Anther Development And Cytochemistry In Asphodelus Aestivus (Asphodelaceae), Fi̇li̇z Vardar, Işil İsmai̇loğlu, Meral Ünal

Asphodelus aestivus Brot. (Asphodelaceae) anthers were analysed to provide a detailed understanding of the events that lead to pollen grain development, accompanied by cytochemical observations at different ontogenic stages. An anther locule of A. aestivus was bordered by 4 different layers: the tapetum, the middle layer, the endothecium, and the epidermis. At the tetrad stage, secretory tapetal cells enlarged maximally, while they showed degeneration at the young microspore stage. During the course of the degeneration, vacuolisation and reduction in the volume were conspicuous. Some species belonging to the genus Orchis Tourn. ex L. (Orchidaceae) are of great economic importance as their tubers or corms are used to produce a hot beverage called salep. Nevertheless, these plants are not cultivated but are rather collected from nature, and due to careless collection many have already been listed as endangered plants. The mature female gametophyte in Trifolium pratense L., a natural tetraploid plant, consists of 7 cells: the egg cell, 2 synergids, the central cell, and 3 antipodal cells. This type of mature female gametophyte is known as polygonum. The central cell occupies the largest portion of the mature female gametophyte and the polar nuclei are situated close to the egg apparatus. The cytoplasm of the central cell is rich in organelles. Sporobolus arabicus Boiss. is a perennial stoloniferous grass that prefers a saline or desert habitat. A natural population of Sporobolus arabicus growing in a salty area was compared with a normal nonsaline population. The salt-tolerant population showed better growth than its counterpart. Physioanatomical adaptations to saline environments in the salt-tolerant population were very specific. They included restricted toxic ion uptake, production of organic osmolytes, sclerification in roots, succulence in stem, and development of vesicular hairs on the leaves. Cistanche phelypaea (L.) Cout. (Orobancaceae) was found parasitising the roots of the succulent shrublets Arthrocnemum macrostachyum (Moric.) K.Koch (Chenopodiaceae) in a coastal salt marsh in Qatar. Measurements were conducted to identify soil properties, host, and noninfected plants by soil excavations to expose the haustoria of the parasite attached to the host roots. The water potential, osmotic potential, pressure potential, and chemical analyses were determined in parasite, host, and noninfected plants. Cadmium is one of the most toxic heavy metals and affects all viable cells, even at low concentrations. It is introduced to agricultural soils mainly by phosphate fertilisers and causes many toxic symptoms in cells. Phytochelatins (PCs) are heavy metal-binding peptides that play an important role in sequestration and detoxification of heavy metals in plants. Callogenesis using vegetative (leaf, stem, petiole) and floral (petal, pistil, anther) explants of Rosa gallica and R. hybrida was investigated using different combinations of 2,4-dichlorophenoxyacetic acid, 6-benzylaminopurine, and gibberellic acid on modified Murashige and Skoog medium. The highest callogenesis was obtained on Murashige and Skoog medium containing ratios of 2 and 3 mg L^{-1} of 2,4-dichlorophenoxyacetic acid to 1 mg L^{-1} of 6-benzylamino purine in both species depending on type of explant. A study was conducted to assess whether foliar-applied nitric oxide (NO) could alleviate the adverse effects of salt stress on wheat (Triticum aestivum L.). Four sodium nitroprusside levels (control [water spray] and 0.05, 0.10, and 0.15 mM) were sprayed as a donor of NO on the leaves of cultivar S-24 plants grown under nonsaline and saline conditions (150 mM NaCl). Data for growth and yield, chlorophyll contents, activities of antioxidants, and concentrations of mineral nutrients were recorded. The effect of water deficit on growth and some physiological and biochemical parameters related to water deficit tolerance was studied in 4 Moroccan alfalfa Medicago sativa L. populations that originated from the mountains and oases of Morocco. The experiment was conducted in greenhouse conditions. Seeds were allowed to germinate in pots filled with sand and peat at a 2:1 ratio, respectively. The transient stability constraint should be taken into consideration for the solution of the optimization problems in power systems. This paper presents a solution for the transient stability-constrained optimal power flow (TSCOPF) by a novel approach based on the artificial bee colony algorithm. The formulas of TSCOPF are derived through the addition of rotor angle inequality constraints into optimal power flow relationships. Purification And Biochemical Characterization Of An Extracellular Lipase From Psychrotolerant Pseudomonas Fluorescens Ke38, Aysun Adan Gökbulut, Alper Arslanoğlu

