Advertisement Call And Distribution Of The Treefrogs Hyla Chrysoscelis And Hyla Versicolor In Virginia, Joseph C. Mitchell, Christopher A. Pague
Virginia Journal of Science
The gray treefrog complex consists of two sibling species that are indistinguishable morphologically, the diploid Hyla chrysoscelis and the tetraploid Hyla versicolor. Identification is possible in the field only by audio recognition of male advertisement call trill rates (pulses/second). During 1979-1983 we evaluated taped calls of these two species taken from 89 populations from throughout Virginia to map their respective ranges and to evaluate differences in call parameters. Hyla chrysoscelis occurs in the Coastal Plain, eastern and southern Piedmont, and in southwestern Virginia. Hyla versicoloroccurs in the Piedmont, Blue Ridge, and Ridge and Valley regions south to Wythe …

Analysis Of Biological Features Associated With Meiotic Recombination Hot And Cold Spots In Saccharomyces Cerevisiae, Loren Hansen, Nak-Kyeong Kim, Leonardo Mariño-Ramírez, David Landsman
Mathematics & Statistics Faculty Publications
Meiotic recombination is not distributed uniformly throughout the genome. There are regions of high and low recombination rates called hot and cold spots, respectively. The recombination rate parallels the frequency of DNA double-strand breaks (DSBs) that initiate meiotic recombination. The aim is to identify biological features associated with DSB frequency. We constructed vectors representing various chromatin and sequence-based features for 1179 DSB hot spots and 1028 DSB cold spots. Using a feature selection approach, we have identified five features that distinguish hot from cold spots in Saccharomyces cerevisiae with high accuracy, namely the histone marks H3K4me3, H3K14ac, H3K36me3, and H3K79me3; …

Human papillomavirus vaccines have potential to reduce cervical cancer incidence and mortality; however, cultural and economic barriers may hinder success in developing countries. We assessed impact of a single vaccine campaign in Mali with use of mathematical modeling. Our model shows that decreases in the prevalence of Human papillomavirus infection are proportional to achieved vaccination coverage.

In this study, we determined the LD50 (50% lethal dose) for cell death, and the ED50 (50% of cell population staining positive) for propidium (Pr) iodide uptake, and phosphatidylserine (PS) externalization for several commonly studied cell lines (HeLa, Jurkat, U937, CHO-K1, and GH3) exposed to 10-ns electric pulses (EP). We found that the LD50 varied substantially across the cell lines studied, increasing from 51 J/g for Jurkat to 1861 J/g for HeLa. PS externalized at doses equal or lower than that required for death in all cell lines ranging from 51 J/g in Jurkat, to 199 J/g in CHO-K1. Pr …

The objectives of this communication were to fabricate pure samples of multi-walled carbon nanotubes (MWCNTs) and to determine their toxicity in tumor cell lines. MWCNTs were dispersed in a concentration of the surfactant T80 that was minimally toxic. Cell-type variation in toxicity to MWCNTs was observed but was not significantly different to unexposed controls. Additionally, we investigated the increased cell killing of the pancreatic cancer cell line PANC1 when exposed to ultrashort (nanosecond) pulsed electrical fields (nsPEF) in the presence of MWCNTs as a potential form of cancer therapy. We hypothesized that the unique electronic properties of MWCNTs disrupt cell …

We investigated the effects of nanosecond pulsed electric fields (nsPEF) on three human cell lines and demonstrated cell shrinkage, breakdown of the cytoskeleton, nuclear membrane and chromosomal telomere damage. There was a differential response between cell types coinciding with cell survival. Jurkat cells showed cytoskeleton, nuclear membrane and telomere damage that severely impacted cell survival compared to two adherent cell lines. Interestingly, disruption of the actin cytoskeleton in adherent cells prior to nsPEF exposure significantly reduced cell survival. We conclude that nsPEF applications are able to induce damage to the cytoskeleton and nuclear membrane. Telomere sequences, regions that tether and …

