The aim of this study was to prepare a supermacroporous cryogel that can be used for the adsorption of cellulase. The macroporous cryogel of poly(2-hydroxyethyl methacrylate-N-methacryloyl-l-phenylalanine) [p(HEMA-MAPA)] was prepared by copolymerization of 2-hydroxyethyl methacrylate (HEMA) with a functional monomer of N-methacryloyl-l-phenylalanine (MAPA). The cryogel was characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, and swelling tests. The effects of several parameters such as medium pH, temperature, ionic strength, and flow rate on cellulase adsorption were also investigated. Cr(III) is considered as essential; however, the Cr(VI) form is classified as carcinogenic.~For this reason, speciation analysis in food samples is a very important question. A new N-carboxamide compound (3) was synthesized by the reaction of dibenzoylaceticacid-$N$-carboxyethyla\-mide (1) and 3,4-dichloroaniline (2). The $N$-(3,4-dichlorophenyl)-3-oxo-3-phenyl-2-(phenylcarbonyl) propanamide (3) subsequently reacted with Cu salt to produce its Cu(II) complex compound (4). The compounds were characterized by analytical and spectral methods. In addition, X-ray diffraction was performed to characterize and obtain detailed information about the structure of 3. The fully optimized geometries of compounds 3 and 4 were calculated at different basis sets by using the Gaussian09 (G09) software to investigate their 3D geometries and electronic structures. L-cysteine-capped Mn-doped ZnS quantum dots (QDs) were used for the determination of glutamic acid in foodstuffs. This method is based on measurement of the quenching of the phosphorescence intensity of the QDs after interacting with glutamic acid. A linear response was observed from 50 to 500 ng mL$^{-1}$ glutamic acid with a limit of detection of 6.79 ng mL$^{-1}$. Room temperature phosphorescence (RTP) intensity of the QDs was quenched rapidly upon the addition of the quencher and the reaction reached equilibrium within 2 min. Production, Purification, And Characterization Of A Thermo-Alkali Stable And Metal-Tolerant Carboxymethylcellulase From Newly Isolated Bacillus Methylotrophicus Y37, Yonca Duman, Yonca Yüzügüllü Karakuş, Arzu Sertel, Fi̇kri̇ye Polat

