While the field of brain aging research continues to grow at a rapid pace, there is still much to learn about how certain factors influence brain aging as well as when and how these factors can be intervened upon. Cerebrovascular and neurodegenerative diseases continue to pose a major obstacle to healthy brain aging and the better we understand the risk factors of these diseases, the greater chance we have at mitigation strategies and prevention. Previous research has identified key factors, consisting of physical, lifestyle, and socio-emotional aspects of life, which have the greatest impact on brain aging. These factors overlap …

Ischemic limb disease continues to pose major clinical challenges. The effect of this disease causes restricted blood flow, which is followed by a limited ability to regenerate in the vascular tissues. The purpose of this study is to address the current limitations in producing large-scale generation of Smooth Muscle Cells (SMCs) derived from human induced pluripotent stem cells (hiPSCs) and then assessing their potential therapeutic efficiency individually and in combination with endothelial cells (ECs) for vascular repair. Providing care for an individual with Alzheimer’s Disease and Related Dementias (ADRD) is considered the most stressful type of family caregiving that impacts caregivers’ health. More than one in five ADRD family caregivers report their overall health as poor to fair. These adverse health outcomes are often exacerbated when caregivers’ emotional, informational, and practical needs remain unmet throughout the caregiving trajectory. Numerous studies have shown that the ways in which caregivers cope to caregiving-related stressors significantly shape their health outcomes. The goal of this dissertation is to examine the varying perspectives, or frames, of providers regarding decision-making in the NICU, which includes how they view neonates and their families, their roles as providers, and the way their frames possibly impact the care they give to the infants. In this dissertation, I conduct in-depth interviews with 34 NICU providers and professionals that provide their services in the NICU, including doctors, nurse practitioners, nurses, various therapists (i.e. respiratory, physical, occupational), and two other providers from two other professions. While drug overdose deaths have generally decreased in the past year, close to 60,000 overdoses involving opioids occurred in 2024. The opioid crisis has lasted over two decades and is now in the fourth wave. The fourth wave has been characterized by overdoses resulting from fentanyl adulteration in the drug supply and polysubstance overdoses involving fentanyl. The purpose of the current study is to compare the perceived harm of fentanyl to other drugs of abuse and identify differences in the perceived harm of fentanyl between groups of people who use substances. Tyrosine kinase inhibition (TKI) is highly effective and promotes favorable long-term outcomes for many patients with chronic myeloid leukemia (CML). However, many patients experience treatment resistance or recurrence after TKI cessation due to leukemic stem cell (LSC) persistence within the hematopoietic stem cell (HSC) niche. Within the niche, macrophages adopt different activation states to alter HSC function, however, CML disrupts niche signals to selectively support LSCs and macrophages may contribute to this process. We hypothesize that macrophages contribute to pro-inflammatory activation within the niche to promote LSC development and nilotinib will reprogram macrophages to selectively support non-leukemic stem cells. Chromatin structure is a central regulator of gene expression and cellular identity, yet the chromatin structural factors driving cardiac disease and regeneration remain incompletely defined. This dissertation investigates how modulation of chromatin architecture shapes pathological and regenerative cardiac phenotypes through integrated single-cell transcriptomic, epigenomic, and functional analyses. To determine whether chromatin structural regulators distinguish healthy from diseased cardiac states, we curated a comprehensive list of chromatin structural genes and applied it to single-nuclei RNA sequencing datasets from human hearts with and without dilated cardiomyopathy (DCM). The purpose of this dissertation was to explore the complexities of the informed consent (IC) process in the context of neonatal cardiac interventions, with a focus on parental knowledge and anxiety during decision-making. Across three manuscripts, this work examined conceptual dimensions, feasibility, and acceptability of a new prenatal treatment (palliative care (PC) support sessions), and the effect sizes of this new treatment on parental knowledge and anxiety levels surrounding the IC discussion for neonatal cardiac palliations. The first manuscript employed dimensional analysis to conduct a concept analysis of IC in surgical care. The review of 27 publications revealed three distinct …

The Centennial Olympiad, hosted by Atlanta, Georgia, in 1996, demonstrated key principles of the postsouthern mindset, a movement typically associated with literary criticism that, if applied to historiographic methods, offers valuable insights into how Atlantans confronted competing identities of being a modern international city hailed as the Black Mecca, and a city forever linked to the Lost Cause, Ku Klux Klan, and racial inequality. Living donor kidney transplantation (LDKT) is the most effective treatment for patients with end-stage kidney disease; however, living donation rates have remained suboptimal over the past two decades. As a result, many interventions have been developed and tailored to increase living donation. The Living Donor Navigator (LDN) Program is one effort designed to help transplant candidates identify potential living donors and guide them through the donation process, using a combination of advocacy and systems training. The LDN Program is a lay-navigator-based intervention that equips participants with the knowledge, communication skills, and confidence needed to identify and engage potential living donors. …

Alzheimer’s disease is a progressive neurodegenerative disorder that causes damage to the neurons that are present in the brain. Progress in the management and treatment of AD is limited by lack of early diagnostics, which are critical to the development of effective therapies. It is therefore critical to develop non-invasive/cost efficient biomarkers to aid in early diagnosis and interventions to prevent or delay dementia onset. The term Early Life Stress (ELS) encompasses traumatic events occurring before the age of 18 such as physical abuse, verbal abuse, household dysfunctions, sexual abuse, childhood neglect, child maltreatment, and adverse childhood experiences. The traumatic psychological and physical experiences in early life are widely known to cause enduring effects on mental and physical health in adulthood. Of note, individuals with ELS have adverse health outcomes earlier in adulthood and with more severity such as in the case of Systemic Lupus Erythematous (SLE), an autoimmune disease. This work addresses key questions relating to the end of the Cold War. Contemporary wisdom suggests that Ronald Reagan’s policies beat the Soviet Union into submission, but this is not the case. Reagan’s initial policies were antagonistic towards the Soviet Union and his devotion to SDI later harmed his ability to establish relations with Michail Gorbachev later on. This thesis argues that it was George H.W. Bush, rather than Reagan, who was the more influential and effective President when dealing with the Soviet Union. Bush’s diplomacy was less idealized and more realistic than Reagan’s. Additionally, Bush was able to successfully …
