Effects Of Leptin And Thyroglobulin Gene Polymorphisms On Beef Quality In Holsteinbreed Bulls In Turkey, Süleyman Kök, Güldan Vapur
Turkish Journal of Veterinary & Animal Sciences
The relationship between three single-nucleotide polymorphisms (SNP) markers and the beef quality of Turkish Holstein bulls (THB) was investigated. The markers in leptin (LEP) gene (E2JW and E2FB) and thyroglobulin (TG) gene (C422T) were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) in one hundred THB. Texture, pH, and marbling score (MS) are three of the most dominating determinants of meat quality. In THB, the relationship between 3 SNPs and beef quality characteristics of musculus longissimus dorsi (MLD) was investigated. The mean pH value of the raw beef samples with the TG G422T CC genotype was 5.33 ± 0.30 …

This study was designed to determine carnitine concentrations in newborn calves with suspected septicaemia, healthy calves, and colostrum samples as well as to detect alterations in blood carnitine concentrations after colostrum intake. Carnitine concentrations were detected in both neonatal calves with suspected septicaemia (n = 195) and healthy calves (n =10) as well as in colostrum/milk samples from their dams (n = 20). The mean carnitine concentration on the 2nd day after colostrum intake (32.61 μmol/L) was significantly higher than that measured before colostrum intake (17.61 μmol/L) (P < 0.05). The striking result was significantly lower mean carnitine concentration detected in calves with suspected septicaemia (17.11 μmol/L), compared to healthy calves (24.92 μmol/L) (P < 0.001). Apart from a sudden increase on day 7, no significant alteration was observed in colostrum/milk carnitine concentrations throughout the postpartum period. The results indicated that colostrum contained carnitine, which passed into the bloodstream of the calf by passive colostral transfer, and that a dramatic reduction in blood carnitine concentrations of calves with suspected septicaemia existed. The results might be of help in dealing with septicaemia suspected neonatal calves through both measurement of blood carnitine and, in response, to supplement such cases with carnitine.

Turkish Journal of Veterinary & Animal Sciences
It was aimed to monitor postpartum (pp) uterine involution and follicular wave pattern by transrectal ultrasonography and detection of concentrations of milk progesterone (P4) in eleven water buffaloes during the summer season in Turkey. The data obtained from the first day of calving to positive pregnancy diagnosis were evaluated on a daily basis. Uterine position scores and nonechogenic material showed a decreasing grade pattern related to the time (P < 0.0001). A time effect was evident with uterine horn thickness (P < 0.05) or intercaruncular zone thickness (P < 0.01) and uterine horn lumen (P < 0.01). Uterine horn thickness showed a gravidity x time interaction (P < 0.05). Gravidity x time interaction was not evident in total uterine horn thickness but time effect was seen (P < 0.0001). Time effect was also evident for the thickness of uterine body (P < 0.0001) and cervix uteri (P < 0.01). The uterine body and horns were returned to previously nongravid state on day 26.00 ± 2.38 and the interval from calving to complete disappearance of uterine fluid was accomplished on day 31.44 ± 3.23 pp. The previously gravid and nongravid uterine horns, uterine body and cervix uteri involuted on day 31.42 ± 1.39, 28.37 ± 0.65, 29.37 ± 0.73 and 27.87 ± 0.47 pp, respectively. The interval from calving to first observation of dominant follicle of nonovulatory wave was 18.54 ± 6.91 days and on average 3.45 ± 1.91 nonovulatory waves were observed. Postpartum first ovulation occurred on day 64.45 ± 18.49. One and two wave models or combination of both waves were observed. The interval from calving to pregnancy was 72.36 ± 21.33 days. The maximum concentrations of milk P$_{4}$ during short or normal cycles and pregnancy were 0.62 ng/mL, 2.2 ng/mL, and 4.04 ng/mL, respectively. In conclusion, the presence of one or two waves should be considered when estrous synchronization programmes are planned in Anatolian water buffaloes during summer season.

Turkish Journal of Veterinary & Animal Sciences
This research examined the effects of liquid feeds on calf performance, rumen development, rumen cellulolytic bacteria count, and total bacteria diversity. Twenty Holstein bull calves were allocated to 2 groups either fed with milk replacer (MR) or whole milk. Calves fed milk tended to have higher (P = 0.058) average daily weight gain as compared to calves fed with MR during the entire experimental period. Starter mixture intake was not influenced by either milk or milk replacer feeding. Calves fed with milk had higher (P < 0.001) proliferative cells in the rumen papilla. The expressions of MCT1, MCT4, NHE1, NHE3, ACSS1, AceCS2, and ACSM1 genes, which are related to volatile fatty acid absorption, in the dorsal sac of the rumen were similar for calves fed either MR or whole milk. Fibrobacter succinogenes (P = 0.017) and Ruminococcus flavefaciens (P = 0.037) were significantly higher in the rumen of the milkfed calves. Moreover, denaturing gradient gel electrophoresis (DGGE)-derived dendrograms showed noticeable differences in ruminal microbial population between MR-fed and milk-fed calves. In conclusion, feeding calves with whole milk, which contains better quality protein and nutrients, instead of milk replacers, positively affects rumen development, cellulolytic bacteria numbers and total bacteria diversity.

