Moisture extremes (excess and deficit) impact crop loss and water quality. Due to excessively wet springs and dry summers, crop damage can occur within the same county or field within the same year. To determine the magnitude of this problem in eastern South Dakota, indemnified crop insurance data for drought and excessive moisture claims were assessed for the years 1991-2020 for the occurrence of both excess moisture and drought in four counties in eastern South Dakota. Results show that there is greater than a 70% chance of the moisture extremes happening in the same year, making subsurface drainage, controlled drainage, …

Exploring Bioprocessing Technologies For Diverse Industrial Application Of Canola, Ahmad Fawzi N Alhomodi

Globally, canola is the second largest oilseed crop after soybean, which is processed for the commercial production of high value oil. The industrial processing steps for canola oil extraction include preprocessing (cleaning, pressing, flaking, and cooking), mechanical pressing, and/or organic solvents. This process results in large quantities of protein-rich (~ 40% dry basis) meal as a co-product. The meal is used partially in animal diet (<30% inclusion) due to high levels of antinutritional factors (ANFs) such as high fibers, phytic acid, and glucosinolates, and low metabolizable energy. Thus, this research was designed to examine various bioprocessing technologies (i.e., traditional sprouting, solid state fermentation, submerged state fermentation, co-culture fermentation, mild pretreatments, and their combinations) for possible advancement in canola use. Canola seed sprouting for 6-day period led to an increase in protein content and a reduction in ANFs and oil content of sprouts compared to ungerminated seed. Subsequent submerged state fermentation of 6- day old sprouts using three different strains (Aureobasidium pullulans, Trichoderma reesei and Neurospora crassa) further increased protein content and lowered ANFs. Solid-state fermentation of 144 h old sprouts using A. pullulans, N. crassa and T. reesei enhanced the protein content and reduced ANFs of sprouts. Sprouting canola seed for three days helped in hull removal, leading to high protein meal accompanied by low fiber and phytic acid level. Three-day seed sprouting had no effect on oil yield, but free fatty acid content was higher compared to seed oil. Co-culture fermentation of HECM under solid state process showed maximum reduction in fiber content with co-culture of A. pullulans and N. crassa while a combination of A. pullulans and T. reesei promoted the highest GLS and phytic acid reductions compared to other combinations, which indicated the advantage of coculture inoculation over monoculture in terms of ANFs reduction. Mild pretreatment of HECM using deionized water resulted in washed HECM (WHECM) with lower soluble sugars and GLS compared to untreated HECM, whereas protein and amino acid were concentrated due to the removal of soluble components. WHECM compared to HECM showed higher protein digestibility when fed to rainbow trout. Subsequent mono- and coculture fermentation of HECM and WHECM under submerged process resulted in higher protein and amino acid content and lower ANFs levels compared to uninoculated controls. The results of cellulase, endoglucanase and β-glucosidase activity indicated the crucial role of used substrates, fungi, fermentation modes (solid state/submerged stated) and inoculation methods (mono-/co-culture) on enzyme activities.

The objective of this thesis was to determine the influence of beef carcass weight on chilling and pH decline of beef carcasses, as well as color, case life and tenderness of steaks from the round, loin, rib and chuck. Twelve carcasses were allotted by hot carcass weight (HCW) into Heavyweight (HW) and Lightweight (LW) groups. Temperature decline, pH decline and thermal image temperature were measured in the round, loin, rib, and chuck for approximately 48 hours. Phosphorus (P) pollution has become a concern among multiple scientific organizations as it leads to eutrophication, an algal bloom that depletes lacustrine and marine ecosystems of native species. Multiple strategies can be implemented to reduce phosphorus loss from agriculture fields, which is often implicated as a cause of eutrophication. Soil phosphorus chemistry results in phosphate fertilizers absorbing to clay minerals over time. Soil phosphorus is lost from agricultural fields primarily through wind and water erosion. No-till practices prevent soil erosion, which reduces the phosphorus from loading into waterways. Fertilizer placement affects phosphorus loss. Future climate projections of warming, drying, and increased weather variability indicate that conventional agricultural and production practices within the Northern Great Plains (NGP) will become less sustainable, both ecologically and economically. As a result, the livelihoods of people that rely on these lands will be adversely impacted. This is especially true for Native American communities, who were relegated to reservations where the land is often vast but marginal and non-tribal operators have an outsized role in food production. In this study, the role of IFITM3 on PRRSV replication was studied in vitro by expressing exogenous IFITM3 in MARC-145 cells. An average of 31% reduction in PRRSV N protein expression and an average of 5.4 fold decrease in virus titer in the supernatant were observed in IFITM3 overexpressing cells as compared to vector control cells at 24 hours post infection (hpi). Moreover, there was a positive correlation between interferon- induced IFITM3 up-regulation and reduced PRRSV replication. To determine the role of endogenous IFITM3 in PRRSV replication, siRNA induced knockdown of IFITM3 was employed. RT-PCR validated the successful silencing of …