Covalent attachment of carbohydrates to proteins is one of the most common post-translational modifications. At the cell surface, sugar moieties of glycoproteins contribute to molecular recognition events involved in cancer metastasis. We have combined glycan metabolic labeling with mass spectrometry analysis to identify and characterize metastasis-associated cell surface sialoglycoproteins. Our model system used syngeneic prostate cancer cell lines derived from PC3 (N2, nonmetastatic, and ML2, highly metastatic). The metabolic incorporation of AC4ManNAz and subsequent specific labeling of cell surface sialylation was confirmed by flow cytometry and confocal microscopy. Affinity isolation of the modified sialic-acid containing cell surface proteins …

Stimuli directed towards activation of apoptosis mechanisms are an attractive approach to eliminate evasion of apoptosis, a ubiquitous cancer hallmark. In these in vitro studies, kinetics and electric field thresholds for several apoptosis characteristics are defined in E4 squamous carcinoma cells (SCC) exposed to ten 300 ns pulses with increasing electric fields. Cell death was [95% at the highest electric field and coincident with phosphatidylserine externalization, caspase and calpain activation in the presence and absence of cytochrome c release, decreases in Bid and mitochondria membrane potential (Δψm) without apparent changes reactive oxygen species levels or in Bcl2 and Bclxl levels. …

This article focuses on the writing strategies second-language students use to compose on social media sites. These alternative and unconventional sites for learning provide language learners opportunities to acquire language by using multiple modalities to respond to various rhetorical situations. In comparison to these sites, academic writing contexts, particularly the developmental-writing course, impose monolingual norms and deficient identities on students. Where these courses articulate these language learners as possessing inadequate skills to perform well in mainstream writing courses, the students' social-media compositions demonstrate that these students have the potential to respond to communicative situations in rhetorically complex ways. Research on assessment and monitoring methods has primarily focused on fisheries with long multivariate data sets. Less research exists on methods applicable to data-poor fisheries with univariate data sets with a small sample size. In this study, we examine the capabilities of seasonal autoregressive integrated moving average (SARIMA) models to fit, forecast, and monitor the landings of such data-poor fisheries. We use a European fishery on meagre (Sciaenidae: Argyrosomus regius), where only a short time series of landings was available to model (n=60 months), as our case-study. We show that despite the limited sample size, a SARIMA model could …

In many coastal and continental margin sediments, pore-water property-property plots yield values of rc:s, the stoichiometric ratio of dissolved inorganic carbon (DIC) produced to sulfate reduced, that are lower than the commonly assumed value of 2. Remineralization of organic matter more reduced than CH2O can cause such observations, as can DIC loss due to authigenic carbonate precipitation. However, through studies of Santa Monica Basin sediments, we have observed that these observations could also be related to the occurrence of anaerobic oxidation of methane (AOM) in sediments. Specifically, using a reactive transport sediment model, we have shown …

The meso- and bathypelagic ocean comprises the largest habitat on earth, yet we know very little about the distribution and activity of protists in this environment. These small eukaryotes are responsible for controlling bacterial abundance in the surface ocean and are major players in the material and energy transfer of pelagic food webs. In this paper, we quantify microbial eukaryotes in the deep North Atlantic, as well as provide a basic characterization of eukaryote community changes through the water column. To this end, we counted organisms using 2 different approaches: (1) catalyzed reporter deposition fluorescence in situ hybridization (CARD-FISH, also …

Diel vertical migration is widespread across diverse taxa in the world's lakes and seas, yet its biogeochemical consequences are still poorly understood. The biologically mediated vertical flux of material in the ocean (also known as the "biological pump") is a matter of major interest and concern, as it is thought to play an important role in regulating ocean carbon storage, and by extension, the global carbon cycle. Recent studies spanning multiple trophic levels from fish to dinoflagellates have led us to a concept of coupled vertical migrations that we refer to as "cascading migrations". That is, migrations that …

Strain HIMB100 is a planktonic marine bacterium in the class Alphaproteobacteria. This strain is of interest because it is one of the first known isolates from a globally ubiquitous clade of marine bacteria known as SAR116 within the family Rhodospirillaceae. Here we describe preliminary features of the organism, together with the draft genome sequence and annotation. This is the second genome sequence of a member of the SAR116 clade. The 2,458,945 bp genome contains 2,334 protein-coding and 42 RNA gene