A carboxymethylcellulose (CMC)-degrading bacterium was isolated from soil, identified as Bacillus methylotrophicus according to the physiological properties and analyses of 16S rRNA and a partial sequence of the gyrase A (gyrA) gene, and named as B. methylotrophicus Y37. The CMCase enzyme was purified to homogeneity by 20.4-fold with 21.73% recovery using single-step hydrophobic interaction chromatography and biochemically characterized. CMCase showed a molecular weight of approximately 50 kDa as determined by SDS-PAGE. The activity profile of the CMCase enzyme exhibited optimum activity at 45 $^{\circ}$C and pH 5.0. Two new solid phase extraction methods have been developed for simultaneous separation and preconcentration of Cu(II) and Cd(II) in some food and environmental samples before their determination by flame atomic absorption spectrometry (FAAS). One of these methods is based on the absorption of the Cu(II) and Cd(II) complexes with [3-(cyanomethyl)-5-(4-methylphenyl)-4$H$-1,2,4-triazol-4-yl]carbamate (L1) and the other is based on the absorption of the Cu(II) and Cd(II) complexes with 4-(4-methylphenyl)-5-{[pyridine-4-yl-4$H$-1,2,4-triazole-3-yl)thio]methyl}-4$H$-1,2,4-triazole-3-thiol (L2) on Amberlite XAD-1180 resin. The sirtuin (SIRT) gene family is reported to regulate critical intracellular processes from aging to cellular metabolism and repair. SIRT3 knockout (SIRT3-/-) mice develop receptor positive mammary tumors starting at 13 months and SIRT3 expression is decreased in human breast cancer samples as well as several other diseases. It is established that carcinogenesis results from the accumulation of multiple aberrant genetic events including the activation of oncogenes and inactivation of tumor-suppressor genes. Cyanobacteria can be used to remove nitrogenous compounds from wastewater, but a major bottleneck in the process is the separation of cyanobacterial biomass from the treated water discharge, which may cause eutrophication. The current study assessed the suitability of three periphytic cyanobacteria (Geitlerinema sp., Gloeotrichia sp., and Lyngbya sp.) isolated from shrimp ponds. These cyanobacteria were immobilized by self-adhesion to polyvinyl chloride sheets, forming mats, and were screened for their efficacy to reduce nitrogenous compounds and phosphorus. Secondary dental caries are one of the major reasons for restoration replacements. Incorporating antimicrobial properties into dental materials would limit the initiation and progression of dental caries. In the current study, dental composites having 1%, 5%, and 10% (w/w) sodium pentaborate pentahydrate were prepared and analyzed for their mechanical properties, degree of monomer conversion (DC) rate, antibacterial effects against Streptococcus mutans, and biocompatibility with human dental pulp stem cells (hDPSCs). Incorporation of boron into the composites significantly decreased flexural strength and DC in a dose-dependent manner, but the value for 1% boron-containing composite still remained within acceptable levels. Reactive oxygen species (ROS) overproduction may severely affect cell functions and even provoke cell death. Diazoxide, an opener of ATP-sensitive potassium channels, induces pharmacological preconditioning in different types of cells. However, the target of action of diazoxide is not clear enough because this substance can activate ATP-sensitive potassium channels localized in mitochondria (mitoKATP) as well as in plasma membranes (KATP channels). In this experiment, the ultrastructural effects of orally administered lead acetate on rat (Rattus rattus) spleen were studied. Rats weighing approximately 200-250 g were used in this experiment. They were divided into five groups, each one including five rats. Group I served as the normal control group while the others served as the experimental groups. An intracellular nitrilase (Nit1) with cyanide-degrading activity was isolated from Trichoderma harzianum VSL291, cultivated on benzonitrile as the sole carbon source. Nit1 was purified to homogeneity by ion exchange and gel filtration chromatography with a recovery of 7.15% and a fold of 22.5. The molecular weight was estimated to be 47.7 kDa and the purified enzyme was sequenced with a system of liquid chromatography and mass spectrometry (LC-MS). The enzyme consists of 436 amino acids with a predicted molecular weight of 47.088 kDa. This study investigates the mechanism of action of DNA insecticides, a novel preparation against gypsy moth (Lymantria dispar) based on the DNA fragments of the antiapoptotic gene of its nuclear polyhedrosis virus. In our experiments gypsy moth showed significant susceptibility to applied DNA insecticides on the basis of BIR and RING domains fragments of LdMNPV IAP-3, whereas common fruit fly and tobacco hornworm did not. The aim of the current study was to evaluate lead accumulation in the ovaries, fallopian tubes, and uterus and to take note of any consequent histo-architectural changes. The experiment involved a 12-month chronic exposure of 28 Wistar female rats at sexual maturity (221 ± 0.88 g/individual) to lead acetate in drinking water. A gene encoding ß-mannanase from Aspergillus niger GIM3.452 was amplified and inserted into a pPIC9K vector. The resulting recombinant plasmid, pPIC9K-MAN, was transformed into Pichia pastoris GS115. One strain (GSKM-1) having the highest ß-mannanase activity of 26.6 U/mL was obtained. In order to increase the secretion of ß-mannanase in P. pastoris, we constructed a double recombinant yeast and made it coexpress protein disulfide isomerase. One strain (GSKZ\alphaM2) with the highest ß-mannanase activity of 40 U/mL was then obtained and used to optimize expression conditions. Our earlier studies found that in vitro estradiol modulates mitochondrial Ca2+ transport in discrete brain regions. The present study examined the role of estradiol receptors (ERs) in estradiol-induced inhibition of Ca^{2+} efflux from synaptosomal mitochondria isolated from rat caudate nuclei and brain stems. Radioactively labeled CaCl_2 (0.6?0.75 µCi ^45CaCl_{2}) was used for Ca^{2+} transport monitoring. In the present study we have introduced a proteomic workflow, 2-dimensional electrophoresis coupled with matrix-assisted laser desorption-ionization time-of-flight mass spectrometry (MALDI-TOF MS) to create a 2-D map reflecting the characteristic pattern of serum proteins in growing piglets. Blood samples were collected from 6 piglets aged 50 days. Serum proteins were resolved at pH 3-10, followed by 12% SDS-PAGE. Reproducible protein spots (183) were picked from the gels and submitted for identification by peptide mass fingerprinting. Sine oculis homeobox 1 (Six1), a member of the Six homeoproteins, plays an important role in skeletal myogenesis and the specification of myofiber diversity. In this study, in order to scale up the production of recombinant porcine Six1 (pSix1), a pET-30a(+)-pSix1 plasmid was constructed and transformed into Escherichia coli BL21 (DE3). A specimen of the serpent eel, Ophisurus serpens, with a total length of 1917 mm was caught on 2 March 2014 by an angler on a boat off the coast of Karaburun, in front of the Sarpıncık lighthouse. This specimen is the second record for the central Aegean Sea since that of Aliağa, Çandarlı Bay, in August 1966.

The encyrtid species Copidosoma boucheanum Ratzeburg (Hym.: Chalcidoidea, Encyrtidae) was identified and is reported for the first time from Iran. These specimens were reared from leafroller larvae (Lep.: Tortricidae) on Populus sp. as a polyembryonic endoparasitoid. Copidosoma species of Iran are listed with their distribution.

Both sexes of Diarthrodella ergeneae sp. nov. (Copepoda, Harpacticoida, Paramesochridae), which were collected from the intertidal zone of Kurtpınarı Beach along the Mediterranean coast of Turkey, are described in detail. The new species is morphologically most similar to D. convexa Kunz, 1981; D. secunda secunda Kunz, 1954; and D. lancifera Kunz, 1981 according to the setal formula of the swimming legs, but D. ergeneae sp. nov. can easily be differentiated from D. convexa by the ratio of P1 exopod/enp-1, by the lack of an inner seta on P2 enp-2, and by the shape and armature of P5; from D. secunda …

This study was carried out on the genus Glyptholaspis in Kelkit Valley, Turkey, during 2007-2012. Three species, G. americana (Berlese, 1888), G. confusa (Foa, 1900), and G. saprophila Masan, 2003, were recorded from Kelkit Valley. G. americana has been previously recorded from Turkey; G. confusa and G. saprophila are first records for the Turkish fauna. The male of G. saprophila is described for the first time in this study. Original drawings for all known species of the genus from Kelkit Valley are included. Additionally, an identification key to the Turkish species of Glyptholaspis is given here.
