A (beta)-glucosidase gene was cloned from a cellulolytic bacterium, which was obtained by hybrid-formation between Bacillus cereus and a Cellulomonas sp. through protoplast fusion, into Escherichia coli plasmid pBR322 using recombinant DNA techniques. E. coli strain transformants of JM83 harboring this cloned plasmid were able to utilize cellobiose as a carbon source. (beta)-glucosidase activity was assayed in these cells by P-nitrophenyl (beta)-D-glucopyranoside (PNPG) hydrolysis. Hybridization of the cloned DNA fragment with DNA from the hybrid organism was performed by Southern transfer (92) and DNA/DNA dot blot procedures. This study was conducted to determine the effect of instruction in a metacognitive self-monitoring strategy on functional reading ability of 15- and 16-year-old learning disabled students. Metacognitive strategy training has been shown to be effective with older or better readers when used with content or narrative materials This study was unique in that the metacognitive self-monitoring instruction was provided utilizing functional or everyday reading materials. An analysis of pan evaporation by synoptic weather types revealed that pan evaporation rates vary significantly by weather type conditions. Fair weather types are associated with the greatest evaporation rates as was expected, and stormy weather types with the least. These findings are primarily related to the variation of solar radiation by weather type. An investigation of the relationship of pan evaporation to solar radiation by synoptic weather types revealed that the ratio of pan evaporation, expressed in energy units, to incoming solar radiation, also varies considerably by synoptic weather types. The purpose of this study was to determine whether educational technology variables differentiate between "effective" and "ineffective" elementary schools. It was hypothesized that "effective schools" use educational media in instruction more frequently than "ineffective schools". It was also expected that "effective schools" use educational media in qualitatively superior ways to "ineffective schools". Research was conducted to develop methods for improving rice through somaclonal variation using plant tissue culture techniques. Experiments were directed toward developing an efficient culture system by which plants could be efficiently regenerated in rice somaculture. Histological studies of the process of callus formation and plant regeneration were also carried out. Explant sources evaluated for their potential for callus induction and plant regeneration included seedling mesocotyl, root-tips, embryos from mature seeds, nodes, immature panicles, and mature panicles. European-type mixed-breed goat kids were infected with Ostertagia ostertagi or were given chronic pentagastrin treatment. Selected clinicopathological parameters and fecal egg counts were monitored throughout the study. At necropsy the abomasal physical parameters were recorded and tissue samples were saved for parasite recovery, quantitative and qualitative light microscopy and transmission and scanning electron microscopy. Serum pepsinogen and gastrin immunoreactivity increased in parasitized animals. The increase was associated with the tissue phase of the parasitic development. Pentagastrin did not alter pepsinogen or gastrin immunoreactivity. All 8 infected animals had patent infection, and approximately 5% establishment of the infective dose of 15,000 …

The purpose of the present study was to examine whether instruction in story structure improved preschoolers' comprehension of stories. Preschool children were pretested on: (a) oral story production, (b) story rule knowledge (structural elements), (c) free recall of a story, and (d) probed recall. Students who were identified as less skilled by these four measures were randomly assigned to one of three groups: instruction, story, or control. The objective of this study was to compare benthic microfloral production on the west and gulf coasts of the U.S.A., and to determine what factors govern that production. Intensive, month long field studies were conducted at two sites in a salt marsh lagoon (Mugu Lagoon) in southern California and at two sites in a shallow water estuary (Barataria Estuary) in southwest Louisiana. For both studies, fluctuations in production during a single month approximated those measured previously over an entire year: daily production varied from 0 to 1500 mg C (.) m('-2). The thermal death time of Clostridium botulinum type E spores in crawfish tailmeat was determined at temperatures ranging from 80 to 95(DEGREES)C. Crawfish paste containing 10('6) spores per gram was dispensed into 16 x 125 mm test tubes, which were subsequently placed in water baths preset at the test temperatures. Sample tubes were removed from the baths at predetermined time intervals and incubated anaerobically for up to two months. Information integration theory suggests that differences in the moral judgments of children and adults is an example of a development process of information integration where younger children employ a unidimensional