Identification And Functional Analysis Of Soybean Gmsbh1 Gene Promoter Conferring High Temperature- And Humidity-Induced Expression, Ming Chen, Peipei Qian, Zhankui Wang, Yingjie Shu, Yuan Tao, Liyan Huang, Yilong Wang, Haihong Zhao, Hao Ma

Turkish Journal of Botany

Soybean homeobox gene GmSBH1 has previously been proven to be involved in response to high temperature and humidity (HTH) stress. To investigate its expression patterns and active cis-elements, a 2040-bp 5'-upstream genomic DNA fragment of GmSBH1,named GmSBH1P, was isolated by PCR walking and characterized. Sequence analysis revealed that the fragment contains a series of cis-acting elements related to stress responses. The transient expression assay in the leaves of Nicotiana benthamiana and in the cotyledonary nodes of soybean indicated that the GmSBH1P strongly and rapidly mediates the induction of GUS expression under HTH stress. Deletion and mutation analysis of the promoter …

Two Pronounced Chlorella Strains For Efficient Biodiesel Feedstock Production, İlhami̇ Tüzün, Zeynep Eli̇bol Çakmak, Emmanuel Young, Turgay Çakmak

Turkish Journal of Botany

In this study, two indigenous Chlorella strains were evaluated for triacylglycerol (TAG) production and related parameters during 10 days of N-deprivation. The strains were identified based on morphological characteristics and genomic information, named as Chlorella sp. IMU12 and Chlorella sp. IMU17. Time-dependent reduction of growth was accompanied by decreased chlorophyll content, reduced oxygen evolution ratio, and elevated carotenoid content of both strains cultivated in N-free Bold's basal medium. Nitrogen deprivation induced total lipid and neutral lipid content in both strains, supported by FTIR measurement of TAGs. Chlorella sp. IMU17 showed higher production of TAGs as a response to N-deprivation. Strikingly, …

Morphological Characterization And Relationships Among Some Important Wild And Domestic Turkish Mustard Genotypes (Brassica Spp.), Fatma Kayaçeti̇n

Turkish Journal of Botany

Mustards occupy an important place among oilseed species due to their considerable economic and nutritional value and multiple uses. This study made use of 77 mustard genotypes collected from the USDA gene bank and diverse ecologies of Turkey to determine the extent of morphologic variability among them. The genotypes were screened to select genotypes appropriate for use in mustard breeding programs during 2015?16 and 2016?17 under the Mediterranean rainfed conditions of Yenimahalle (spring sowing and autumn sowing) and Haymana (spring sowing). Cluster analysis showed significant diversity among the genotypes. Promising genotypes with different genetic backgrounds for spring and autumn sowing …

Turkish Journal of Biology

The clinical use of cisplatin, which is a first-line anticancer agent, is highly restricted due to its adverse effects on kidneys that lead to nephrotoxicity. Therefore, some potential reno-protective substances have been used in combination with cisplatin to cope with nephrotoxicity. Due to its high antitumor activity and oxygen-carrying capacity, we investigated the molecular effects of squalene against cisplatin-induced oxidative stress and kidney damage in mice. Single dose of cisplatin (7 mg/kg) was given to male Balb/c mice. Squalene (100 mg/kg/day) was administered orogastrically to mice for 10 days. Following sacrification, molecular alterations were investigated as analysis of the levels …

Alfalfa is a fodder crop that accounts for one of the best sources of protein and is widely cultivated around the world. In vitro regeneration of alfalfa has been studied earlier; however, most of the studies were almost intervened with callus formation. In this study, 3 explant sources (cotyledonary node, hypocotyl, and root crown) of two Turkish cultivars (Nimet and Savaş) were excised from young seedlings. Explants were subjected to different concentrations of BAP, BAP-IBA, and TDZ to evaluate the direct in vitro regeneration potential of selected plant parts. Moreover, we transformed the alfalfa plant with pBin19 harboring 35s.GUS-INT_35s.nptII construct …

Turkish Journal of Agriculture and Forestry

In this study, the effects of cooking and the addition of extra virgin olive oil on the in vitro bioaccessibility of carotenoids in tomato sauce were evaluated. The results indicated that total antioxidant activity, α-tocopherol, lycopene, β-carotene, lutein, and chlorophyll a contents were positively affected by the combination of cooking and addition of extra virgin olive oil. Moreover, after in vitro digestion, for intestinal phase, lycopene and Β-carotene were observed in all samples, whereas α-tocopherol and chlorophyll a were not detected in any of the samples. Lycopene (≈8.5-fold) and β -carotene (≈5-fold) were significantly improved (P < 0.05) by the effect of cooking and addition of extra virgin olive oil to the samples compared to control tomato sauce (without oil and cooking).

Deciphering Global Dna Variations And Embryo Sac Fertility In Autotetraploid Rice Line, Fozia Ghouri, Jiani Zhu, Hang Yu, Jinwen Wu, Faheem Shahzad Baloch, Xiangdong Liu, Muhammad Qasim Shahid

Turkish Journal of Agriculture and Forestry

Autotetraploid rice is a new germplasm resource obtained by doubling chromosomes through colchicine treatment. There have been many studies on the reproductive characteristics of autotetraploid rice, but little is known about global DNA variations and reasons for low embryo sac fertility in autotetraploid rice. Therefore, an autotetraploid rice line (T446) was used for resequencing and embryo sac fertility was observed. Whole-genome resequencing data revealed 87,229 SNPs and 11,022 InDels in the genome of T446 versus E246 (diploid rice), which had an average of 23.37 SNPs and 2.95 InDels per 100 kb. A total of 17,375 and 17,171 structural variations and …

The Application Of Hemp (Cannabis Sativa L.) For A Green Economy: A Review, Tahseen Karche, Manager Rajdeo Singh

Turkish Journal of Botany

The concern for the issues related to the growing needs of human civilization like resource depletion, pollution, climate changes, and health risk has led to a search for eco-friendly alternatives to environmental problems. Recently, hemp started gaining popularity for its medicinal, nontoxic, greenhouse negative, and biodegradable properties. Originating from the steppes of Central Asia, Cannabis sativa L. (hemp) is one of the oldest domesticated plants known to humans. Since 5000-4000 BC, different parts of the plant were used for spinning, weaving, papermaking, the seed for human feed, animal feeds, medicinal, and health purposes. Reports also suggest the use of hemp …

The footprints in this study, which were considered to have been made by ungulates, were discovered on mudstone that was approximately 5 cm thick, near an abundant mud-cracked plane located on the stratigraphic subsurface of the late Oligocene Karayün Formation, which crops out over wide areas in southern Sivas (Turkey) and has terrestrial, fluvial sediment characteristics. These ungulate footprints documented from the late Oligocene of the Karayün Formation in southern Sivas represent the first reported vertebrates in Anatolia. The footprints of three different species of ungulates were identified. The shapes, depth, and widths of the footprints provided some basic ichnotaxonomic …
