Development And Evaluation Of Tracer Particles For Use In Microzooplankton Herbivory Studies, Mary Putt

Two methods of preparing algae for use as tracer particles in single species measurements of microzooplankton herbivory were evaluated. Algae were either heat-killed and labelled with 5-(4,6-dichlorotriazin-2-yl amino fluorescein) (DTAF) (Rublee & Gallegos 1989; Mar, Ecol. Prog. Ser. 51: 221-227) or stained with hydroethidine (HYD). Both DTAF and HYD-stained algae were readily visible within the digestive vacuoles of most microzooplankton collected in estuarine and coastal waters of Massachusetts (USA) and preserved with Lugol's iodine. However, DTAF was ineffective at staining several chromophytic algae and the heat-kill process reduced cell volume by ≥ 50% in several of the algae which were …

When mortality rates by age are calculated from recorded deaths and enumerated populations, rates at higher ages are typically in error because of misstated ages. Mortality rates for China in 1981 have been calculated from the number of deaths in 1981 in each household recorded in the 1982 census, and from the census population back-projected one year. Because age was determined from date of birth, and because persons of the Chinese culture have very precise knowledge of date of birth, the mortality rates even at high ages should be unusually accurate. Weekly period meanders and eddies are persistent features of Gulf Stream frontal dynamics from Miami, Florida, to Cape Hatteras, North Carolina. Satellite imagery and moored current and temperature records reveal a spatial pattern of preferred regions for growth and decay of frontal disturbances. Growth regions occur off Miami, Cape Canaveral, and Cape Fear due to baroclinic instability, and decay occurs in the confines of the Straits of Florida between Miami and Palm Beach, between 30° and 32°N where the stream approaches the topographic feature known as the Charleston bump and between 33°N and Cape Hatteras. Dissolved lignin-derived compounds in seawater indicate the presence of organic matter originating from vascular plants and therefore from terrestrial (upland and coastal marsh) ecosystems. We used a hydrophobic resin to concentrate lignin-rich humic substances and to determine concentrations of lignin oxidation products (vanillyl lignin phenols) for waters of the continental shelf of the southeastern U.S. Lignin phenol concentrations ranged from 0.05 to 4.2µg liter‒1 and accounted for 0.002–0.13% of the total dissolved organic carbon (DOC) pool in continental shelf waters. Self-activation and self-inhibition of cell number density (or growth factor concentration) due to a spatially localized source are studied. Both the time-independent and time-dependent models are examined, and the linear stability of the resulting three steady states of the former is discussed.

This note develops a simple model for the competition between activator and inhibitor control mechanisms in one-dimensional tissue growth. The pedagogic usefulness of such a model is that it is easily accessible to undergraduate applied mathematicians and is suggestive of behavior known to occur in more realistic biological systems (e.g., some types of cancer). The limitations of the model are obvious and can provide a basis for discussion of the applicability of complementary levels of description in mathematical modeling.

Waters associated with the Chesapeake Bay plume were highly variable, unstable, and subject to the influence of flow patterns into and out of the Bay. Seasonal phytoplankton assemblages were identified for this region, with the phytoplankton development similar to the seasonal spring-fall growth maxima over the shelf, yet influenced by the multi-pulsed patterns often noted in the lower Bay. Major dominants included species common to both the shelf and lower Chesapeake Bay.

Marge Piercy's novel Small Changes is encyclopedic in its incredibly detailed, all-encompassing feminist analysis of female and male behavior in the late 60's and early 70's. The behavior of the younger generation is compared and contrasted with that of their parents. The overall impression given by the novel is that, despite the very different life-styles of the two generations, very little change has, in fact, occurred. At the end of the novel, sexism prevails and no significant threat to male control of the power structure has developed. From examination of the title, Piercy seems to place her emphasis not upon …

Cultured human keratinocytes were grown in vitro to assess the effects of glutathione, a thiol, on human keratinocyte proliferation. Single keratinocytes were harvested from newborn (greater than 5 days old) foreskin and were grown in culture flasks such that experiments could be performed in approximately 2 to 3 weeks. In order to evaluate the effects of glutathione (GSH) on keratinocyte growth, experiments were performed to determine the growth curve, plating efficiency, and incorporation of radiolabelled thymidine. Growth studies, performed with different concentrations of glutathione, revealed that 0.065 μM was the most effective concentration when compared to control cell cultures, and …

