Six experiments were conducted for this dissertation research to evaluate strategies to improve growth performance and health of calves. Growth performance, nutrient utilization, and health effects were evaluated when calves were offered water treated with reverse osmosis system (RW), fed starter pellets supplemented with microbially-enhanced soy protein (MSP) or supplemented condensed whey solubles (CWS). Various forms of deep eutectic solvents have been synthesized with advantages of 100% atom economy, low vapor pressure, ease of preparation, cost efficiency, low toxicity, and tunability. Though several DESs have been synthesized and characterized, including binary and ternary solvents, none of the binary DESs have been formulated with water as the sole hydrogen bond donor(HBD). Hepatocellular carcinoma (HCC) is considered the third leading cause of death from cancer. Overall survival rate is significantly low, due to the emerging resistance to chemotherapeutic agents and lack of selectivity. Recent studies have demonstrated that epidermal growth factor receptor (EGFR) is a promising molecular target for cancer therapy, especially HCC. Current studies showed that cucurbitacins are potent anticancer compounds which target EGFR. Phytochemicals such as tannins included in dairy cow nutritional programs, as a natural feed additive, promise to improve rumen bypass protein and consequently reduce the feeding cost by decreasing the amount of CP needed in the ration. Additionally, if tannins are fed along with high quality protein sources, this could improve the AA profile reaching the small intestine and potentially improving health and performance of dairy cows. In pregnant pigs, amino acid (AA) requirements represent the sum of those required for maintenance functions, protein retention and efficiency of utilizing AA intake for the aforementioned body processes. The NRC (2012) model assumed AA efficiency is constant across period of gestation; however this is not reflective of the changes in metabolic demand during gestation. Therefore, two experiments were conducted to evaluate the efficiency of utilizing SID Lys and Thr for whole body protein retention (kSIDLys and kSIDThr) in pregnant gilts during early, mid and late gestation. Computational fluid dynamics software was used to simulate the motion of circulating tumor cells in a variety of microfluidic cell isolation devices. Design of several novel microfluidic cell isolation devices was aided by viewing streamlines of fluid in devices in simulation. Devices that performed best in simulation used 5-micrometer wide guiding channels to guide cells to the capture location in the device. While these devices performed better than other devices in simulation and captured all particles regardless of position along inlet, experimental results differ from simulation.

Corn dried distillers’ grains with solubles (DDGS) can be a good alternative feedstuff to the traditional corn soybean meal diets since it has high fat and protein content. However, it has a high fiber content, which is not well digested by pigs and can reduce nutrient utilization by encapsulation. The nutritive value of fibrous feedstuffs like DDGS can be improved by supplementation with fiber-degrading enzymes. Six primer pairs, each with a pair of genus-specific and universal second internal transcribed spacer (ITS2) primers were evaluated to amplify and sequence the ITS2 region of nuclear ribosomal DNA to identify the species and genotypes of Haemonchus from livestock fecal samples containing mixtures of trichostrongyle nematode eggs. Based upon the use of universal ITS2 primers on Haemonchus adults, the ITS2 region has been shown to contain three fixed single-nucleotide differences between H. placei and H. contortus. The main purpose of this body of research was to evaluate carinata meal as a feedstuff for dairy heifers and lay the foundations for its future evaluation in lactating dairy cow diets. This research focused on evaluating the taste preference of carinata meal compared with other oilseeds meals and distillers dried grains with solubles (DDGS). Despite containing glucosinolates, carinata meal was preferred similarly to canola meal, had greater preference compared to camelina meal, and less preference compared to the other meals. Humus is a class of complex, organic molecules that are largely responsible for improving soil water holding capacity, nutrient mineralization, nutrient storage, and other critical soil functions. In addition to the best feedstock sugarcane, industrial beets are of great potential to become another ideal feedstock for production of advanced biofuels and high-value chemicals. Beets produce readily-fermentable sugars (especially sucrose), eliminating the cost for starch hydrolysis required for corn-based biofuel production. However, the whole beet has very high-water content (75% water), which is a big challenge for transportation and storage. Air-propelled abrasive grit management has been reported to control weed seedlings in corn and soybeans while maintaining yield. This research examined the weed control, corn and soybean yields, nitrogen mineralization, and nitrogen yield responses from grits. Although the current FDA approved cyanide antidotes (i.e., sodium nitrite, sodium thiosulfate, and hydoxocoboalamine) are effective for treating cyanide poisoning, each individual antidote has major limitations, including large effective dosage, delayed onset of action, or dependence on enzymes generally confined to specific organs. To overcome these current limitations, next-generation cyanide antidotes are being investigated, including 3-mercaptopyruvate (3-MP). Analytical methods capable of detecting 3-MP from plasma are essential for the development of 3-MP as a nextgeneration cyanide antidote. Effects of enzymatically hydrolyzed yeast (EHY) and supplementation frequency (FREQ) on immune parameters among beef cows and calves was evaluated. Eighty multiparous (parity = 4.2 ± 0.3) cows were fed a common brome hay-based diet (CP = 8.0 ± 0.17%). Cows were blocked by expected calving date and stratified by body condition score (BCS) before random assignment of treatment. Bovine viral diarrhea virus (BVDV) and bovine herpesvirus type 1 (BHV-1) are important pathogens that affect cattle. In this in vitro study, the main goal was determining the direct effects of BVDV and BHV-1 infections of monocyte-derived dendritic cells (MDDC) on cytokine mRNA expression. For the BVDV infected MDDC experiments, Holstein Friesian and Brown Swiss calves were included with a breed comparison with cytokine expression. Surface marker expression was also measured in BHV-1-infected MDDCs. The swift fox (Vulpes velox), a native species once abundant throughout the Northern Great Plains (NGP), has declined due to changes in land use, historic predator eradication programs, and predation by larger canid species. Currently, the species is estimated to occupy 44% of its historic range. In the NGP, the status of the swift fox varies by state, ranging from furbearer to endangered species. Infected individuals vary in their contribution to disease persistence, and chronically infected individuals may sustain disease in a population. One disease that might persist in a population through chronically infected individuals is pneumonia in wild sheep. Mycoplasma ovipneumoniae (Mo), a pathogen of Caprinae commonly present in domestic sheep and goats, strongly correlates with pneumonia epizootics when it infects wild sheep populations. These epizootics can cause 40-100% herd mortality in an initial all-age dieoff, precipitate annual lamb mortality as high as 100% in following years, and sustain adult mortality long after initial all-age dieoffs.
