Cystic Fibrosis (CF) is a genetic disorder that primarily impacts the respiratory and gastrointestinal systems. With advances in treatment and medications, the life expectancy of people with CF is continuing to increase with current life expectancy at 47 years of age. Given the increased life expectancy, people with CF are interested in childbearing but may have CF specific fertility issues that should be addressed by their CF healthcare provider. Currently, these conversations are not occurring or are suboptimal. Our cross-sectional study aimed to investigate joint specificity of concentric muscle torque enhancement after a maximum eccentric contraction for the knee versus ankle joints across two different movement velocities (120°/s and 180°/s). After a familiarization session, 22 healthy young adults randomly performed concentric (CONC) and maximum eccentric preloaded concentric (EccCONC) muscle strength tests of the knee extensors and ankle plantar flexors of the non-dominant leg on an isokinetic strength testing device. We calculated the ratio between EccCONC and CONC (EccCONC/CONC) for all the conditions as the marker of concentric muscle torque enhancement. Tissue damage, whether a traumatic injury or surgically-induced, can compromise organ structure and function. Traumatic injury affects 38 million people annually in the U.S alone, costing around 4.8 billion dollars. Much research has been accomplished on defining the pathways associated with recovery, but research on understanding the role of nutrition in wound healing is limited. Zebrafish (Danio rerio) provide a high-throughput model to elucidate nutrients that are important in the regenerative process due to their reproductive capacity, rapid development, cost compared to other animal models and, lastly, their ability for tissue regeneration. The overall risk of infection after total knee arthroplasty (TKA) has not improved over the past 20 years, remaining 10-40 times greater than the risk of infection of a native knee. Despite extensive research into periprosthetic joint infection (PJI), little is known about the synovial fluid immune homeostasis of the native knee joint and how TKA surgery itself impacts the local immune composition. While many postulate a role for cartilage in maintaining this immune homeostasis, the exact method in which it contributes remains elusive. Although people can lose weight by restricting calories and increasing energy ex-penditure, only a small percentage are successful at maintaining the loss. PURPOSE: Therefore, the purpose of this study is to identify factors that may predict who is more likely to gain weight and visceral fat back after weight loss, and to evaluate the efficacy of a weight maintenance support group (EatRight) during 12 months of weight mainte-nance. METHODS: Subjects (n=85) were healthy, overweight [body mass index (BMI) 27–30 kg/m2, mean weight 77.6 kg] premenopausal women (age 34). Pathogens and plants are involved in a coevolutionary battle for dominance. As plants lack adaptive immune systems, they rely on their innate immunity to protect themselves from infectious agents. Upon pathogen attack, plant cells activate their pro-survival strategy by recognizing the pathogen and actively transcribing and translating proteins to effectively defend against the pathogen infection. As the intensity of infection increases, the heightened demand for proteins places an excessive burden on the ER machinery resulting in the accumulation of misfolded or unfolded proteins within the ER. The gain of function of nuclear factor erythroid 2 like 2 (Nrf2), in the absence of oxidative stress (or toxic insults), its pathophysiology arising from the endoplasmic reticulum and exercise intervention to mitigate the pathology are the focal point of this dissertation. Nrf2, when constitutively expressed, has been previously associated with cardiomyopathy. Chapter II investigated the hypothesis that cardiac-specific expression of Nrf2 (mNrf2-TG) would establish a pro-reductive state and trigger adaptive cardiac remodeling by enhancing the basal antioxidant defense. Obesity is a global epidemic that affects countless numbers of people caused by genetic and environmental factors including circadian disruption, which poses significant health risks to skeletal muscle physiology and other tissues. While a feeding fasting intervention known as time-restricted feeding (TRF) has been shown to mitigate obesity-related muscle dysfunction, the underlying mechanisms remain a subject of investigation. Objective: Exogenous ketone administration presents a possible treatment strategy for obesity and obesity-related diseases. The exogenous ketone ester, R,S-1,3-butanediol diacetoacetate (BD-AcAc2) decreases adiposity and hepatic steatosis in high-fat dietinduced obese mice. In our previous work, carbohydrate energy was removed and replaced with the ketone ester to produce isocaloric diets. The current investigation aimed to determine whether BD-AcAc2 decreases markers of hepatic steatosis and inflammation in lean mice on a high-fat, high-sugar (HFHS) diet without removing carbohydrate energy. In the United States, acute kidney injury (AKI) affects nearly 20% of all intensive care unit patients. Additionally, over a third of Americans aged 50 or older suffer from stage 3 chronic kidney disease (CKD) or higher. AKI and CKD are encompassing terms for many etiologies and pathological processes that affect different regions of the kidney but converge at tissue inflammation and fibrosis. Macrophages perform vital homeostatic functions and can initiate or ameliorate inflammation and fibrosis.