Sediment denitrification potential from two sites in the Elizabeth River estuary was studied over a nine-month period using the acetylene blockage method. Rates of microbial processes in this environment are of interest because of the high concentration of toxics present in some parts of the system. Highest rates were found in the highly polluted Southern Branch of the Elizabeth River with nitrate amended sediment ranging from 2-262 nmol N20/h per 20ml of sediment and exhibiting maximal rates during spring and fall. A comparison of the total percentage cell abundance and cell biovolume relationships of major phytoplankton categories was made between two station sets across the shelf margin. Diatom values for abundance and biovolume were greater at oceanic stations compared to the outer shelf stations, with dinoflagellates having the reverse pattern. The composite contributions to biovolume and abundance in the standing crop from other phytoplankton categories were greater over the outer shelf than beyond the shelf margin. Male Long-Evans rats were conditioned 6 days, alternating drug and non-drug conditions, using injections of .6 mg/kg d-amphetamine, amphetamine plus 60mg/kg ethanol, or amphetamine plus 120 mg/kg ethanol. These injections were given in a specific side of the shuttlebox to assess the development of a place preference or aversion. The amount of time spent in the drug-paired side was analyzed on 2 non-drugged test days. Rats spent significantly more time in the drug-paired side on Test Day 1 than on Test Day 2. When the ethanol groups were combined, these rats spent significantly more time in the drug-paired side than …

The purpose of this study was to isolate and purify the enzymes, transfer RNA-guanine ribosyltrasferase and protein kinase C, and to determine whether the phosphorylating enzyme activates the insertion enzyme in vitro. Transfer RNA-guanine ribosyltransferase lost activity within several days after isolation and total, complete reactivation was accomplished in the presence of protein kinase C. This demonstrated that ribosyltransferase's instability is related to the degree of its phosphorylation. It is proposed that modification of tRNA is controlled by protein kinase C. Deactivation of the insertion enzyme leads to hypomodified tRNA which in turn is associated with neoplasia. Potential use …

Polycyclic aromatic hydrocarbons (PAHs) such as naphthalene (Nap) and benzo[a]pyrene (B[a]P) are common in the water column and sediments of estuarine and coastal areas. The mud crab Rhithropanopeus harrisii was exposed to varying concentrations of Nap or B[a]P to determine the effects on reproductive success, embryological development, larval physiology and mortality. Nap and B[a]P decreased hatching success and prolonged embryological development. Both pollutants produced high abnormality frequencies for egg exposed larvae. Molt frequency and respiratory rates of larvae from non-exposed eggs were more chronically affected by these pollutants than were egg exposed larvae. The EC50 concentrations were determined for …

The formation and cycling of iron sulfides within anoxic sediments can affect the cycling of trace metals. The sediments at station CB-27 (near Annapolis, Maryland) in Chesapeake Bay were rich in FeS (39% of TS), low in greigite (3.6% of TS), and has a relatively constant pyrite concentration (50 - 60% of TS) with depth. The percent composition of total metals in pyrite were 25.7% cu, 11.8% Zn, 15.8% Fe, and non-detectable to 2.1% Cd. A biomass difference approach was used to measure biomass and production over eleven months in four different communities within the Great Dismal Swamp. On all sites, fine roots were the moat dynamic root component. Both biomass and production were highest on the mixed hardwood site, the least flooded site, and second highest on the cedar site, the site with the longest duration of saturation. The maple-gum and cypress sites had lower amounts of biomass and rates of production. Environmental parameters that influenced production included frequency and duration of flooding, density of the stand, and soil type.

