Although membrane proteins represent most therapeutically relevant drug targets, the availability of atomic resolution structures for this class of proteins has been limited. Structural characterization has been hampered by the biophysical nature of these polytopic transporters, receptors, and channels, and recent innovations to in vitro techniques aim to mitigate these challenges. One such class of membrane proteins, the ATP-binding cassette (ABC) superfamily, are broadly expressed throughout the human body, required for normal physiology and disease-causing when mutated, yet lacks sufficient structural representation in the Protein Data Bank. Objectives Sources of immunization data include state registries or immunization information systems (IIS), medical records, and surveys. Little is known about the quality of these data sources or the feasibility of using IIS data for research. We assessed the feasibility of collecting immunization information for a national children's health study by accessing existing IIS data and comparing the completeness of these data against medical record abstractions (MRA) and parent report. Staff time needed to obtain IIS and MRA data was assessed. Mechanical loading – or physical activity – is essential in the bone remodeling process as well as optimizing the densitometric and geometric properties of bone throughout the lifespan. Participation in sports is a common mode of physical activity that can enhance bone mass accrual at younger ages and facilitate bone mass maintenance at older ages. Research suggests that sport participation continued from adolescence into high school and college provides added benefits on aBMD and cortical bone measures and these benefits remain 10-15 years after retirement from sport. The ruminal microbiome allows ruminant animals to convert cellulosic biomass into food products. A majority of ruminal microorganisms remain uncharacterized due, in part, to the complexity of ruminal microbial communities. In order to gain further insight, selection-based batch culturing from bovine rumen fluid, in combination with metagenomics, was used to identify and characterize previously uncharacterized rumen bacteria capable of metabolizing cellulose, which was supplemented as a purified substrate. 16S rRNA-based population analysis was used to identify rumen bacteria enriched within 14 days of culturing. Developing perennial grain crops is an effective and crucial way to prevent soil erosion caused by conventional agriculture of using annual crops while meeting the increasing need of global food demand. We hypothesized that the regrowth in Zea might be controlled by two dominant complementary genes. F1 hybrids were created by crossing Zea diploperennis Iltis, Doebley & R. Guzman with annual Zea mays L. ssp. mays inbred line B73. A Total of 134 F2 plants derived from nine F1 were planted and phenotyped. From 2015-2017, we evaluated a newly established bighorn sheep (Ovis canadensis) herd in the Deadwood Region of the Black Hills, South Dakota. Our objectives were to 1a) determine annual survival rates, 1b) determine cause-specific mortality, 1c) estimate population size, 2a) assess genetic diversity, 2b) assess disease prevalence, 3) evaluate movement patterns post-release, 4a) evaluate 3rd-order habitat selection, and 4b) estimate herbaceous biomass at foraging sites post-release of translocated bighorn sheep. Profile by Sanford is a membership based weight loss program that helps its members make lifestyle changes with diet, exercise, and one-on-one interactions with a weight loss coach. Discovery of characteristics and behaviors influencing weight loss will benefit current and future members of Profile. This research utilizes massive data from Profile by Sanford to analyze member behavior. Fourteen data sets are evaluated, some containing millions of observations. All data is combined into one comprehensive table of 33,487 members. Members of Profile by Sanford are 77% female and two-thirds of all members start the program classified as obese. Main limitations of dried distillers’ grains with solubles (DDGS) as meal for monogastric animals is the presence of high fiber content, reduced fat content, and poor amino acid balance. This causes a reduced economic value of DDGS for these species due to its low inclusion rates. The goal of this theses was to create new strains of a fungus, Aureobasidium pullulans, with enhanced cellulase production and use it to optimize an enzymatic saccharification, pretreatment, and fungal conversion process to enhance the nutritional value of DDGS. The northern Great Plains provide critical breeding habitat for waterfowl and gamebirds in the United States. Peak commodity prices in the late 2000s resulted in increased agricultural production and large-scale conversions of grassland habitat to monoculture row crops. However, recent declines in commodity prices have created a renewed interest for private landowners to diversify crops and enroll in government subsidized conservation programs that convert idle grassland and unproductive cropland to wildlife friendly perennial grassland plantings. Diverse cropping systems and no-till (NT) are beneficial in improving soil organic carbon (SOC) and other soils properties. The present study was conducted to evaluate the interactions and impacts of long-term tillage and diverse crop rotation systems on soil quality indicators that include a range of soil chemical, physical, hydrological, and microbial properties. This thesis examines the importance of conversional subsidies in accounting for an increase in organic acreage in the 12 North-Central States in the United States. Monthly time series data that spans from January 2002 to December 2014 was used in the analysis. Empirical evidence suggests that increase in organic acreage is due in part to the availability of conversion subsidies. Without government assistance, most small-scale farmers are not sufficiently motivated to switch to organic production due to the high initial costs involved in transitioning. Objective: The purpose of the study was to determine the relationship between hope, health-promoting behaviors, and health-related quality of life (HRQOL) in persons with Parkinson’s disease.
Background: The incidence of Parkinson’s disease is high in Midwest and Northeast regions of the United States. Parkinson’s disease affects motor and non-motor symptoms and has a variety of complications. Parkinson’s disease is related to genetic and environmental factors. HRQOL decreases in Parkinson’s disease; thus, the effect of hope and health-promoting behaviors on health is crucial as the disease advances.