An extracellular lipase producing bacterium was isolated from a soil sample, and identified as a strain of Pseudomonas fluorescens by 16S rRNA gene sequencing. It was named Pseudomonas fluorescens KE38. KE38 showed psychrotolerant properties with an optimum growth temperature of 25 °C. The lipase enzyme secreted by KE38 was purified 41.13-fold with an overall yield of 54.99%, and a specific activity of 337.3 U/mg. The molecular mass of purified lipase was estimated to be approximately 43 kDa by SDS-PAGE. Yeast transformations with Brassica napus and Perilla frutescens \omega-3 desaturases were carried out. Feeding experiments on exogenous 18:2 at 10 mg/L and 100 mg/L for both genes under this investigation showed the same activity levels over the 3 days of the incubation, producing almost the same amount of 18:3. To further investigate the characteristics of B. napus and P. frutescens \omega -3 desaturases, a time-course in vivo activity study was conducted. Three different soybean [Glycine max (L.) Merr.] cultivars (A-3127, Derry, and Yemsoy) were evaluated for dry matter (DM) yield, plant components, and crop growth rate (CGR) at different row spacings and harvesting stages in 3 locations with Mediterranean-type climate in a split-split plot design with 3 replications in 2009 and 2010. In addition, crude protein (CP), crude protein yield (CPY), acid detergent fiber (ADF), neutral detergent fiber (NDF), total digestible nutrients (TDN), and relative feed value (RFV) of harvested forage soybeans were determined at the Bursa (Turkey) location. The purpose of this research was to develop an efficient machine vision-based experimental sorting system for tomatoes. To establish an efficient protocol for cell suspension culture and growth, the effects of different plant growth regulators (2,4-D, NAA, BAP, and kinetin) on callus induction and cell suspension culture of Iranian poppy were evaluated. Mutation breeding techniques in combination with tissue culture and molecular marker methods provide a powerful tool for improvement of vegetatively propagated plants. The aim of this study was to develop a protocol for shoot regeneration and mutation induction of Etlingera elatior. The results of irradiation on in vitro buds of E. elatior showed LD_{50} to be 10 Gy, with the survival of explants being sharply reduced at this dosage. Table grapes (Vitis vinifera) of the cultivar Shahroudi were coated with 0.5% or 1% chitosan and then stored at 0 °C for 60 days. Shelf-life qualitative traits including rachis and berry appearance and berry cracking, shattering, decay, browning, acceptability, and flavor were evaluated. Treated berries showed less weight loss, decay, browning, shattering, and cracking. Flavor-related factors such as total soluble solids (TSS), titratable acidity (TA), and TSS/TA ratio levels were higher in treated fruits; however, there was no significant difference between the fruits treated with 0.5% and 1% chitosan. Pine processionary moth, Thaumetopoea wilkinsoni Tams, is an important defoliating lepidopteran pest of pine trees. The aim of this study was to determine the required spore-crystal concentration of local Bacillus thuringiensis (Bt) isolates, optimal ambient temperature, and larval stage to control this troublesome forest pest. The susceptibility of T. wilkinsoni larvae decreased with older stage and lower temperatures. The optimum temperature was found to be 15 °C or higher for the control of early larval stages. The effects of Meloidogyne incognita, Xanthomonas campestris, and Rhizobium sp., alone or in combination, on the disease complex in chickpea were examined. Individual inoculation with M. incognita and X. campestris caused significant reductions in plant growth, while inoculation with Rhizobium sp. resulted in a significant increase in plant growth. Inoculation with M. incognita and X. campestris together caused a greater reduction in plant growth than the damage caused by each of them alone. The present study was done to select the potential autochthonous lactic acid bacteria (LAB) for the production of shalgam, which is a traditional Turkish lactic acid-fermented beverage. Eighteen LAB belonging to the genera Lactobacillus, Lactococcus, Pediococcus, and Leuconostoc isolated previously from shalgam samples produced in the university laboratory and by small- and large-scale producers in industry were used. Pasteurized black carrot juice was inoculated individually with these selected LAB strains and fermented for 10 days. In this study, the effects of the plant growth regulator trinexapac-ethyl (TE) combined with different sowing densities on field-grown rice (Oryza sativa L.) plant length, lodging, and grain yield were evaluated over 2 growing seasons.