The embryonal carcinoma PCC4uva differentiates into neurons in response to treatment with retinoic acid and dbcAMP. We used this in vitro model system to study the effects of laminin on early neural differentiation. Laminin substrata markedly potentiate neural differentiation of retinoic acid and dbcAMP-treated cultures. Only laminin induced more rapid neural cell body clustering, neurite growth and neurite fasciculation as compared to type IV collagen, type I collagen, and fibronectin substrata. Exogenous laminin substrata promoted greater cell attachment, cellular spreading and growth to confluence than type IV collagen, type I collagen, fibronectin and glass substrata. This study was performed to determine if there is a quantifiable difference between quantitative fit testing using a probed facepiece and a probed cartridge. Four subjects were tested on four different respirators. The respirators used were MSA & 3M full and half face respirator. Each subject performed each method five times per respirator. The results indicated no correlation between the two fit testing methods overall. However, a moderate correlation was found between the MSA respirators and the two methods. This study explores the feasibility of using an applied soil repellent finish, Scotchgard, on 100% cotton denim fabric in order to enhance the removal of atrazine and alachlor residue during the laundering process. The experimental findings provided no statistically significant evidence that Scotchgard fabric protector reduced pesticide residuals on the 100% cotton denim fabric either before or after laundering. A significant finding secondary to the study involved the possible interaction of Scotchgard with the pesticide alachlor, leaving greater amounts of residual on the fabric after laundering than those found with no fabric treatment. This study examined the relationships of body-image attitudes and sexual attitudes, experiences, and functioning in a college population. Sixty-five males and 120 females completed a variety of established self-report measures assessing the constructs under study, as well as two new measures developed in this study. The new measures included (1) The Sexual Experiences and Problems Survey for self-assessment of difficulties in sexual functioning, and (2) The Body Exposure in Sexual Activities Questionnaire to measure self-consciousness and avoidance of body exposure in sexual contexts. The goal of this thesis is to develop an artificial neural approach toward addressing the intractability involved with the decomposition problem. The search for the lattice of substitution property (s. p.) partitions essential to decompositions is cast into the framework of constraint satisfaction. An artificial neural network is developed to provide solutions by performing optimization of a mathematically derived objective function over the problem space. Triphenylethylene antiestrogen compounds are used to stimulate ovulation in anovulary women and spermatogenesis in oligozoospermatic males. Clomiphene Citrate (CC) is given therapeuticaly as a mixture of the trans (E) and cis (Z) isomers. Each of these isomers has its own characteristic agonist-antagonist profiles. A sensitive and simple microwell based competition enzyme immunoassay for the quantitative determination of estradiol (E2) and estriol (E3) was developed. Nicrowells coated with antibody were incubated with antigen followed by adding horseradish peroxidase (HRPO) conjugate. The assay, which can be performed within two hours at room temperature, involved simultaneous incubation of E2 - or E3-HRPO conjugate and serum sample in polystyrene microwells coated with anti- E2 or anti-E3 gamma globulin fraction.

An enzyme-labeled immunoassay of sufficient range and sensitivity was developed for serum testosterone determination. In this method, the testosterone in the serum sample and testosterone-HRPO conjugate compete with each other for the limited antibody binding sites on the anti-testosterone gamma globulin coated microwell. The assay can be performed in only 2.5 hours.

The gamma globulin fraction was isolated from the anti-testosterone whole serum by DEAE-cellulose column chromatography. Testosterone-peroxidase conjugate was prepared by a carbodiimide coupling method. The interference which arises from the binding of testosterone to steroid hormone binding protein was eliminated by the addition of 17 beta-estradiol. All assay …

The direct stimulation of sensory nerves or the intradermal injection of neuropeptides triggers a cutaneous inflammatory response that involves the degranulation of mast cells. These neurogenic inflammatory reactions are physiologically similar to IgE-mediated hypersensitivity responses, which also depend on mast cells. Although non-antigenic stimuli distinguish neurogenic from IgE-mediated inflammation, the similarity of their effector stages suggested that peripheral nerves may also participate in lgE-mediated responses. To examine this hypothesis, IgE responses were elicited in denervated skin. Phytoplankton In Lake Drummond And Two Drainage Ditches In The Great Dismal Swamp, Virginia, Christine Gregory Phillips

A twelve month study was conducted on the phytoplankton of Lake Drummond and Washington and Jericho Ditches from December 1988 to November 1989. Surface water samples were collected for nutrient and chlorophyll analysis from June 1989 to November 1989. The dominant phytoplankton component was the pico-nanoplankton (< 3 micron) category. A total of 57 species were identified from Lake Drummond, 51 species from Washington Ditch, and 31 species from the Jericho Ditch. Three major phytoplankton groups were identified at these sites: bacillariophyceans, cyanophyceans and cryptophyceans. The dominant species was the diatom Asterionella formosa. Nutrient and chlorophyll concentrations increased from summer to fall. Based on nutrient and chlorophyll g concentrations and species diversity indices, Lake Drummond is classified as eutrophic.