Iron (Fe) availability restricts diatom growth and primary production in large areas of the oceans. It is a challenge to assess the bulk Fe nutritional health of natural diatom populations, since species can differ in their physiological and molecular responses to Fe limitation. We assayed expression of selected genes in diatoms from the Thalassiosira genus to assess their potential utility as species-specific molecular markers to indicate Fe status in natural diatom assemblages. In this study, we compared the expression of the photosynthetic genes encoding ferredoxin (a Fe-requiring protein) and flavodoxin (a Fe-free protein) in culture experiments with Fe replete and …

The Ross Sea polynya is among the most productive regions in the Southern Ocean and may constitute a significant oceanic CO2sink. Based on results from several field studies, this region has been considered seasonally iron limited, whereby a "winter reserve" of dissolved iron (dFe) is progressively depleted during the growing season to low concentrations (~ 0.1 nM) that limit phytoplankton growth in the austral summer (December-February). Here we report new iron data for the Ross Sea polynya during austral summer 2005-2006 (27 December-22 January) and the following austral spring 2006 (16 November-3 December). The summer 2005-2006 data show …

A microfluidic device integrated with 3D thin film microelectrode arrays wrapped around serpentine-shaped microchannel walls has been designed, fabricated and tested for cell electrofusion. Each microelectrode array has 1015 discrete microelectrodes patterned on each side wall, and the adjacent microelectrodes are separated by coplanar dielectric channel wall. The device was tested to electrofuse K562 cells under a relatively low voltage. Under an AC electric field applied between the pair of the microelectrode arrays, cells are paired at the edge of each discrete microelectrode due to the induced positive dielectrophoresis. Subsequently, electric pulse signals are sequentially applied between the microelectrode arrays …

Although the information on domains and heterogeneities of natural organic matter (NOM) can provide insights into its formation mechanisms, the appropriate solid-state NMR technique for measuring them is still lacking. The traditional technique requires mobility differences in NOM whereas NOM components are primarily rigid. We introduced a new 1H spin diffusion technique, 1H-13C two-dimensional heteronuclear correlation (2D HETCOR) NMR with 1H spin diffusion, for characterization of domains and heterogeneities in aquatic and sedimentary organic matter. It was achieved by collecting a series of 2D HETCOR spectra with a variable mixing time, tm', and …

From Isotropic To Anisotropic Side Chain Representations: Comparison Of Three Models For Residue Contact Estimation, Weitao Sun, Jing He
Computer Science Faculty Publications
The criterion to determine residue contact is a fundamental problem in deriving knowledge-based mean-force potential energy calculations for protein structures. A frequently used criterion is to require the side chain center-to-center distance or the C-alpha-to-C-alpha atom distance to be within a pre-determined cutoff distance. However, the spatially anisotropic nature of the side chain determines that it is challenging to identify the contact pairs. This study compares three side chain contact models: the Atom Distance criteria (ADC) model, the Isotropic Sphere Side chain (ISS) model and the Anisotropic Ellipsoid Side chain (AES) model using 424 high resolution protein structures in the …

Distributions And Abundances Of Microstegium Vimineum Along Forest Roadsides At The Grassy Hill Natural Area, Franklin County, Virginia, Gregory D. Turner
Virginia Journal of Science
In summer 2005, plots were surveyed along roads passing through forest habitats at the Grassy Hill Natural Area Preserve in Franklin County, Virginia to assess the distributions and abundances of Microstegium vimineum in transects located at increasing distances away from roadsides into forest interiors. Across plots, Microstegium was encountered almost exclusively in roadside transects, where abundances were relatively high. While forest composition and topographic features were similar across plots, percent canopy cover and leaf litter depth were greater in interior compared to roadside transects due to undisturbed tree canopies and ground cover located in interior plot areas. Results imply that …

The article presents a study on the marine and terrestrial factors that affect Adélie penguin Pygoscelis adeliae chick growth as well as fledging mass off the western Antarctic Peninsula. In order to examine these factors, the researchers used an individual-based bioenergetics model which stimulates the growth of the penguins. The researchers believe that the results provide insight into climate-driven processes which influence chick growth and emphasize the need for field research that is designed to analyze the factors that identify the availability of Age-Class 3 yr (AC3) and older Antarctic silverfish.
