Despite being relatively benign and not an indicative signature of toxicity, fibril formation and fibrillar structures continue to be key factors in assessing the structure-function relationship in protein aggregation diseases. The inability to capture molecular cross-talk among key players at the tissue level before fibril formation greatly accounts for the missing link toward the development of an efficacious therapeutic intervention for Type II diabetes mellitus (T2DM). We show that human α-calcitonin gene-related peptide (α-CGRP) remodeled amylin fibrillization. Furthermore, while CGRP and/or amylin monomers reduce the secretion of both mouse Ins1 and Ins2 proteins, CGRP oligomers have a reverse effect on …

Representations Of Female Agency In Medieval French Literature, Mathilde Pointiere Apr 2021

Representations Of Female Agency In Medieval French Literature, Mathilde Pointiere

LSU Doctoral Dissertations

This dissertation examines the different ways authors portray female agency in medieval French literature. In focusing on three medieval writers, Chrétien de Troyes, Heldris de Cornouailles and Christine de Pizan, I contend that female agency arises as a result of trauma or crisis. I define my terms as follows: agency is the capacity and intention of performing actions on one’s own behalf. For a fictional character to have agency, therefore, she must be portrayed as having a sense of control and of being the owner of the action she executes. Additionally, I argue that as women characters assume their agency, …

Mental Imagery In Fear Extinction: A Multi-Component Examination Based On Behavioral, Physiological, And Neurological Measures, Xinrui Jiang Apr 2021

Mental Imagery In Fear Extinction: A Multi-Component Examination Based On Behavioral, Physiological, And Neurological Measures, Xinrui Jiang

LSU Doctoral Dissertations

Imagery has long been utilized in clinical treatments of affective symptoms with the assumption that mental imagery can stand in for its perceptual counterpart and exert regulatory effects over emotional responses. While this assumption has its ground in theoretical framework of mental imagery supported by evidence of neurological overlaps between imagery and perception, and clinical applications of imagery interventions were found to be successful, very little has been done through means of experimental examinations.

This investigation began with a differential fear conditioning study (study 1) to simulate and assess imagery extinction. Results provided support for the efficacy of imagery exposure …

Effects Of Planning And Repeated Writing Interventions On Writing Fluency, Katherine Lea Moore Apr 2021

Effects Of Planning And Repeated Writing Interventions On Writing Fluency, Katherine Lea Moore

LSU Doctoral Dissertations

Writing is an essential skill for academic success. Evidence shows students with writing difficulties experience lower overall academic performance (Graham & Perin, 2007). The development of fluent writing is particularly important, as it has been shown to be a strong predictor of global writing quality (Malecki & Jewell, 2003). Limited research exists on writing interventions specifically targeting writing fluency. While performance feedback procedures have been found effective for increasing writing fluency (Hier & Eckert, 2016), interventions such as story-mapping and self-regulation strategy development (SRSD), which target higher order writing skills, have also been shown to improve writing fluency abilities (Harris, …

© 2020 Wiley Periodicals LLC. Novel bio-based microparticles and latexes are synthesized using green chemistry principles through the reaction of aconitic acid (a sugarcane byproduct) and epoxidized linseed oil (ELO). No volatile solvents have been used, and no initiators are present in this polymerization. Using different agitation techniques, we investigate the particles and latexes formed via suspension polymerization in contrast to bulk dispersion polymerization. With the addition of water and a small amount of surfactant to the ELO and aconitic system, a thermally stable material up to 250°C is created that exhibits adhesive and hydrophobic properties. These microparticle latexes are …

This Letter reports the experimental realization of a novel, to the best of our knowledge, active power stabilization scheme in which laser power fluctuations are sensed via the radiation pressure driven motion they induce on a movable mirror. The mirror position and its fluctuations were determined by means of a weak auxiliary laser beam and a Michelson interferometer, which formed the in-loop sensor of the power stabilization feedback control system. This sensing technique exploits a nondemolition measurement, which can result in higher sensitivity for power fluctuations than direct, and hence destructive, detection. Here we used this new scheme in a …

A Theoretical Calculation Of Stacking Fault Energy Of Ni Alloys: The Effects Of Temperature And Composition, Mohammad S. Dodaran, Shengmin Guo, Michael M. Khonsari, Nima Shamsaei, Shuai Shao

Faculty Publications

Combining cluster expansion (CE) method with one dimensional axial Ising model, this work investigated the effects of alloying elements and configurational variations due to temperature on the stacking fault energy (SFE) of FCC Ni binary alloys. Ensembles of large numbers of atomistic structures, each with more than ~400 atoms, were generated to consider sufficient long-range chemical disorder and temperature effects due to configurational entropy. A Monte Carlo Metropolis algorithm was used to generate these structures, whose energies were then evaluated based on the effective cluster interactions obtained from CE. As a baseline, this work had shown the SFE of pure …

Drought stress is an important concern worldwide which reduces crop yield and quality. To alleviate this problem, Trichoderma asperellum has been used as a plant growth-promoting fungus capable of inducing plant tolerance to biotic and abiotic stresses. Here, we examined the effect of T. asperellum inoculation on sugarcane plant above and belowground development under drought stress and investigated the role of this fungus on inducing tolerance to drought at physiological and biochemical levels. The experiment was performed in pots under greenhouse conditions, with four treatments and four replicates. The treatments consisted of sugarcane plants inoculated or not with T. asperellum …

