Single-sourced, newly weaned steers (n=256; initial BW=542 ± 3.7lb; n=64 steers/treatment; 8 steers/pen) were blocked by location in a 2×2 factorial arrangement of DFM (Certillus CP B1801 Dry; Bacillus subtilis, Lactobacillus plantarum; 28 g/steer·d-1) and YCW (Celmanax; 18 g/steer·d-1). Temperature-humidity index (THI) was calculated as: THI=0.81×ambient temperature+[relative humidity×(ambient temperature-14.40)]+46.40. Single-sourced, newly weaned steers (n=70; initial body weight (BW)=560±12.9lb) were allotted to 10 pens (n=5 pens/treatment with 7 steers/pen). Previously identified pregnant females from four different herds and postpartum females from one herd were utilized. Blood samples were collected (n = 1,753; study 1) between d 28 and 285 of gestation and (heifers n = 418 and cows n = 657; study 2) once a week for up to 12 weeks after calving. Blood samples were collected from six different Bos taurus herds between day 27 and 285 of gestation (heifers n = 1,205 and cows n = 1,539). Blood samples to determine PAG clearance interval were collected weekly postpartum for up to 12 weeks (heifers n = 418 and cows n = 657). Study Description:
Semen from 22 dairy bulls was used to evaluate the presence, localization, and quantification of DAG1 and SERPINA5 on sperm. Sperm motility parameters and viability was also evaluated for semen from each bull. Semen from 19 out of the 22 dairy bulls was used for in vitro embryo production (two Low-SCR and one High-SCR were not available for in vitro embryo production). Bulls were classified based on their sire conception rates (SCR) values as High-SCR (SCR > 1.0) or Low-SCR fertility (SCR < -4.0). Low fertility bulls were subdivided based on their blastocyst rate (BL) as High-BL (Low-SCR/High-BL BL ≥ 31%) or Low-BL (Low-SCR/Low-BL BL ≤ 26%), and High-SCR bulls were not subdivided. The GLM procedure in SAS was used with bull as a fixed effect to determine if variance was greater between bulls compared to within bull. Correlations were determined among DAG1 and SERPINA5 concentrations, percentage of tail labeled for SERPINA5, SCR, sperm total motility, progressive motility, and viability, and in vitro embryo produced cleavage rate (CL) and BL. The GLIMMIX procedure of SAS was used to evaluate the relationship of bull field fertility (High- and Low-SCR), and field and in vitro fertility (High-SCR, Low-SCR/High-BL, Low-SCR/Low-BL) classifications with sperm total (TMOT) and progressive (PROG) motility, viability, CL, BL, DAG1 and SERPINA5 relative concentration, and proportion of sperm tail labeled for SERPINA5. Both SERPINA5 and DAG1 were localized on the sperm head; however, SERPINA5 was also localized on the sperm tail. There was greater variance in concentration among bulls compared to within bull for both DAG1 (P < 0.01; 69.4 vs 49.1, respectively) and SERPINA5 (P < 0.01; 325.8 vs 285.4, respectively). There was a positive correlation between concentration of DAG1 and SERPINA5 (P = 0.01; r = 0.54). Concentrations of SERPINA5 were also correlated with CL (P = 0.04; r = 0.48), and percentage of sperm tail labeled for SERPINA5 was correlated with viability (P = 0.05; r = 0.44) and tended to be correlated with CL (P = 0.10; r = 0.39). There was no relationship between SCR or BL rate classifications and DAG1 (P ≥ 0.66), SERPINA5 (P ≥ 0.54), or percentage of sperm tail labeled for SERPINA5 (P ≥ 0.48).

Ejaculated and epididymal semen was collected from mature Angus bulls (n = 9), and then centrifuged to separate sperm and fluid. Fluids were collected and sperm pellets were resuspended in a high ionic solution and vortexed to remove loosely attached proteins. Sperm samples were centrifuged, and the supernatant was collected. Heifers were stratified into two groups based on initial body weight and each group was randomly assigned to one of two treatments: a control group (n=30) and a precision group (n=30). Treatment groups are separated into different pastures to graze dormant native range from November to April. Groups are rotated monthly to reduce individual pasture influence. Six herds of yearling steers (n=127) will graze native summer pastures from May to August at the Cottonwood Field Station near Philip, South Dakota. Six pastures are divided into two groups: a continuous grazing (CG) treatment with a low, moderate, and high stocking density and a virtually fenced rotational (VFR) grazing system with the same stocking densities. Cattle DMI is an essential component of calculating cattle stocking rates, determining nutrient requirements, and evaluating feed efficiency. Cattle DMI and digestion of forages impact enteric greenhouse gas (GHG) emissions; a major public and environmental concern. Increased GHG levels indicate energy loss during the rumen fermentation process. Obtaining data for rangeland cattle DMI and GHG emissions is needed to understand and enhance individual animal performance and reduce negative environmental impacts. Data were collected from Wind Cave National Park and 6 satellite bison herds across the upper Midwest ranging across South Dakota, Indiana, and Kansas with a MAP gradient ranging from 34 to 82 mm and a MAT gradient ranging from 9 to 13°C. Fecal samples were collected non-invasively from 50 freshly voided bison dung patties at each herd. Helminth eggs were recovered and identified using sugar floatation; Trichostrongyle-type eggs were hatched by coproculture to identify to species. Twelve head of fed beef cattle were harvested at the SDSU Meat Laboratory over two days. Carcasses were allotted into two weight groups based on hot carcass weight (HCW): Heavyweight (HW) and Lightweight (LW). Data logging thermometers were placed in the left side of each carcass within the round, loin, rib, and chuck primals to track temperature decline. Carcass measurements including 12th rib fat thickness, ribeye area and marbling score were collected approximately 48 hours postmortem. Steaks from each primal were collected to measure Warner-Bratzler shear force (WBSF), objective and subjective color.

