As rendering engines become increasingly important in film and television, with their use in virtual production (VP) to display rendered imagery, some underlying issues become more apparent. This thesis aims to investigate how we can improve asset color matching of VP elements with real-life objects found on sets. Experiments were conducted in which objects were exposed to various types of lighting setups, and digital twins were rendered using traditional computer graphics techniques. Degenerative disc disease is a major contributor to low back pain, characterized by inflammation of the intervertebral disc (IVD), degradation of the extracellular matrix, loss of hydration, and cell death. Current therapies fail to address these underlying mechanisms, underscoring the need for regenerative strategies. Mesenchymal stem cells (MSCs) exhibit immunomodulatory and regenerative potential, but their efficacy is hampered by the harsh microenvironment of the degenerated IVD. Acellular MSC-derived extracellular vesicles (EVs) have shown therapeutic potential and offer a promising alternative for IVD regeneration. This chapter tells intertwined stories of the rescue and preservation of two archival collections of instructional (classroom) television programs and attendant materials from the ITV new wave of the 1970s and 1980s. A.I.T., the Agency for Instructional Television/Technology created innovative programming combining instructional design and media artistry, all focused on learners. The ability, afforded by these collections, to address key lacunae in the history could have significant implications for the design of learning media in its current forms of interactive media and video games for learning as well as the development of the next evolution of learning media and technology.

Cells constantly sense and respond to biophysical cues in their microenvironment. Understanding how they interpret these mechanical signals is essential for advancing tissue modeling, studying disease mechanisms, and developing more predictive in vitro platforms. This dissertation presents a series of engineered microphysiological systems (MPS) designed to investigate how mechanical stimuli shape cellular behavior in distinct human tissue microenvironments. In Aim 1, I developed a reconfigurable microfluidic platform that mimics key features of human vascular barriers. The increasing demand for efficient separation processes of micron-sized particles, including microorganisms, highlights the importance of developing low-cost, non-labor-intensive, and time-efficient techniques. Insulator-based electrokinetic (iEK) systems have proven to be a practical alternative to bench-scale methodologies such as centrifugation and membrane filtration. Several studies have reported the separation of microparticles by employing direct current (DC) voltages. However, all of them have overlooked the phenomenon of nonlinear electrophoresis (EPNL) and its role in the electrokinetic migration of particles. We present a unified framework for estimating stochastic parameters in general variational problems. This nonlinear inverse problem is formulated as a stochastic optimization problem using the output least-squares (OLS) objective, which minimizes the discrepancy between observed data and the computed solution. A key challenge in OLS-based formulations is the efficient computation of first- and second-order derivatives of the OLS functional, which depend on the corresponding derivatives of the parameter-to-solution map—often costly and difficult to evaluate, especially in stochastic settings. This study focused on the Leatherback Turtle (Dermochelys coriacea), a species currently classified as Vulnerable on the IUCN Red List, evaluating the conservation policies and measures implemented by selected English-speaking countries. My Two Bodies examines the intersection of visibility, memory, and gendered absence through photographic and material strategies. Engaging with censorship and instability of the image, it investigates how realities are shaped by what is seen, obscured, or erased. Referencing the gendered absence of women in Iranian visual culture, the project considers how censorship intensifies rather than eliminates visibility. This thesis looks at how to balance energy efficiency with embodied energy in small commercial retrofits, focusing on restaurant buildings. Rather than tearing down older buildings and starting from scratch—something that wipes out all the embodied energy already invested—retrofitting can improve performance while preserving what's already there. Using EnergyPlus for operational energy modeling and the Athena Impact Estimator for embodied energy analysis, this study compares different retrofit scenarios. We examine the behavior of C-shaped superellipse sector particles (SeSPs) as they travel through an obstacle field. SeSPs are two-dimensional particles parameterized by corner sharp- ness, aspect ratio, and opening angle; these parameters are sufficient to represent a large variety of shapes, including discs, rods, and concave and convex particles. Our obstacle field is a rhombic Galton Board, an array with alternating rows of pegs.
