The aim of the present study is to provide a strategy for predicting the production of antigenic regions of Clostridium novyi alpha toxin. The selection is based on B-cell epitopes and MHCII binding protein by immunoinformatics tools. The study resulted in identifying antigenic regions in the beginning and middle (3-17 and 965-997 amino acid residues) as suitable binders to MHCII and the carboxyl terminal of the protein (1800-1958 amino acid residues) in B-cell epitopes. Fructose ingestion is often associated with hepatic steatosis and hypertriglyceridemia. The homeostasis of hepatic lipids is mainly determined by the interplay of lipogenesis and fatty acid β-oxidation. In this study, we hypothesized that high fructose intake disturbs hepatic lipid metabolism through an imbalance between these processes. Therefore, we analyzed the effects of a 9-week-long consumption of a 60% fructose solution on physiological parameters, glycemia, and blood lipid profiles in male Wistar rats. The aim of this study was to fabricate and characterize acellular porcine pericardium that can be used to patch cardiovascular repair. Porcine pericardial tissues were treated with 10 mM Tris-HCl and sodium dodecyl sulfate (SDS) at different concentrations and time intervals. The optimal decellularization protocol was determined according to elimination of cellular components and DNA via histology and DNA quantification, respectively. There were no significant changes in stress and fracture strain after decellularization. Liquid extracts from the decellularized pericardium caused no cytotoxicity towards human fibroblasts, were capable of supporting an appropriate attachment of human endothelial progenitor cells, and did not …

We have previously shown that DNA damage induced by doxorubicin was associated with an increase in the level of DNA methylation, 5meC, in mouse embryonic fibroblasts using a developed immunostaining methodology. Doxorubicin (also known as adriamycin), which induces double-strand DNA breaks, has been widely used in cancer therapy. Cytosine modifications are dynamically regulated in the genome during lifespan, but abnormal changes in the 5meC pattern can be associated with diseases in particular cancer progressions and DNA damage. Turkish Journal of Biology

Hybrid materials based on tetraethyl orthosilicate (TEOS), hydroxypropyl cellulose (HPC), or hydroxypropyl methyl cellulose (HPMC) with silver nanoparticles were synthesized. They were analyzed and characterized using differential thermal analysis and thermogravimetry, atomic force microscopy, and static contact angle measurements. It was experimentally demonstrated that the silver-doped hybrid materials have pronounced antibacterial behavior by studying the reduction of Pseudomonas aeruginosa PAO1 biofilm formation on the tested materials. The results revealed biofilm reduction of 35.7% and 30% by SiO2/HPC/2.5% Ag and SiO2/HPMC/2.5% Ag hybrid materials, respectively, compared to the control. Galega officinalis L. is a medicinal plant being used as a galactagogue and an antidiabetic in folk medicine. A new effective micropropagation protocol was established from leaf, stem, node, and root explants of G. officinalis for in vitro plant regeneration and callus induction systems. Although interspecific chimeras can be identified using laborious techniques, more accurate and rapid detectable methods need to be established. The present study develops a nested polymerase chain reaction (nPCR) assay to detect human-mouse chimeras. A set of previously validated outer primers, specific to the human SRY gene, was used for conventional one-step PCR, while the inner primers for nPCR were designed. The aim of this research was to investigate the effects of the methanol extract obtained from Lycium barbarum fruits on anxiety, depression-like behaviors, and spatial memory in Wistar albino rats. A total of 28 rats were selected, randomly assigned to four experimental groups, and tested by means of the open field and elevated plus maze tests for anxiety-like behaviors, the forced swim test for depression-like behaviors, and the Morris water maze test for spatial memory. We investigated the implication of AMP-activated protein kinase (AMPK) in melatonin-induced protection in a renal ischemia/reperfusion (I/R) model. Animals were divided into four groups: rats of the sham group were not subjected to renal I/R. Rats in the I/R group received the vehicle 30 min before renal ischemia for 1 h followed by reperfusion for 6 h. Rats in the Mel+I/R group received melatonin (10 mg/kg) 30 min before the onset of I/R. The present study addressed the successful in vitro seed germination, bulblet regeneration, increased bulblet size, hardening, and acclimatization of Persian lily (Fritillaria persica). Seed germination rate was recorded as 86.7% and 96.7% after 2 and 3 months of cold treatment at 4 °C, respectively. Bulblet explants taken from a germination experiment were cultured on Murashige and Skoog (MS) medium supplemented with 0.5-2.0 mg/L thidiazuron (TDZ). This study examined the effect of yohimbine alone and in combination with glibenclamide on ischemia and reperfusion-induced arrhythmias in diabetes. Six minutes of ischemia were produced in 1-week diabetic rats by ligation of the left coronary artery, and 6 min of reperfusion were produced by releasing the artery. A dose of 5 mg/kg of yohimbine and glibenclamide was administered for 7 days prior to the coronary ligation. Boron, which is mostly found in the form of boric acid (BA) or sodium borate (NaB), is known to bind NAD+. Sox6, a member of the Sox (Sry-related HMG box) family of transcription factors, plays an important role in embryonic muscle development and adult fast myofiber maintenance. Here we report the expression and purification of a His-tagged version of recombinant porcine Sox6 (pSox6) in Escherichia coli. The recombinant pSox6 was expressed as a C-terminal fusion protein with His6 tag in the E. Coli Rosetta (DE3) host strain. Findings in the last decade suggest that there is a considerable amount of cardiomyocyte turnover in the human heart throughout life, albeit not sufficient for heart regeneration following myocardial infarctions. Only a few species are known to be remarkably efficient in cardiac regeneration. They restore lost cardiomyocytes via a process of cardiomyocyte dedifferentiation, which is followed by robust proliferation of cardiomyocytes and incorporation into the myocardium. Mesenchymal stromal cells (MSCs) have been characterized as an important component of the hematopoietic niche that plays a role in regulating hematopoietic stem cell (HSC) quiescence, self-renewal, and lineage commitment. Macrophages, one kind of innate immune cells, are mainly developed from HSCs in niche. However, how MSCs and their subpopulations regulate macrophage commitment is still unknown. The current study compared the contribution of MSCs and Sca-1+MSCs to macrophage commitment and further modulation on the function of macrophages. We aimed to determine whether increased levels of asymmetric dimethylarginine (ADMA) directly cause hepatic deterioration and whether it is decreased by ascorbic acid (AA) as an antioxidant. The study included three groups: Control (n = 10), ADMA (n = 10), and ADMA + AA (n = 10). ADMA was administered at 2 mg kg-1 day-1 and AA was given at 50 mg kg-1 day-1 for 10 days. Polyhydroxybutyrate (PHB) is a polymer used to restore tissues or regenerate bones. However, its compatibility with human bone marrow-derived mesenchymal stem cells (MSCs) has not been examined. PHB membranes of random (r-PHB) or aligned (a-PHB) electrospun nanofibers that generate bone and spinal axon scaffolds were combined with human MSCs. The adhesion and proliferation of cells on these membranes were examined. The orientation of cells on PHB membranes was analyzed by confocal microscopy and scanning electron microscopy (SEM). Eichhornia crassipes is an abundant floating aquatic weed that has great potential for cadmium (Cd) remediation owing to its large biomass and relatively high tolerance and accumulation capabilities. This study was conducted with Eichhornia in 5, 10, 15, and 20 mg L-1 CdCl2 in a hydroponic system for 21 days, and the Cd concentrations in the roots, shoots, and leaves were estimated. The plant showed tolerance, but at high Cd concentrations declines in biomass, root length, and leaf area were observed.