Mice are often the model of choice for genomic research because of their high similarity to the human genome and relatively easy maintenance. Thus, understanding the mechanisms and principles of their genome is vital to advancing our knowledge about human biology. Long interspersed elements type 1 (LINE-1s or L1s) are transposable elements within mammalian DNA. Promoter activities have been identified for the human L1 5’ untranslated region (5’UTR) in both the sense and antisense orientation, but it was previously unknown if mouse L1 5’UTRs have antisense promoter activity. Benign gynecological disorders can affect a high percentage of women of reproductive age, ages 15-44 years. These conditions can affect the lifestyle of the individual and can be associated with infertility. The gold standard to identify and diagnose endometriosis requires invasive surgical procedures, while the Rotterdam Criteria is used to identify and diagnose polycystic ovary syndrome. The purpose of this paper is to discuss, describe, and characterize potential non-invasive biomarkers that are present in various pathological stages of both endometriosis and polycystic ovary syndrome. Drinking water quality requirements focus mainly on removing hazardous substances from the water supply, making it safe to drink and appropriate for livestock use. However, there is a concern that high mineral content in water may affect the drinking behavior of heifer calves, which may affect health and performance. In South Dakota, the primary water sources for agricultural use are surface or groundwater. Water quality varies between different regions of the state; in the eastern part of the state, the water is classified as “hard water” and calcium is the primary mineral found in the water. Conventional agricultural practices can have unintended consequences on the environment and wildlife. Insects and birds are declining at rapid rates around the world, and the current conventional agricultural paradigm is a major driver through habitat loss and the intensification of production. Invertebrates in agroecosystems provide services to both farmers and the rest of society. Regenerative systems may promote the functioning of an agroecosystem by influencing invertebrate abundance, diversity, and ecosystem services and mitigate bird and insect declines through conservation practices that increase soil health, reduce disturbances, and increase biological diversity. Prairie cordgrass (Spartina pectinata) (PCG) is a warm-season perennial grass that can be used as a biofuel feedstock and can be grown on marginal lands. Previous studies on intercropping of a perennial legume i.e., kura clover (Trifolium ambiguum) (KC) with PCG can improve soil biochemical properties, increase biomass production, mitigate greenhouse gas (GHG) emissions while reducing the chemical fertilizer requirement. Grasslands historically covered 46 million km2 of Earth’s surface representing nearly 42% of the living vegetation. Encroachment of woody species onto grasslands and savannas is a widely researched global phenomenon, with eastern redcedar (ERC) (Juniperus virginiana L.) trees being the most prominent woody encroacher in the Northern Great Plains (NGP) due to the lack of prescribed fire and planted ERC escaping from shelterbelts. This encroachment poses a threat to native plant communities in terms of their reproduction, regeneration, and diversity. Advances in space technology and interest toward remote sensing mission have grown in the recent years, requiring the attitude control subsystems of observation satellites to increase their performances in terms of pointing accuracy and on-board implementability. The objectives of this thesis project were to 1) evaluate the influence of grain- and grass-finishing systems on carcass characteristics, proximate composition, nutritional profile, and fatty acid composition of bison bull meat, and 2) evaluate the influence of animal age on carcass characteristics of bison bulls. For objective 1, bison bulls were allowed to graze native range in north-central Nebraska until approximately 26 mo of age, when they were randomly assigned to either grain-finishing (n = 98; in an open lot with ad libitum access to prairie hay, alfalfa hay and corn for 95 d prior to slaughter) or grass-finishing …

Pallid Sturgeon (Scaphirhynchus albus) are a federally endangered species experiencing widespread reproduction and recruitment failures. Recruitment failure is hypothesized to be caused by habitat modifications made to the Missouri River that disrupt connectivity and alter temperature profiles downstream of constructed dams. Hypolimnetic releases from Missouri River dams affect the temperature downstream, creating colder conditions during the downstream drifting phase for dispersing larvae. Understanding the influence of water temperature changes on larval development is crucial for recovery efforts.