integration rule, and older children and adults employ multidimensional integration rules. Studies that utilize the information integration approach involve single-episode stories in which two or three levels of intention and consequence story cues are manipulated in a within-subjects factorial design. The effect of heat stress on plasma profiles of progesterone, luteinizing hormone (LH) and cortisol was evaluated following cloprostenol (500 (mu)g) induced luteolysis in lactating and non-lactating cows in a controlled environment. Another experiment was conducted to evaluate the effect of heat stress on the superovulatory response in lactating cows. The plasma progesterone, LH and total estrogen concentrations were not altered by heat stress (P > 0.05) when the average rectal temperatures of both lactating and non-lactating cows were raised to 39.83(DEGREES)C (103.7(DEGREES)F). However, plasma cortisol was significantly suppressed (P < 0.05) during heat stress. Luteolysis was followed by a preovulatory LH surge in all cows following cloprostenol and thus heat stress did not appear to alter the responsiveness of the corpus luteum to cloprostenol and the factors which regulate the hypothalamic control of LH release. Preovulatory LH peaks were preceded by total estrogen peaks and therefore heat stress did not appear to alter the positive feedback of estrogens from the preovulatory follicles to release LH from the pituitary gland. The magnitude of the diurnal variation of cortisol was higher during lower stress. LH peaks were seen irrespective of cortisol concentrations during both higher and lower stress treatments. However, LH peaks coincided with those of cortisol and total estrogens during lower stress treatments in both lactating and non-lactating cows. An altered diurnal variation in rectal temperatures was seen following LH peaks during lower stress. Fifteen transferable embryos were recovered during lower stress (15/34 or 44%) compared to one transferable embryo (1/4 or 25%) during higher stress (P > 0.05) in the superovulation study. The spatial and temporal variability of the Polar Front in the Sea of Japan is investigated by analyzing both the conventional hydrographic data from the files of NODC and sea surface temperature (SST) data and sea height data derived from satellites. The root mean square variability of the annual mean frontal position, which extends from 38.5(DEGREES)N near the Korean Peninsula to 41.5(DEGREES)N near the Tsugaru Strait, is about 100 to 120 km in the western and central region and is 300 km in the eastern region. A questionnaire was devised to determine the general status of modal counterpoint classes in selected colleges and universities in the United States and to discover the most-used modal counterpoint textbooks, specifically those exhibiting the direct approach; it was sent to 152 NASM approved schools, and 117 (76.97%) responses were obtained. Information was requested about the types of counterpoint classes offered, the respondents' ratings of the modal counterpoint texts they use, teaching aids and techniques, and ways in which modal counterpoint is included in the music curriculum. Ninety-one (77.8%) of the 117 responding institutions included modal counterpoint in their course offerings. …

This study focused on the instructional use of three prereading vocabulary strategies, Directed Reading Activity, Reconciled Reading Lesson and List-Group-Label, as a means for improving vocabulary and comprehension of basal reader stories. Subjects were 66 average and above-average second-grade students. Data were collected over a seven-day period with instruction and post-testing occurring on alternating days. Data were analyzed in three ways. First, a repeated measures analysis of variance on the pre- and post-test vocabulary scores indicated that statistically significant amounts of learning took place within each instructional group. As part of a chemical-ecological study of the Florida scrub community, Ceratiola ericoides (Empetraceae) and Calamintha ashei (Lamiaceae) were chemically investigated for natural products with possible allelochemical activities. The chromatographic separations and chemical investigations were guided by germination and radicle growth bioassays performed on seeds of commercial lettuce (Lactuca sativa) and Schizachyrium scoparium, a native grass of the Florida sandhill community. From the dichloromethane extract of aerial parts of C. ericoides, 2', 4'-dihydroxychalcone, the dihydrochalcones angoletin and 2',6'-dihydroxy-4-methoxy-3',5'-dimethyldihydrochalcone, the flavanones 8-methylpinocembrin, 6,8-dimethylpinocembrin and 7-hydroxyflavanone were isolated along with the triterpenes erytrodiol and ursolic acid. Screen washing of material from new and previously known fossil localities in Late Cretaceous/early Tertiary deposits in Big Bend National Park, Texas resulted in establishing the presence of sediments of early Paleocene (Puercan) age in the upper Javelina Member of the Tornillo Formation. Deposition of the Tornillo Formation was probably continuous across the Cretaceous/Tertiary boundary. Preliminary paleomagnetic zonation indicates that the highest occurrence of dinosaurs at Bid Bend is in a reversed interval. At least part of the early mafic stage of Laramide magmatism in the Big Bend area was post-early Paleocene in age. A new Maastrichtian locality in the …