Toxicity Remission Of Paes On Multireceptors After Molecular Modification Through A3d-Qsar Pharmacophore Model Coupled With A Gray Interconnect Degree Method, Xinyi Chen, Yu Li
Turkish Journal of Chemistry
In the proposed model, the gray interconnect degree method was employed to process the acute toxicity values of phthalate acid esters (PAEs) to green algae, daphnia, mysid, and fish (predicted by EPI Suite software) and to obtain the comprehensive characterization value of the multireceptor toxicity effect (MTE) of PAEs. The 3D-QSAR pharmacophore model indicated that hydrophobic groups significantly affected the MTE of PAEs. Based on this, 16 PAEs derivative molecules with significantly decreased comprehensive characterization value (more than 10%) of the toxic effects of multireceptors were designed. Among them, 13 PAEs derivative molecules reduced the toxicity values (predicted by the …

Regarding the abundant use of metals for different purposes, it becomes more critical from various scientific and technologi¬cal perspectives to discover novel agents as selective probes for the detection of specific metals. In our previous studies, we have shown that aqueous solutions of natural urolithins (i.e., hydroxyl-substituted benzo[c]chromen-6-one derivatives) are selective Iron (III) sen¬sors in fluorescence assays. In this study, we have extrapolated these findings to another coumarine compound (i.e., 3-Hydroxy-7,8,9,10- tetrahydro-6H-benzo[c]chromen-6-one) and compared the selective metal binding properties with Urolithin B (i.e., 3-Hydroxy-6H-benzo[c]chromen-6-one). Following the synthesis and structure identification studies, the fluorometric studies pointed out that the lactam group …

The phytochemical analyses were conducted with gas chromatographymass spectrometry/flame ionisation detector (GC-MS/FID) and liquid chromatography-mass spectromtetry (LC-MS/MS) techniques. The extracts and essential oil were studied for α-amylase and acetylcholinesterase activities with two different spectrophotometric methods. Antimicrobial activities of the extracts were investigated by microdilution. The extracts were evaluated in vitro for cytotoxic effects against cancer and normal cell lines by MTT assay. The essential oil (EO) contained α-pinene (12.5%) and β-caryophyllene …

Heavy metals, due to their inability to degrade, pose a serious environmental and nutritional problem. The accumulation of essential and non-essential heavy metals in living organisms reduces normal growth and development, resulting in acute poisoning, disease and even death of organisms. Melatonin is a very important multifunctional molecule in protecting plants from oxidative stress due to its ability to directly neutralize reactive oxygen species (ROS). Also, melatonin has a chelating property, which may contribute in reducing metal-induced toxicity. In this paper, the protective role of melatonin in counteracting metal-induced free radical generation was highlighted. Using the HPLC-FLD technique melatonin was …

Resistance to therapeutic agents and the highly toxic side effects of synthetic drugs has spurred new research in the treatment of colon cancer, which has high morbidity and mortality ratios. This study aims to clarify the molecular mechanisms of the anticarcinogenic properties of methanol extract of Viburnum opulus L. (EVO)and its main active compound, trans-p-coumaric acid (p-CA), on human colon cancer cells (DLD-1, HT-29, SW-620, Caco-2) and healthy colon epithelial cells (CCD-18Co). The effects of EVO on controlled cell death (apoptosis) and the cell division cycle were determined by flow cytometry. Alteration in mRNA and protein expressions of switch genes …

Colorectal cancer is one of the most common types of cancer. Drug resistance and drug-induced damage of healthy tissues are major obstacles in cancer treatment. Therefore, to develop efficient anticancer therapy, it is necessary to find compounds that affect tumor cells, but do not exhibit toxicity to healthy cells. Caffeic acid phenethyl ester (CAPE) has been demonstrated to have anticancer properties in many types of cancer. In this study, the cytotoxic and apoptotic effects of CAPE on the RKO colorectal cancer cell line and CCD 841-CoN normal colorectal cell line was investigated. In addition, changes in the survivin expression were …

Recent studies have indicated that polysaccharides, the main component of the Lycium barbarum L. fruit, have beneficial effects (e.g., anxiolytic, antioxidant, and neuroprotective) on humans and rodents. However, the effects of different dosages of such polysaccharides on ovariectomized rats and their underlying mechanisms in the brain have not been evaluated in the literature. Here, we aimed to evaluate the effects of the high and low doses of polysaccharides obtained from Lycium barbarum fruits (HD-LBP and LD-LBP, respectively) on anxious behaviors via behavioral (using the OFT and EPM), biochemical (using ELISA), and immunohistochemical (using immunohistochemical staining) measures in detail. Two weeks …

COVID-19, a novel infectious disease, caused by SARS-CoV-2, affected millions of people around the world with a high mortality rate. Although SARS-CoV-2 mainly causes lung infection, gastrointestinal symptoms described in COVID-19 patients and detection of the viral RNA in feces of infected patients drove attentions to a possible fecal-oral transmission route of SARS-CoV-2. However, not only the viral RNA but also the infectious viral particles are required for the viral infection and no proof has been demonstrated the transmission of the infectious virus particles via the fecal-oral route yet. Growing evidence indicates the crosstalk between gut microbiota and lung, that …
