Application Of Green Fluorescent Protein As A Marker For Selection Of Transgenic Mouse Embryos Before Implantation, Haydar Bağiş, Levent Keski̇ntepe

In this study, we evaluated the application of green fluorescent protein (GFP) from the jellyfish Aequorea victoria, for efficient selection of possible transgenic and non-mosaic mouse embryos after microinjection. We injected 353 one-cell mouse zygotes with a DNA fragment carrying Gfp gene under the control of \beta-actin gene promoter. Eighty-seven per cent (307 embryos) of the injected embryos survived after microinjection. The surviving embryos were cultured for an additional day and then analysed for detection of GFP expression using a standard fluorescein isothiocyanate (FITC) filter set attached to a microscope. Nitric oxide synthase (NOS) that uses NADPH as a cofactor is an enzyme which produces nitric oxide (NO) from L-arginine. Endothelial-derived (eNOS), inducible (iNOS) and neuronal (nNOS) nitric oxide synthase are 3 known isoforms. Endothelial NO regulates the vascular tonus and causes the dilatation of vessels, while NOS localized in the sarcolemma of striated muscle plays a role in the regulation of muscle hemodynamics. Many authors have found differences in NO production in some pathological conditions such as diabetes mellitus (DM), hypertension and atherosclerosis. We examined the effects of daily melatonin (Mel) infusions in pinealectomized prepubertal male Siberian hamsters in three different conditions. In one study we investigated the body weight maturation response to one hour daily infusions of 10 ng, 25 ng, or 50 ng of Mel in pinealectomized hamsters. Animals received, at day 15 of life, programmed subcutaneous infusions of Mel or vehicle at one of five time points (1900-2000, 2000-2100, 2100-2200, 2400-0100, and 0300-0400 hours) for 30 days. In long-day-born (16L) animals, Mel infusion right after lights off (2000-2100 hours) significantly inhibited body growth; this dose was ineffective at other times. …

Diabetes causes metabolic defects and related vascular alterations such as atherosclerosis. These vascular alterations may also reflect the renal structural degeneration under the influence of diabetes. Genetic alterations in proto-oncogenes or tumour suppressor genes are believed to be one of the key events in the multistage process of carcinogenesis. Activating point mutations occurring in either one of the three ras proto-oncogene families are common genetic alterations in human and animal neoplasms. However, the mechanisms leading to oral cancer are not completely understood. This study was carried out to evaluate the effects of ration supplemented with zinc on the electrocardiograms of 60 broiler chicks. The chicks were divided into four groups,one control and three experimental groups (II,III,IV). Zinc was given to the experimental groups at doses of 125, 500, 1000 mk/kg respectively, in their ration for 60 days.The electrocardiogram recordings were performed on chicks on the 60 th day of the experiment. The Development Of European Sea Bass (Dicentrarchus Labrax L., 1758) Eggs In Relation To Temperature, Şahi̇n Saka, Kürşat Firat, H. Okan Kamaci

In this study, the development of European Sea Bass (Dicentrarchus labrax L., 1758) eggs was investigated at different temperatures. The experiments were carried out at 15ºC and 17ºC. The average diameters of the eggs were determined tobe approximately 1.162±0.004 mm. The embryonic development of the eggs was observed every hour and then photographed. The hatching rates were found to be 84.8-89.4% at 15ºC and 82.3-86.3% at 17ºC, respectively. Hatching of the larvae took 87 hours at 15ºC and 69 hours at 17ºC.

A total of 200 wild pigeons, 82 young and 118 adults, captured from 12 localities in Ankara District, were examined for blood parasites. Haemoproteus columbae was found in 57% (114/200) of the pigeons examined, and no other blood parasites were detected.

In this research, some physical characteristics and uses of the coarse fibers obtained from hair goats raised in some provinces of Turkey were examined. For the general provinces, the diameter of fiber and natural length of fiber were 92.1±0.56 µ and 11.6±1.37 cm resepctively. According to the findings, the coarse fibres of hair goats can be used in products such as tents, stiffening cloth, card weaving, floor weaving (çul ve cicim), saddle-bags, bags, sacks, animal feeding bags and socks.

Genetic determinants of lactose fermentation, proteolytic activity and bacteriocin production in the strains L. lactis subsp. lactis MPL114, L. lactis subsp. cremoris MPC112 and L. lactis subsp. lactis biovar. diacetylactis MPD166 were studied. As a result of mutation and genetic transfer studies, it was determined that all of these characters were encoded by the 65.4 kb plasmid in L. lactis subsp. cremoris MPC112. Lactose fermentation and proteolytic activity were encoded by the 68.0 kb plasmid and bacteriocin production was encoded by the 12.4 kb plasmid in L. lactis subsp. lactis MPL114. In this study, congenital malformations are described in one calf. In the calf, accessory tongue, which is a rare condition, was determined together with palatoschisis and growth malformations of cranial bones. The accessory tongue observed in this study is the first case described in calves.

This research was carried out with the aim of improving the conservation and feed value characteristics of citrus pulps. In order to ensure and improve citrus pulp conservation even for short periods (1 week), several methods intended to inhibit yeast activity were examined. Orange and grapefruit pulps were used as feed materials and three heads of merino male yearlings fitted with rumen cannula were used as animal materials. Candida utilis NRRL Y-900 was grown on horn hydrolysate for single-cell protein production. First, ram horns obtained from slaughterhouse in Erzurum were hydrolyzed by physical and chemical methods and crude horn hydrolysate (CHH) was obtained. The European pine sawfly, Neodiprion sertifer (Geoffr.) (Hymenoptera: Diprionidae), is a serious pest of pine plantations and ornamentals. Chemical pesticides used to control this pest have a detrimental effect on the environment. The Neodiprion sertifer nuclear polyhedrosis virus (NsNPV) frequently causes considerable decreases in the populations but doses not harm other animals or plants. In this study, we tested five different concentrations of NsNPV against the larvae of N. sertifer and showed viral infection using Giemsa staining.
