Thrombin is a key enzyme involved in the development and progression of many cardiovascular diseases. Direct thrombin inhibitors (DTIs), with their minimum off-target effects and immediacy of action, have greatly improved the treatment of these diseases. However, the risk of bleeding, pharmacokinetic issues, and thrombotic complications remain major concerns. In an effort to increase the effectiveness of the DTI discovery pipeline, we developed a two-stage machine learning pipeline to identify and rank peptide sequences based on their effective thrombin inhibitory potential. The positive dataset for our model consisted of thrombin inhibitor peptides and their binding affinities (KI) curated from published …

Recommender systems have revolutionized the way users discover and engage with content. Moving beyond the collaborative filtering approach, most modern recommender systems leverage additional sources of information, such as context and social network data. Such data can be modeled using graphs, and the recent advances in Graph Neural Networks have led to the prominence of a new family of graph-based recommender system algorithms. In this work, we propose the RelationalNet algorithm, which not only models user–item, and user–user relationships but also item–item relationships with graphs and uses them as input to the recommendation process. The rationale for utilizing item–item interactions …

Community detection has been (and remains) a very important topic in several fields. From marketing and social networking to biological studies, community detection plays a key role in advancing research in many different fields. Research on this topic originally looked at classifying nodes into discrete communities (non-overlapping communities) but eventually moved forward to placing nodes in multiple communities (overlapping communities). Unfortunately, community detection has always been a time-inefficient process, and datasets are too large to realistically process them using traditional methods. Because of this, recent methods have turned to parallelism and graph stream models, where the edge list is accessed …

This paper develops a new approach for the design of polynomial-based reference and feedforward control trajectories for the high precision control of flexible servo actuators. First, a robust frequency-domain controller is designed for reference tracking and disturbance rejection. This controller is designed to achieve a desirable loop shape with reasonable bandwidth and stability margins. Then, the polynomial trajectories are derived based on a simplified rigid body representation of the system. To avoid the excitation of the system's resonant modes, this paper proposes the design and implementation of a set of feedforward notch filters. We further investigate the main cause of …
