated rice production system, which results in variable volumetric water content (VWC), temperature, and oxidation-reduction (redox) potential conditions. The spatial and temporal variations in environmental factors can affect greenhouse gas (GHG) emissions. The objective of this field study was to document and evaluate the continuous variations in soil water content, temperature, and redox potential over two growing seasons (2018 and 2019) among site positions (up-, mid-, and down-slope) and tillage treatments [conventional tillage (CT) and no-tillage (NT)] under furrow-irrigated …

Testing Flowering Perennial Plants In A Bermudagrass (Cynodon Spp.) Lawn, Paige Elizabeth Boyle, Michelle Wisdom Aug 2020

Testing Flowering Perennial Plants In A Bermudagrass (Cynodon Spp.) Lawn, Paige Elizabeth Boyle, Michelle Wisdom

Horticulture Faculty Publications and Presentations

Most pollinating insects require a season-long succession of floral resources to fulfill life-cycle requirements. Incorporating forbs into turfgrass sites may create a season-long sequence of flowers to support foraging pollinators. However, persistence of forbs in warm-season turfgrasses such as bermudagrass (Cynodon spp.) may be affected by the competitive nature of the turfgrass and routine management practices such as mowing. A 2-year study was conducted to evaluate seven forbs (Bellis perennis L., Lotus corniculatus L., Prunella vulgaris L., Trifolium fragiferum L. ‘Fresa’, Trifolium repens L. ‘Durana’ and ‘Resolute’, Trifolium subterraneum L.) for persistence and ability to produce floral resources …

Design And Testing Of A Multi-Depth Soil Temperature Sensor Array Intended As A Candidate Irrigation Scheduling Tool, Lillian Glaeser Aug 2020

Design And Testing Of A Multi-Depth Soil Temperature Sensor Array Intended As A Candidate Irrigation Scheduling Tool, Lillian Glaeser

Biological and Agricultural Engineering Undergraduate Honors Theses

A crucial measure in the advancement of water conservation and sustainable agricultural strategies is increasing efficiencies of irrigation systems. Because of the lack of availability of affordable, durable, and scalable soil moisture monitoring devices, this thesis proposes a low-cost, multi-depth soil temperature sensor array as a candidate for monitoring soil moisture content and guiding farmer’s irrigation scheduling. Propagation of diurnal temperature waves through soil is dependent on the thermal diffusivity of the soil, which can be determined by examining temperature waves at different soil depths. Thermal diffusivity is dependent on several factors, though, in undisturbed soil, water has the largest …

By identifying and analyzing the research output from faculty and graduate students from the Food Science Department to determine where they published and the sources …

Dissecting The Cellular Control Of Septin Organization In A Global Cereal Killer, Nawaraj Dulal Jul 2020

Dissecting The Cellular Control Of Septin Organization In A Global Cereal Killer, Nawaraj Dulal

Graduate Theses and Dissertations

Rice blast disease, caused by the filamentous fungus Magnaporthe oryzae, destroys sufficient rice each year to feed 60 million people, and is a serious threat to global food security. A wheat-adapted lineage of M. oryzae now poses threat to global wheat production. Rice blast disease is currently controlled using limited fungicides, and the emergence of fungicide resistance within M. oryzae populations is a growing concern. There is a pressing need to identify new classes of fungicides to control the disease, which requires better understanding of the basic biology of the pathogen. To establish disease, M. oryzae forms a specialized dome …

The Role Of Designers In Promoting Healthy Masculinity With An Approach To Stopping Violence Against Women And Girls, Fatemeh Abolbashari Jul 2020

The Role Of Designers In Promoting Healthy Masculinity With An Approach To Stopping Violence Against Women And Girls, Fatemeh Abolbashari

Graduate Theses and Dissertations

While awareness about violence against women and girls is growing, there is still a lack of evidence about what changes behavior to prevent it from happening. As a graphic designer, I wanted to find the root problem of this issue and work towards a solution. Through my research and questioning, I concluded that this problem begins with men’s thoughts on hierarchy, language, and behavior, and supported by a prevalence of toxic masculinity in men’s culture.

Where does this violence come from? What has society done to raise this kind of man? Violence against women and girls is being couched in …

Diaporthe species (anamorph: Phomopsis) are associated with a wide range of plant hosts as plant pathogens, asymptomatic endophytes, and saprobes. One of these hosts is soybean, which is one of the most important crops in U.S. agriculture. Several Diaporthe species cause important diseases on soybean in the U.S., and specifically in Arkansas. The taxonomy, genetic diversity, and pathogenicity of Diaporthe species associated with asymptomatic infection of soybean are rarely studied with accurate molecular tools. Therefore, this dissertation aimed to assess the diversity and boundaries of Diaporthe associated with soybean in Arkansas. Furthermore, pathogenicity and alternative lifestyles were assessed among Diaporthe …