This work combines microbiology, molecular biology, Next-Generation Sequencing and system biology approaches to explore the mechanism of antibiotic persistence: a multi-drug tolerant, non-dividing, and metabolically altered state present in a subpopulation of cells due to phenotypic diversity rather than genetic variation (i.e. mutations). Persister can survive lethal antibiotic state and resuscitate after the treatment period is over. They are considered as the major contributing factor behind recurring infections. They also have a high mutation rate, which increases the chances of bacteria gaining antibiotic resistance. In the design of greener chemicals, deep eutectic solvents (DESs) are considered as one of the most versatile alternative solvents with widespread applications. DESs have the advantages of being nonflammable with negligible vapor pressure compared to traditional solvents. They share many characteristics of ionic liquids, but DESs are cheaper to formulate, typically nontoxic, recyclable, biodegradable, and are suitable for use with biological systems. Identity Negotiations In Family Relationships: Discussing Substance Use Disorder Through The Communication Theory Of Identity, Sarah-Michele Q. Weaver

Guided by the communication theory of identity (CTI), the current study explores identity negotiations among family members who have experience with substance use disorder. Semi-structured interviews were conducted with 9 participants. Personalenacted, enacted-relational, and personal-relational identity gaps guide the scope and use of CTI. Findings mirrored 3 major themes in consideration of these identity gaps: (1) participants embraced or rejected SUD as a central aspect of personal identity, (2) participants experienced turbulence in relational identities due to SUD, and (3) personalrelational identity gaps and alignment surface in multiple contexts where interpersonal communication occurs.

Cheddar cheese is a commodity and a top-selling cheese in the United States. Enhanced flavors of cheddar cheese requires aging, which will allow proteolysis increase. Proteolysis is the most complex process that happens in cheese manufacturing and can be difficult to control because of environmental factors and compositional characteristics. The ripening process, which is the most timely and costly procedure in cheese manufacturing, can also be unpredictable when aspects, such as cheese composition, are changed. Lipid-laden macrophages contribute to atherosclerotic plaque formation in atherosclerosisrelated heart diseases. Thus, understanding formation and degradation of lipid would help identify therapeutic targets for the treatment of atherosclerosis. In the first chapter of this dissertation, I have explained the current understanding of cholesterol metabolism in macrophages and how mTOR could play a potential role in this process. Additionally, I have described how CRISPR/Cas9 whole-genome screens present an effective tool to identify genes regulating cholesterol metabolism including positive and negative regulators of lipid droplet formation and lipid droplet degradation. Wheat is one of the most important staple crops providing 20% of energy for 35% of the world population. Fusarium head blight (FHB), primarily caused by the fugal Fusarium graminearum Schwabe, is a damaging disease in wheat that affects global wheat production every year and causes food safety issues. The disease not only reduces the grain yield and quality but also produces mycotoxin in the diseased kernels making them unsuitable for human consumption or as livestock feeds. Most wheat varieties are susceptible to bacterial leaf streak (BLS), a major disease caused by Xanthomonas translucens pv. undulosa (Xtpvu). BLS is challenging to manage since common chemicals do not provide adequate control. Plant-based diets contain numerous flavonoid compounds that produce antibacterial effects and gut health improvement. Genistein is among the most abundant isoflavones present in a plant-based diet and can be found in high amounts in soy products (up to 1g Kg-1). We describe here a robust method to identify genistein tolerant and metabolizing bacteria in swine gut microbiota and to screen the strains that have antibacterial and immunestimulatory properties. Such strains could be developed as non-antibiotic alternatives to prevent enteric infections in pigs and improve gut immunity. Invasive species are forces of environmental, economic, and social change with detrimental impacts on ecosystems, commerce, agriculture, and human health. However, we still have a poor understanding of the processes that could be driving invasive potential. Hybridization may contribute to the establishment and spread of invasive lineages, as it may enhance genetic variation and facilitate rapid adaptation. Focusing on Russian thistle (Salsola tragus s.l., family Amaranthaceae) and closely related species, my thesis aims to clarify phylogenetic relationships and investigate how hybridization has shaped the invasion dynamics of these lineages.