High Temperature Polymer Electrolytes For Hydrogen Fuel Cells And Electrochemical Pumps, Gokul Venugopalan Apr 2021

High Temperature Polymer Electrolytes For Hydrogen Fuel Cells And Electrochemical Pumps, Gokul Venugopalan

LSU Doctoral Dissertations

Hydrogen fuel cell and separation technologies such as proton exchange membrane fuel cells (PEMFCs) and electrochemical hydrogen pump (ECHP) offer a profound advantage in the transition to a low-carbon economy. An imperative hitch in hydrogen fuel cells and ECHP technology has been the electrocatalyst poisoning by carbon monoxide (CO) and other contaminants in the reactant mixture. By operating, hydrogen fuel cells and ECHPs at high temperatures (>200 °C), the effect of CO adsorption on the electrocatalyst surface could be curtailed. The high-temperature operation of devices necessitates a proton exchange membrane (PEM) to operate under anhydrous conditions.

In this work, …

Somatic cells age and die, but the germ-cell lineage is immortal. In Caenorhabditis elegans, germline immortality involves proteostasis renewal at the beginning of each new generation, when oocyte maturation signals from sperm trigger the clearance of carbonylated proteins and protein aggregates. Here, we explore the cell biology of this proteostasis renewal in the context of a whole-genome RNAi screen. Oocyte maturation signals are known to trigger protein-aggregate removal via lysosome acidification. Our findings suggest that lysosomes are acidified as a consequence of changes in endoplasmic reticulum activity that permit assembly of the lysosomal V-ATPase, which in turn allows lysosomes to …

Suboptimal uterine fluid (UF) composition can lead to pregnancy loss and likely contributes to offspring susceptibility to chronic adult-onset disorders. However, our understanding of the biochemical composition and mechanisms underpinning UF formation and regulation remain elusive, particularly in humans. To address this challenge, we developed a high-throughput method for intraorganoid fluid (IOF) isolation from human endometrial epithelial organoids. The IOF is biochemically distinct to the extraorganoid fluid (EOF) and cell culture medium as evidenced by the exclusive presence of 17 metabolites in IOF. Similarly, 69 metabolites were unique to EOF, showing asymmetrical apical and basolateral secretion by the in vitro …

Structure-Activity Relationship Analysis Of Psychedelics In A Rat Model Of Asthma Reveals The Anti-Inflammatory Pharmacophore, Thomas W. Flanagan, Gerald B. Billac, Alexus N. Landry, Melaine N. Sebastian, Stephania A. Cormier, Charles D. Nichols

Division of Basic Science Faculty Publications

Psychedelic drugs can exert potent anti-inflammatory effects. However, anti-inflammatory effects do not appear to correlate with behavioral activity, suggesting different underlying mechanisms. We hypothesized that the distinct structural features of psychedelics underlie functionally selective mechanisms at the target 5-HT2A receptor to elicit maximal anti-inflammatory effects. In order to test this hypothesis, we developed a new rat-based screening platform for allergic asthma. Next, we investigated 21 agonists at the 5-HT2A receptor from the three primary chemotypes (phenylalkylamine, ergoline, and tryptamine) for their ability to prevent airways hyperresponsiveness as a measure of pulmonary inflammation. Furthermore, we assessed each drug for in vitro …

Anesthetic Combination Of Alfaxalone, Dexmedetomidine And Hydromorphone For Comparison Of Endoscopy-Guided Intubation And Blind Orotracheal Intubation In Domestic Rabbits (Oryctolagus Cuniculus), Stephanie Reabel

LSU Master's Theses

Domestic rabbits have a high risk of anesthesia-related mortality compared to other companion animals. Factors that may contribute to increased mortality include drug-induced cardiopulmonary effects (Grint & Murison 2008; Navarrete-Calvo et al. 2013), difficulty performing intubation (Aeschbacher 1995; Flecknell 2016; Wenger et al. 2017) or damage to laryngeal and tracheal tissues following orotracheal intubation (Grint et al. 2006; Phaneuf et al. 2006; Engbers et al. 2017).

Alfaxalone is a neuroactive steroid that produces induction of anesthesia and cardiorespiratory depression in a dose-dependent manner when injected intravenously. The ability to induce anesthesia via an intramuscular (IM) route has several advantages in …

Effects Of Historical Land-Use Change On Surface Runoff And Flooding In The Amite River Basin, Louisiana, Usa Using Coupled 1d/2d Hec-Ras–Hec-Hms Hydrological Modeling, Alexandre G. H. Cowles

LSU Master's Theses

The Amite River Basin is a largely rural watershed spanning parts of four counties in southern Mississippi and seven parishes in southeast Louisiana, with basinwide imperviousness increasing from 0.82% in 1938 to 3.85% in 2016. The Basin has been the subject of significant research interest since catastrophic flooding in 2016 caused 13 deaths and widespread damages. Rapid development in recent decades has led to an expansion of impervious surfaces in Baton Rouge and surrounding areas, encroaching on floodplains and wetlands. At the basin scale, differences in flooding due to impervious cover changes were found to be somewhat limited, particularly along …