The goal of this work was to develop electrochemical sensors for heavy metal ions (e.g., Cd2+, Hg2+, Pb2+) and phosphate detections in fertilizers with high sensitivity, high detection range, repeatability and portability. The heavy metal electrochemical sensors were made using novel graphene oxide/silver nanowire (GO/AgNWs) composite as working electrode. The phosphate electrochemical sensors were fabricated using ammonium molybdate tetrahydrate/silver nanowires (AMT/AgNWs) modified screen printed electrode. Sporeformers are common dairy contaminants, which are of a great concern to the dairy industry. Quality of dairy products is affected by these sporeformers such as Bacillus species, which are a significant cause of spoilage of dairy products. Thermal processes such as pasteurization have been used to inactivate pathogens, but some of the sporeformers and their endospores are resistant to such heat treatments. Remote ischemic postconditioning (rIpost) is a potent vasculoprotective stimulus that is thought to reduce reperfusion injury associated with heart disease. However, certain animal models of cardiometabolic risk factors such as dyslipidemia, diabetes, and hypertension suggest the beneficial effects of rIpost to lessen reperfusion injury may be diminished. These findings have not been studied in clinically relevant humans. To determine the effects of a diet that provides 30% energy from protein with ½ as lean, red meat on risk factors of metabolic syndrome in humans. This pilot study was a 3- month, randomized, control, intervention trial with 33 participants (Beef-Intervention n=18; DASH-Control n=15) who displayed markers of metabolic syndrome. Registered Dietitians Nutritionists recruited and educated participants on Beef-Intervention Lean Beef Pattern, (30% of energy from protein with ½ as lean red meat, 40% carbohydrate, 30% fat) or DASH-Control dietary pattern, (15% of energy from protein, 55% carbohydrate and 30 % fat). The purpose of this study was to determine the relationship between relative maximal (1RM) strength (i.e. back squat and power clean) to sprinting performance in 60-400m collegiate sprinters. A secondary purpose was to determine the distribution of athletes within the theoretical relationship between relative squat strength and performance capabilities. Fifty-six (n = 56) male and sixty-four (n = 64) female collegiate track and field sprinters were observed from DI (n = 88) and DII schools (n = 32) that participated in a year round strength and conditioning program. Background: Childhood obesity is a complex issue common in the U.S. today as it not only is associated with health-threatening comorbidities, but also increases a child’s likelihood of becoming overweight or obese during adulthood. Because of its complexity, several factors, including parents and the home environment, must be considered when assessing child weight status and also when aiming to prevent or treat childhood obesity.

Context: Shoulder injuries are frequently sustained in American football due to the contact and collision aspect of the sport. Injuries to the shoulder account for about 10% to 20% of all musculoskeletal injuries that occur in football and the shoulder is the fourth most commonly injured joint behind the hand, knee and ankle. The effectiveness of the dynamic stabilizers of the glenohumeral joint is a factor that could affect the rate of injuries that occur at the joint. The CKCUEST has been shown to be an effective test for assessing the dynamic stability of the glenohumeral joint.

To improve breastfeeding support in local businesses in Brookings, SD, researchers from South Dakota State University partnered with Brookings Health System, the Brookings Area Chamber of Commerce, local breastfeeding advocates, and an expert public deliberation moderator to conduct community-based participatory research leading to a public deliberation event. The collaborative team took a mixed-methods approach, using qualitative, quantitative, and rhetorical methods to collect and analyze data across two phases of the project: formative research (Phase I) and implementation (Phase II).