Three different treatments of salt 6, 8 and 10% were used in the process of aging hot pepper mash in oak and plastic barrels over a period of two years. The chemical characteristics of the changes during the aging process of these mashes were determined. These characteristics included pH, titratable acidity, salt content, color changes and total pectic substances. The pectic substances were degraded by the hot pepper pectic enzymes very rapidly after only a few days of aging. In an effort to unravel the dependence of the dynamics of three-bound-particle systems on the relative masses of the particles, an adiabatic treatment in hyperspherical coordinates is used to study the ground state and some doubly excited states of Ps('-) (positronium negative ion) and other three-particle systems, including H('-) and M('-) (muonium negative ion). Two different methods for the solution of the adiabatic eigenvalue equation are presented; one uses a "prediagonalized" basis of hyperspherical harmonics, and the other uses an asymptotic basis which consists of hydrogenic functions for the positronium. The pathogenesis of equine herpesvirus type 1 (EHV-1) and type 2 (EHV-2) respiratory infection was studied in cell and organ cultures by infectivity assays following experimental infections (growth kinetics), and by light, scanning and transmission electron microscopy. The cell cultures included fetal donkey and pony alveolar macrophages (FDAM/FPAM), fetal equine kidney (FEK), rabbit kidney (LLC-RK(,1)), and african green monkey kidney (VERO) cells. Organ cultures were derived from respiratory tissues (nasal, tracheal and pharyngeal mucosae) of seronegative donkey (F1D1) and pony (F1M1) fetuses, and of a seropositive adult pony (A1). A model integrating elements of job stress theory, coping theory, and Type A behavior research was developed and tested in the present study. The model was employed to generate numerous hypotheses concerning proposed relations among job stresses, coping strategies, Type A behavior, and stress-related outcomes. These hypotheses were tested utilizing questionnaire data collected from 136 recently-graduated nurses in ten hospitals. As predicted, Type A was positively associated with job stresses, both Type A and job stresses were related to felt strain and organizational commitment, and commitment predicted turnover intention. The objective of this research was to investigate the level of alienation among a sample of aged respondents from East Baton Rouge Parish Council on Aging Senior Centers. The second objective was to determine if black aged respondents would be more alienated than white aged respondents, and the third objective was to determine the relationship between seven independent variables (RACE, EDUCATION, CHURCH WORK, LIVING ARRANGEMENT, VOLUNTEER WORK, INCOME and HEALTH RATING) and four subtypes of alienation (PERSONAL ISOLATION, GROUP ISOLATION, POWERLESSNESS and NORMLESSNESS). We study the elements of a semigroup which are minimal or maximal with respect to Green's quasiorders. Part 1 begins with a preliminary review. The sets of minimal elements are characterized in terms of minimal ideals. We discuss the relationship between the min set of a semigroup and the min set of a subsemigroup. The sets of maximal elements are characterized, and it is shown that these sets do not necessarily satisfy any inclusion relationships to each other. Lipogenic responsiveness to insulin was investigated at different times of day in freshly isolated hepatocytes and in primary cultured hepatocytes of Syrian hamsters, Mesocricetus auratus held on 14 hour daily photoperiods (lights on: 08.00) by following (('14)C)acetate incorporation into total cell lipid. The influence of prolactin on lipogenesis was tested by adding the hormone in vivo and directly to primary cultured hepatocytes. The findings of these studies are unique in that they demonstrate important roles of temporal interaction between prolactin and insulin in regulation of lipogenesis in cultured hepatocytes. Jurors' Decisions In A Simulated Trial Of The Pms Defense: Background Characteristics And Attitudes Toward Menstruation., Marjorie Carole Donohoe

This mock jury study assessed the feasibility of utilizing Premenstrual Syndrome (PMS) as a substantive defense in a capital case. One hundred-eighty mock jurors were asked to render judgments in identical cases of spouse murder with one of three defenses: (1) temporary insanity due to PMS; (2) temporary insanity with a reported history of psychiatric treatment; and (3) temporary insanity brought about by extreme emotional trauma. Results indicated that mock jurors rejected an insanity defense for the PMS defendant. Jurors were more willing to render an NGRI (Not Guilty by Reason of Insanity) verdict for the defendant with a documented …
